Abstract:<正> With cell-mediated 51Cr cytotoxicity assay,it was demonstratedthat the RNA extracted from H22 tumor cells(H22TuRNA)could.significantly inhibit the cytotoxic effect of lymphocytes of normalmouse spleen when treated with H22TuRNA at a dosage of≥250μg H22TuRNA/1×107,lymphocytes at 37℃ for 60 min.The inten-sity of inhibition depended on the incubation time and the dose ofH22 TuRNA.The inhibition capacity was not abolished by treating with:RNase.Polyacrylamide gel electrophoresis showed that only thesmall molecules of RNA with the sedimentation rate about 4sremained after the treatment with RNase.Thus,it was consideredthat the RNA with small molecules which possesses anti-RNaseproperty was one of the active components in H22TuRNA.H22TuRNA could also inhibit the ability of lymphocytes toadhere to glass surfaces.This suggested that the function of H22-TuRNA to inhibit the cytotoxic effect of lymphocytes was to inter-fere with the contact between lymphocytes and target cells.
Abstract:<正> Lung lymph fistula preparation was used to study the effectsof hypoxia and endotoxin on pulmonary pressor response andpulmonary fluid exchange in 9 conscious goats.The results suggestthat 1)the mechanism of pulmonary pressor response induced byhypoxia is somewhat different from that induced by endotxin,2)hypoxia(at a simulated altitude of 4,000 m for 5 hours)exertsno obvious effect on the pulmonary fluid filtration and pulmonaryvascular permeability for protein,and 3)alveolar hypoxia can notenhance the pulmonary injury induced by endotoxin.
Abstract:<正> In 78 cases of hepatocellular carcinoma(HCC),the local immuneresponse in cancerous tissue was investigated with immunofluorescenttechnique.In addition,analyses of clinical and postoperative follow-up data were also done to explore its clinical significance.The resultsindicate that intensity of local immune response is closely correlatedwith the clinical staging of HCC,tumour size,hepatic metastasisand cancerous thrombosis.The local immune response can representthe host immune function against HCC and directly influence theprognosis.It can provide a valuable reference for predicting prog-nosis and selecting general immunotherapy after operation.
Abstract:<正> The toxic effects of nitroquine-dapsone compound(NQD)in mice and dogs were studied.The therapeutic index of NQDin mice is 1911,the greatest among the 6 antimalarials tested.Thetoxic effects of NQD(50 mg/kg/day for 3 days per os)and nitro-quine in dogs were manifested by injuries on the adrenal cortexand intestinal epithelium.When folic acid(4 mg/kg/day for 4 days)or calcium leucovorinum(0.3 mg/kg/day for 4 days)were usedconcomitantly with NQD,the death rate and the incidence of dia-rrhea in the toxicated dogs were greatly reduced,the injury on theintestinal epithelium was much milder,and the goblet cells weremuch more numerous than those without treatment.The results suggestthat folic acid and calcium leucovorinum can protect the undifferen-tiated cells in the intestinal crypts from being injured by NQD.
Abstract:<正> Interferon-gamma(IFN-γ)was induced by pokeweed mitogen(PWM)or esculentoside in human splenic lymphocytes.PWM-inducedIFN-γ was partially purified to a specific activity of 106 RU/mgprotein by batch adsorption on controlled-pore glass(CPG)beadsand desorption by ethylene glycol in phosphate-buffered saline(PBS).Use of Chinese made micro-pore glass(MPG)instead of CPG topurify IFN-γ showed similar purification efficacy.Both CPG andMPG recovered could be regenerated for repeated use.The yieldfrom glass beads was further purified to a specific activity of 107RU/mg protein,with about 78% recovery,by the anti-new-borncalf serum antibodies-Sepharose 4B(ACS)column chromatography.Almost all the antiviral activity of the IFN-γ induced by PWM oresculentoside was destroyed by adjusting to pH 2 or heating to 56℃,and was neutralized by monoclonal antibody to human interferon-γ(Hu IFN-γ),thereby the product was identified as Hu IFN-γ.Bio-gel P-200 chromatography resulted in two peaks of activity for PWM-induced IFN,a major one with molecular weight(MW)of 40,000-45,700 and a minor one of 25,700,but only one peak of 45,700-49,000for esculentoside-induced IFN.
Abstract:<正> Fetal lymphocyte inhibitory factor(FLIF)was prepared bymeans of a successive two-step salting-out procedure using ammo-nium sulphate.FLIF was found to be noncytotoxic and thermostableat 56℃ for 30 min and has the ability of suppressing lymphocytetransformation of human adults,and decreasing both 3H-UR and3H-TdR incorporation into lymphocytes.Our experiments showedthat FLIF was effective only when the cell was on its course ofactivation.
Abstract:<正> A sensitive and specific double-sandwich ELISA system utili-zing ascites fluid containing monoclonal antibody as a coatingantibody was developed for the detection of Clostridium botulinumtoxoid,toxin and artificially contaminated food specimens.Theminimal detectable amount(MDA)of toxoid varied from 4 to 31ng/ml.The lowest concentration for cross-reaction to botulinum AToxoid ranged from 4 to 8μg/ml.The specific index obtained wasbetween 250 and 1000.The MDA of botulin B was 16 ng/ml(4 LD50/ml),whereas that of type A toxin was larger than 32 μg/ml(301LD50/ml);the specific index was over 2000.The MDA of botulin Bin artificially contaminated food varied from 0.67 to 7.8 LD50/mland no cross-reaction was shown between types A and B.The ELISAsystem is close to RPHA in sensitivitv but is apparently superiorto the latter in specificity.
Abstract:<正> A method,which was a modification of that of Stothert et al,was adopted to establish chronic lung lymph fistula in goats fromChongqing area.The procedure consisted mainly that a segment ofthe thoracic duct was isolated to serve as the efferent of the caudalmediastinal lymph node to collect lung lymph,and an irrigationtube was inserted for the periodic administration of heparin solutionto prevent the sanguinous lymph from clotting in the early post-operative days.A series of difficulties including high surgical morta-lity,stoppage of lymph flow,and chyle contamination of the lunglymph were overcome.A trial of the operated goats on steaminhalation injury precipitated instant increase of lung lymph flowand an elevation of lymph/plasma ratio of total protein concentration,which suggested that our animal model was suitable and effective inthe study of the exchanges of fluid and protein in the lungs.Thedetails of technique were described and discussed.
Abstract:<正> The growth of 111 cultures of human epidermal keratinocytes inexplant culture,dispersed cell culture and 3T3 feeder layer culturewas studied.The rate of monolayer formation was 80.0% in 3T3feeder layer cultures,52.5% in explant cultures and only 11.8% indispersed cell cultures.It is concluded that among the modelsstudied the 3T3 feeder layer culture is the most effective andpossesses many advantages for the cultivation of human keratino-cytes;the explant culture would be a simple and easy method if wecan effectively control the contamination by fibroblasts.The growthpattern of cultured keratinocytes in each model was described.
Abstract:<正> 8 specimens of meningioma(4 meningotheliomatous,3 fibrousand 1 transitional)were observed with transmission electron micro-scope(TEM).The main feature of most tumor cells is that there aremany conspicuous interdigitations of cytoplasm protruding outfrom the cell surface.There are a lot of mitochondria,Golgi com-plexes and microfilaments which are closely packed or scatteredinside the interdigitations.Between the interdigitations,many des-mosomes are seen.The meningiomas have an epithelioid structure.which may explain why the tumors cells would not scatter outwhen the smear samples or histological sections are observed underlight microscope.The three types of meningiomas described inthe present work may mainly originate from epithelioid cells ofarachnoid granulations as inferred from the ultrastructure observed.We did not find any distinct differences in the ultrastructuresamong them.
Abstract:<正> This article states that the tachnique of 51Cr-labelled frog redblood cells can be applied to studying acceleration physiology tomeasure the effects of +3Gz acceleration on regional blood flow oforgans in anesthetized rabbits.The results show that the coronaryblood flow of anesthetized rabbits was unchanged during +3Gzstress.However,the blood flow of the brain,liver,stomach,smallintestines,renal cortex and medulla was markedly reduced,especially,that of the renal cortex was decreased by 96.57% as compared withthe control group.The study indicates that it is appropriate to applybiological microsphere technique to the study of acceleration physi-ology.
Abstract:<正> Ten cases of portal hypertension due to liver cir-rhosis were treated with a new procedure of selective distalsplenocaval shunt in the period from January,1984 to October,1985.The liver function was grade Ⅰ in 6 cases,grade Ⅱ in 2,and grade Ⅲ in the other 2.The operative procedure consisted ofa direct end to side anastomosis between the distal part of thesplenic vein and the infeiror vena cava.There was no operativemortality.After the completion of the anastomosis,the portalpressure was reduced by 4.4 cm H2O on average.All the patientshave been followed up for 3 to 24 months,and they are all doingvery well and have resumed fully or partially their original job.Neither rebleeding nor hepatic encephalopathy occurred postopera-tively.The patency of the anastomosis was demonstrated byultrasonography and angiography.The spleen markedly shrinkedin size and became softened in consistency.Our method is a new type of selective shunt.No report of sucha type of shunt has ever been published in the literature.It isrecommended that this operation be used in the treatment of portalhypertension since it is effective.
Abstract:<正> A new orally active non-glycosidic,non-adrenergic cardiotonicagent,2-[(2-methoxy-4-methylsulfinyl)phenyl]-1H-imidazo[4,5-b]pyridine(AR-L115BS)has been synthesized.The two key interme-diates,2,3-diaminopyridine and 2-methoxy-4-methyl-mercaptoben-zoic acid were prepared according to the routes submitted by Foxand Zipp respectively.The approaches were improved to adaptlarge scale laboratory preparation.The structure of the end productwas proved by elemental analysis,mass spectra,1H nuclear magneticresonance spectra and infrared spectra.
Abstract:<正> The cardiotonic activity of imidazopyridine(MD),a Chinese-madeproduct of sulmazol(AR-L 115 BS),was verified and compared withthat of isoprenaline(ISO)on isolated guinea pig atria.The electri-cally-paced left atria were used to determine their positive ino-tropic action,while the spontaneously beating right atria were usedto determine their positive chronotropic action.AS a result,MD showed a strong positive inotropic actioncomparable to that of ISO,and some positive chronotropic actionwhich,however,was much weaker than that of ISO.The positiveinotropic action of a single dose of MD developed quite slowly andincreased gradually in a long time,whereas the action of ISOdeveloped so steeply that its peak effect was reached within a fewminutes.Propranolol could significantly block both inotropic andchronotropic actions of ISO but scarcely those of MD.This suggeststhat MD is not a β1-receptor agonist.The experimental resultsdescribed above are similar to those reported abroad.
Abstract:<正> Effect of electrical stimulation of lateral hypothalamic area(LHA)on pain threshold of rats was studied by using tailflickresponse to radiant heat as the index of pain response and theinfluence of intraperitoneally injected naloxone on the analgesiceffect of LHA was observed.The result showed that pain thresholdof the animals raised 52.5% in average after LHA stimulation andthis analgesic action of LHA decreased significantly after injecionof naloxone.Quantitatively the analgesic effect of LHA was muchweaker than that of electroaccupuncture.The possible underlyingmechanisms were discussed.
