    • Electrophysiological and Histopathological Studies on Cochlear Changes Induced by Iron Deficiency in Rats

      Abstract:<正> We report a new animal model of cochlear changes induced by irondeficiency.The experimental studies were conducted in 141 rats fed with basiciron-deficient diet for 7 to 100 days,and 130 normal or chronically anemic ratswere used as controls.Electrophysiological findings showed that the incidence ofover 15 dB auditory threshold elevation was 31.85% in the iron-deficient rats ascompared with zero percent in controls.The main cochlear histopathologicalchanges induced by iron deficiency were strial atrophy and reduction of spiralganglion cells.We conclude that the observed anomalies should be attributed tocochlear tissue iron deficiency.

    • Early Morphological Characteristics of High Velocity Fragment Wounds

      Abstract:<正> The soft tissue wounds on the hind limbs of 21 dogs were inflicted bysquare fragments(0.35g in weight)which were shot with a smooth-bore testinggun.The impact velocities were 700-1500 m/s.Debridement and grossexamination were carried out 6 hours after injury.The main conclusions were asfollows:1.The primary wound track in the high velocity group looked like a funnel inshape.The part of the wound cavity near the entrance was the largest indiameter.2.The severity of tissue damage depended on the impact velocity.The higherthe velocity,the larger the wound cavity.3.The tissue damage was irregular or uneven,‘jumping’or‘mosaic’inappearance.In addition,the differences of the morphological characteristics between thewound tracks due to bullets and those due to square fragments were pointed outand their causes discussed.

    • Mechanisms of Natural Killer Cell-mediated Tumor Cell Cytolysis at a Single Cell Level

      Abstract:<正> The interaction between natural killer cell and tumor target cells at asingle cell level was investigated.The study with a modified single cell cytotoxicassay showed the heterogeneity and requirment of target cell membrane fluidity inthe formation of natural killer cell-tumor target cell conjugates,as well as aninternal disintegration model of natural killer cell-mediated target cell lysis,which may have some relation to the increased lysosome reaction in damagedtargtet cells as revealed by ultrastructural study.From this finding,a“target cellsuicide”model of natural killer cell mediated target cell lysis is proposed.Inaddition to these findings,the interaction of natural emperipolesis lymphocyteswith tumor cells and its relationship to natural killer cell-mediated killing werealso studied and are discussed here.

    • Purification of Human Interferon -α by Antibody Affinity Chromatography

      Abstract:<正> Results of the establishment and application of anti-interferon antibodycolumn are reported.Antiserum against human lymphoblastoid interferon(HuLyIFN)was obtained by immunizing sheep with partially purified Namalvainterferon.After they were isolated from antiserum,the immunoglobulins againstHuLyIFN were adsorbed repeatedly on the“mock interferon”column and trypsininhibitor column.Anti-interferon antibody column was established by couplingthe adsorbed anti-interferon antibody to Sepharose 4B.Crude Namalva interferonwas purified by antibody affinity chromatography in one step to a specific activityof 8×106 to 8×107 units/mg protein with the recovery of over 100%.Peak ofpurified interferon activity in elution was seen mostly in fractions 1-3(3-8ml).Similar results were obtained in purification of human leukocyte interferon(HuLeIFN)by this column.The polyclonal antibody column could purifyrecombined leukocyte interferon-xD(LrIFN-αD)to homogeneity as analyzed onSDS-polyacrylamide gel electrophoresis.

    • A Clinical Study of Plasma Fibronectin in Patients with Severe Viral Hepatitis

      Abstract:<正> Plasma fibronectin was determined serially with both simpleradioimmunadiffasion and modified indirect hemoagglutination test(usinggelatin-coated sheep RBC)in 30 patients with severe viral hepatitis(SVH),100normal subjects and 30 cases of chronic active hepatitis(CAH)were also studiedto serve as controls.Liver function test,serum complement 3 and immunoglobulinwere determined simultaneously.Endotoxin was assayed with limulus lysate testin 21 cases of SVH and 14 cases of CAH.The normal value of plasma fibronectin was 284.31±45.56μg/ml and 1:30.64(geometrical mean).Plasma fibronectin level decreased significantly in SVHpatients.Obvious correlations were found between the levels of plasma fibronectinand the valuers of total serum bilirubin,prothrombin time,complement 3,immunoglobulin M,and blood platelet count.Low plasma fibronectin levels wereremarkably associated with the occurrence of endotoxemia.It is concluded that serial determination of plasma fibronectin can reflect theseverity of the illness and the prognosis of the patients with SVH.

    • An Ultrastructural Study on Hyperosmotic Opening of Tight Junction by Lanthanum Nitrate Tracing

      Abstract:<正> A model of hyperosmotic opening of the blood-brain barrier(BBB)was successfully made by infusion of 1.4M mannitol into the internal carotidartery of rabbit.We selected lanthanum nitrate as a tracer.Ultrastructuralchanges of hyperosmotie opening of the BBB were observed through injection oflanthanum nitrate into blood vessels or through impregnation of small blocks ofbrain tissue in lanthanum solution.Electron microscopy showed that lanthanumnitrate passed through the opening of endothelial tight junction from the bloodinto the subendothelial layers and the extracellular spaces of the brain,or fromthe outside into the inside of blood vessels.The data suggest that themorphological basis of osmotic opening of BBB is the reversible widening of thetight junctions between endothelial cells,and that lanthanum nitrate infusion hasan advantage over block impregnation.

    • Mechanisms of Protective Activities of Monoclonal Antibodies Against Malaria Parasite Plasmodium Falciparum

      Abstract:<正> A range of monoclonal antibodies specific for Plasmodium falciparumwere tested in vitro for their abilities to inhibit the multiplication of a partiallysynchronized culture of P.falciparum,to augment the phagocytosis of theparasites by macrophages,and to enhance the hilling of parasites by peritonealcells depleted of adherent cells.Seven of 17 monoclonal antibodies,ranging fromculture supernatant and ascitic fluid to purified IgG,were shown to have a dose-and time-dependent inhibition of the parasite growth in vitro.At a concentrationof 0.6mg/ml,the inhibitory capacity of these monoclonal IgG was above 94% overa 3-day culture period,much higher than that of the relevant polycloaal IgG.Four of six monoclonal antibodies tested augmented the phagocytosis of theparasite by macrophages,resulted from the opsonization of the parasites.Four ofthe seven monoclanal antibodies examined showed cytotoxic activity on malariaparasites.Peritoneal cells depleted of adherent cells were capable of killing theparasites in the presence of monoclonal antibodies.The results indicate that theremay be“monofunction”,“bifunction”,and“multifunction”types of monoclonalantibodies against P.falciparum.The putative protective antigen of malariaparasite purified by“multifunctional monoclonal antibody”affinity chromatographymay have potential interest as a vaccine against the parasite.

    • The Receptor Role of Glycophorin A during the Invasion of Erythrocytes by Plasmodium Falciparum

      Abstract:<正> Under the condition of in vitro culture,the effects of glycophorin A(GPA)of human erythrocyte membrane,anti-GPA IgG,α1-acid glycoprotein,ovomucoid,wheat germ agglutinin,etc.on the invasion of erythrocytes by P.falciparum were observed in this study.The results show that the combination ofGPA with P.falciparum merozoites has high specificity,high affinity and atendency to saturation,and produces specific biological effects.These confirm forthe first time from the characteristics of the ligand-receptor interaction that GPAis a receptor recognized and bound by P.falciparum merozoites.

    • Isolation of CIq in Human Serum with Anti-human CIq McAb Affinity Column Chromatography

      Abstract:<正> This paper is to report a procedure for the isolation of Clq in humanserum with anti-human C1q McAb affinity column chromatography.Thisprocedure is simple and rapid,Which can be performed within 24 to 48 hours.The yield of Clq can reach 7-10 mg per 100 ml serum.The preparation isimmunoglobulin free as proved by double diffusion and immunoelectrophoresisassay and exhibits typical C1q subunit structure on SDS-PAGE.After biologicalactivity evaluation with red blood cell agglutination and latex agglutinationassay,this C1q preparation was found to be satisfactory in its activity.The anti-human C1q McAb sepharose 4B column can be used repeatedly.Itcan be stored in 4℃ for approximately a year without losing any of its absorptionactivity.

    • Effects of Iodium-Heterocyclic Compound on Physiological Properties of Guinea Pig Papillary Muscles

      Abstract:<正> Effects of iodium-heterocyclic compound(IHC-64)on physiologicalproperties of guinea pig papillary muscles were reported.IHC-64 not onlydepressed the myocardiac contraction with IC50119.4±10.9μm,but also prolongedthe effective refractory period(ERP)from 190.5±13.9ms to 202.0±13.6 ms(P<0.01).Moreover,it increased the concentration for adrenaline to induceautomaticity of the papillary muscles by 2.8±1.1 folds(P<0.01)without markedeffects on its excitability.IHC-64 reduced significantly the contractility inducedby isoprenaline in potassium-depolarized guinea pig papillary muscles,makingthe isoprenaline dose-effect curves shift in a non-parallel manner.These resultsdemonstrate preliminarily that IHC-64 may be a new calcium antagonist.

    • The Antipyretic and Bacteriostatic Action of Gegen Qinlan Decoction and Its Effects of Immune Function

      Abstract:<正> The pharmacological effect about the recipe of ancient Chinesemedicine and herb drugs—Gegen Qinlian Decoction(GGQLD)was studied.The results showed that it could markedly lower the body temperature of feverishrabbits caused by infection of pentavaccine.The effect is as strong as that ofAspirin compound(APC),there is no statistical significance between animaltreated by GGQLD and those by APC.It could also inhibit the growth of manykinds of bacteria(Staphylococcus;Bacillus dysenteriae;Pneumococcus)in vivoand in vitro.It was observed that the percentage of ANAE+ lymphocytes in thespleen and lymphonodes and the number of PFC in the spleen of mouse could beincreased by GGQLD.Meanwhile,the phagocytic effect of peritoneal macrophagesin mice treated by GGQLD was strengthened,which suggested that the antifebrileand antibiotic effects of this medicine are partly related to its efficiency to enhancethe immunity against pathogens.

    • On the Chemistry of Pheophorbide a:A Novel Photosensitizer

      Abstract:<正> A method for the preparation of pheophorbide a(a novel photosensiti-zer for tumor photolocalization and photodynamic therapy)from silkwormexcrement oily chlorophyll is reported.The chromatographic,spectroscopic andphotochemical properties of pheophorbide a were studied,and its distribution intissues of S180 tumor-bearing mice was determined.The UV/visspectrum of the obtained pheophorbide a was identical with thatreported by other authers,but it was still found to contain at least twocomponents(probably two optical isomers)by HPLC analysis and1H-NMR spectrometry.The results of a preliminary study on itsdistribution in tumor-bearing mice showed that it was accumulatedselectively in tumor tissues after intravenous injection and at 24 hoursafter drug administration the tumor to skin ratio rose to as high as 15.

      Abstract:<正> Serum renin-angiotensin-aldosterone(RAA)levels were determined on169 occations in 96 cases of hemorrhagic fever with renal syndrome(HFRS).Theresults reveal that RAA system levels rise in vaviable degrees in different stages ofHFRS,the highest value being in the hypotensive and oliguric stages and thenormal value,in the polyuric and convalescent stages.The elevation range iscorrelated with the severity of the disease.Being closely related to the decrease ofblood volume before the bypotensive stage and to the“tubuloglomerular feedback”mechanism after the oliguric stage,the activation of RAA system may be one ofthe contributing factors of the acute renal failure in HFRS.

    • Interposed Jejunal Loop with Artificial Papilla:A New Procedure of Choledocho-duodenostomy

      Abstract:<正> Internal drainage of the biliary tract such as choledocho-duodenostomyand Roux-en-y choledocho-jejunostomy is a common operation for the treatment ofprimary choledocholithiasis and biliary strictrue.Though these procedures cancreat a good drainage,serious reflux cholangitis is frequently resulted.In orderto prevent this tiresome complication,the authors have designed a new procedure:a loop of jejunum is isolated;an artificial papilla is made on its distal end;thenits proximal end is connected to the bile duct and its distal end with the artificialpapilla is inserted into the duodenum;finally the isolated jejunal loop and theduodenum are sutured up.This paper is to report our experience of applying this procedure on 15 casesin the period from Nov.1982 to Nov.1985.After operation,no reflux of theintestinal contents into the interposed jejunal loop was found and barium mealwas able to demonstrate the reflux of the contrast medium only in one case out ofthe 15.14 cases have been followed up for 6-36 months and excellent results werefound.It proves that this operative procedure can prevent the reflux of chyme intothe biliary tract.In addition,interposed jejunal loop with an artificial papilla ismore physiological than Roux-en-y choledocho-jejunostomy since bile can flowthrough the interposed jejunal loop into duodenum to mix with the pancreaticfluid and chyme,which is helpful to better disgestion.
