    • The expression of Leu 7(HNK-1)in human lung

      Abstract:<正> Leu 7(HNK-1),a glycoprotein,is a specific antigen presented on the surfaceof human natural killer cells and killer cells.In this study ABC immunostaining with spe-cific monoclonal antibody to Leu 7 was used to detect the expression of Leu 7 in humanlung.The results demonstrated that the Leu 7 immunoreactivity was localized in theneuroepithelial bodies(NEB)and neuroendocrine cells(NEC)of the human lung.In addi-tion,the anti-Leu 7 monoclonal antibody stained the nerve fibers in the wall of theintrapulmonary bronchi.Adjacent sections immunostained with antibodies to Leu 7 and5-hydroxytryptamin(5-HT)or calcitonin gene-related peptide(CGRP)proved thecoexistence of Leu 7 and CGRP or 5-HT in endocrine cells.The CGRP immunoreactivenerve fibers also showed positive staining with the monoclonal antibody to Leu 7.Ourfindings support the hypothesis that the nervous and endocrine systems share some com-mon antigenic determinants with the immune system.

    • Influence of four kinds of polysaccharides on the induction of lymphokine-activated killer cells in vitro

      Abstract:<正> Effects of Lycium barbarum polysaccharide(LBP),Astragalus polysaccharide(APS),polysaccharide of Acanthopanax senticosus(PAS)and polysaccharide of bacteriallipopolysaccharide(PS)on the induction of lymphokine-activated killer(LAK)cells fromC57BL/6 murine splenocytes were studied using [125I]UdR release assay.The fourpolysaccharides alone were shown to induce no cytotoxicity.When combined with humanrecombination interleukin-2(rlL-2),they augmented LAK cell activities in adose-dependent manner,most markedly at 0.01~0.1 mg·ml-1for LBP,0.01 mg·ml-1 forAPS and PAS,and 0.01 μg·ml-1 for PS.They increased LAK cell activity in a shortrange of rIL-2 concentrations(250~1000U·ml-1).They were shown to inhibit LAK cellactivity when used beyond the suitable dosage.

    • Significance of aberrant melanosomes in the diagnosis of malignant melanoma

      Abstract:<正> In order to clarify the significance of the aberrant melanosomes in thediagnosis of malignant melanoma,17 cases of malignant melanoma were studied with elec-tron microscopy,and 30 cases of nevus were studied likewise for comparison.Aberrantmelanosomes were present in 12 cases of pigmented and 2 cases of amelanotic malignantmelanoma.They were also found in 4 cases of congenital nevus.The findings suggest thatthe presence of aberrant melanosomes is of significance for the diagnosis of malignantmelanoma but this has been overemphasized.It is believed that the diagnosis of malignantmelanoma depends upon a comprehensive judgement of all the ultrastructural findings un-der electron microscopy.

    • Purification and identification of HIV-1 gag p20 protein expressed in E.coli

      Abstract:<正> The human immunodeficiency virus type 1 gag p20 gene fragment was clonedinto plasmid pBV220 to construct a recombinant expression plasmid pCY7.By induction,the p20 protein was expressed in E.coli,and it was confirmed by Western blotting thatthe expressed p20 protein could be specifically recognized by anti-p17 monoclonalantibodies.The recombinant bacteria was lysed and fractionated into snpernatant andprecipitate by centrifugation.It was found that the p20 protein was present chiefly inthe precipitate.After p20 protein being washed twice,its purity reached 27.4%.Then theprecipitate was washed with 1 mol/L urea solution and the purity of p20 protein wasraised to 58.1%.Finally,the precipitate was dissolved in 6mol/L of urea and appliedonto a heparin affinity column.Four peaks were obtained and the p20 protein wasfound mainly in peak 3.The purity of p20 protein at this step reached 84.2% as meas-ured by gel scanning.

    • Prophylactic effect of magnesium sulfate against reperfusion induced arrhythmias in the isolated rat hearts

      Abstract:<正> Prophylactic effect of magnesium sulfate on reperfusion arrhythmias was studiedusing a left anterior descending coronary artery occlusion followed by reperfusion in theisolated rat heart.In the first studies,we have observed a bell-shaped relationship be-tween the incidence of reperfusion-induced ventricular fibrillation(VF)and the durationof preceding ischemia.With 5,10,15,20 and 25 min of ischemia,10,70,60,50 and 20per cent of the hearts exhibited irreversible VF,respectively.In the second studies(10 minischemia),perfusate magnesium sulfate was increased to 3.6,4.8 and 6.0 mmol/L 1 min be-fore coronary ligation,VF fell in a dose-dependent manner from its control total inci-dence of 100%(1.2mmol/L MgSO4)to 82%,73% and 18%(P<0.01),respectively.Heartrate was also reduced in a dose-dependent manner,falling from its control value of326±11 to 227±22 beats/min with the highest concentration of magnesium sulfate.Asperfusate magnesium sulfate was increased to 6.0 mmol/L just before reperfusion,no an-ti-arrhythmic effects were observed.With an anti-arrhythmic concentration of magnesiumsulfate(6.0 mmol/L,increased 1 min before ligation),calcium concentration was increasedby 1.5 mmol/L at the same time,under these conditions the anti-arrhythmic effect of mag-nesium sulfate was lost and its negative chronotropic effect was also partially abolished.We conclude that magnesium sulfate has a certain prophylactic effect againstreperfusion induced arrhythmias and this could be due to a direct or indirect calci-um-antagonist action.

      Abstract:<正> The human rectal adenocarcinoma cell line(HR8348)was treated withlympholdne activated killer(LAK)cells in vitro and the changes of the microtubulin inboth the effector and target cells were investigated with the aid of immunofluorescencemicroscopy.It was revealed that after the attachment of LAK cells to the tumor cells,theeffector-target conjugates formed and distribution of the microtubulin in both the effectorand target cells changed.In the LAK cells,the microtubulin concentrated mainly in thecontact region,forming a crescent-like structure,while in the target cells,themicrotubulin condensed into patches and fused with the crescent-like structure of the LAKcells,Eventually,the target cells degenerated and died.It was suggested that the lysis ofthe target cells may be related to the redistribution of the microtubulin in both theeffector and target cells.

    • Localization of X-gene translation product in human cholangiocarcinoma and its surrounding liver tissues

      Abstract:<正> Intrahepatic cholangiocarcinoma is an uncommon type of primary hepatomaand its mechanism is still unknown.Twenty samples of the cancer and the surrounding tis-sues were all taken from the Xi’an area and examined for the presence of X-gene prod-uct(HBxAg)by the avidin-biotin-peroxidase complex method.It was found that 17 cases(85%)showed positive reaction,HBxAg was present in 15 cancer samples and 16non-tumor tissue samples.This result suggests that there is a close relationshipbetween human cholangiocarcinoma and HBV,and the HBxAg might play an importantrole in the pathogenesis of cholangiocarcinoma.

      Abstract:<正> The specific bindings of estrogen,progestin and androgen were determined inthe cytosol fraction of myomatous,adenomyotic and postmenopausal uterine tissues andof the normal endometrium and myometrium as well.It was found that theconcentrations of estrogen,progestin and androgen cytosol receptors were significantlyhigher in myomatous tissue than in normal myometrium;there was also an obvious differ-ence of the concentration of the sex steroid receptors between normal endometrium andadenomyotic tissue;and the uterine tissues of postmenopausal women still retained highlevels of these sex steroid receptors.In addition,the regulation of sex steroids in thepathogenesis of myoma and adenomyosis is discussed.

      Abstract:<正> The changes of immunoreactive neurotensin(ir-NT)contents in the brain areas,pituitary gland and plasma in the traumatized rats were observed in the present study.The results of radioimmunoassay exhibited significant changes of the ir-NT contents inthe hypothalamus,pituitary gland,plasma,injured tissue,hippocampus,central gray andspinal cord in the posttraumatic rats at different intervals.A predominant characteristicsof the changes of ir-NT levels in the brain areas,pituitary gland and plasma was thedramatical decrease at various times except for the hypothalamus,central gray andhippocampus with biphasic alterations.The ir-NT contents in the frontal cortex andpons-medulla also displayed changes to different extent under the acute craniocerebraltrauma condition.These results suggest that NT may play a role in the pathophysiologyof traumatic head injury.

    • Distribution of peptidergic neurons in the hypothalamic paraventricular nucleus of the rat

      Abstract:<正> Distribution of peptidergic neurons in the hypothalamic paraventricular nucleus(PVN)of the rat was investigated by means of immunohistochemical technique,and thearea,perimeter,maximum diameter,minimum diameter and grey scale of peptidergicneuronal cell bodies were measured with an image analyser.All of these peptidergicneurons,oxytocin(OXY)-,vasopressin(VP)-,substance P(SP)-,corticotropin releasingfactor(CRF)-,thyrotropin releasing hormone(TRH)-,neurotensin(NT)-,cholecystokinin(CCK)-,somatostatin(SOM)-,galanin(GAL)-,leucine-enkephalin(L-ENK)-,vasoactive intestine polypeptide(VIP)-,and ACTH-like neurons,were ob-served in the PVN.The data of image analysis showed that the area of peptidergicneuronal cell bodies in the magnocellular subnucleus part of the PVN was similar,but wasdifferent in the parvocellular subnucleus part of the PVN.

    • Histological changes after tibial lengthening with metaphyseal osteotomy in goats

      Abstract:<正> The histological changes after tibial lengthening with metaphyseal osteotomywere observed in 30 goats.It was found that(1)a slow lengthening rate of 1 mm/d couldenable the tissues to adapt the mechanical traction and consequently increase thelengthening limitation;(2)the newly-formed bone in the lengthened area might be due to acombined osteogenesis of endochondral,intramembranous,and fibrous tissue ossification;and(3)the osteogenetic process could be distinguished into 3 stages as hemorrhage andorganization of the hematocele,callus formation,and remodelling of the newly-formedbones.

    • Recognition with monoclonal antibodies of a Mr 71000 surface antigen of Plasmodium falciparum merozoites

      Abstract:<正> Merozoite surface antigens can induce protective immune responses and may becandidate antigens for malaria vaccine.Four hybridoma cell lines secreting monoclonalantibodies against Plasmodium falciparum Fcc7801/HN were produced.Antibodies fromthree of the four lines showed significant growth-inhibiting effect on P.falciparum invitro.One monoclonal antibody,known as C6,conjugated the antigen located exclusivelyon the merozoite surface and distributed evenly over the entire surface,as wasdemonstrated by immunoelectron microscopy.C6 also precipitated a single protein of Mr71000.

    • Sequential check and computer analysis of a cloned DNA fragment specific for Plasmodium vivax

      Abstract:<正> A cloned DNA fragment specific for Plasmodium vivax was cleaved from theplasmid pVA1 and ligated into phage M13mp18.The nucleotides of the insert were se-quenced by the dideoxy-chain termination procedure.The fragment was composed of 261base pairs and 3 Hinf Ⅰ sites but no tandem repeat sequences.Computer retrieval andanalysis show that the nucleotide sequence of the cloned DNA fragment is unique,forthere has not been any significant homologue with that of other Plasmodium genes pub-lished as yet.

      Abstract:<正> Immunoreactive substance P(Ir-SP)level,and substance P likeimmunoreactivity(SP-Li)in the spinal cord were observed with radioimmunoassay andimmunohistochemistry in rats after they were given an intraperitoneal injection of mor-phine(7.5mg/kg)or electroacupunctured(3V and 3Hz)on the “Jiaji point”.It wasfound that the pain threshold(PT),Ir-SP level and SP-Li in the dorsal horn of the spi-nal cord were more significantly increased in the animals after the administration ofmorphine or electroacupuncture than in the control(P<0.05~0.01).The combined effectsof morphine and electropacupuncture were even more powerful than either of the agentswas administered singly.Naloxone could block the analgesic effect and the elevation ofIr-SP due to morphine or electroacupuncture.The findings suggest that there is a synergismbe tween morphine and electroacupuncture and the analgesic effect of the 2 depends uponthe increase of Ir-SP level of the spinal cord mediated through the opiate receptors.

      Abstract:<正> Earthworm capsule is an extract from earthworm made in our laboratory.From1986 to 1989,486 patients with carcinoma of esophagus or lung were randomly divided into 2groups,one group received radiotherapy alone(control group)and the other radiotherapycombined with earthworm capsules(treatment group).All cases were diagnosedpathologically or cytologically.In a two-month treatment course,earthworm capsuleswere given orally twice a day,2~3 capsules each time,each capsule containing 60 mg ofthe extract.Compared with the control group,the CR(complete remission)rate andCR+PR(partial remission)rate in patients with carcinoma of esophagus treated byradiotherapy in combination with earthworm capsule increased 9.7%(P<0.01) and 9.6%(P<0.05),while in patients with carcinoma of lung,they increased 10.6%(P<0.05)and13.5%(P<0.05),respectively.The earthworm capsule caused irritation in the alimentarytract,the patients displaying slight nausea and vomiting(16.7% in thecombination treatment group and 10.0% in the control group).However,these side ef-fects could be alleviated when capsules were taken after meal or with honey concomitantly.No deleterious effects on the heart,liver,kidney,skin,hematopoietic or nervous systemwere seen.

    • Enhanced myocardial protection with Ginsenosides in St.Thomas’ Hospital cardioplegic solution

      Abstract:<正> Several studies suggest that Ginsenosides have a protective effect on myocardialischemia and reperfusion injury.Using the isolated working rat heart model the effect ofthe St.Thomas’ Hospital cardioplegic solution containing different concentrations ofGinsenosides on the enhancement of myocardial protection following hypothennic ischemicarrest(20 ℃,120min)was evaluated.The concentrations of Ginsenosides in thecardioplegic solutions were 0(control),50,100,and 150μg/ml,respectively.Recoveries ofaortic flow,cardiac output,and minute work markedly improved by adding Ginsenosidesto cardioplegia.Dose-response tests indicated the best protective effect in the group using100 μg/ml,where the generation of thiobarbituric acid reactive materials(malondialdehyde,MDA)markedly reduced.These results showed that Ginsenosides may enhance themyocardial protection of the St.Thomas′ Hospital cardioplegic solution,and that additionof Ginsenosides to cardioplegia may protect the ischemic myocardium from free-radical in-jury.

    • Application of α-globin 3′hypervariable region to gene diagnosis of adult polycystic kidney disease

      Abstract:<正> Seventy-four members from 9 adult polycystic kidney disease(APKD)familieswere analysed with Southern blot and 3′hypervariable region/Pvu Ⅱ,3′HVR/Bgl ⅡRFLPs.The results showed that 3′ HVR fragments were highly polymorphic.Theheterozygote frequencies of Pvu Ⅱ,Bgl Ⅱ and PvuⅡ+Bg1Ⅱ fragments were 92%,84.7%and 98.6%,respectively.The maximum lod score for linkage between 3′HVR and APKDwas 9.71 at a recombination fraction 0.045.We successfully applied 3′HVR probe to thegene diagnosis of 17 symptomatic patients and 7 patients in the presymptomatic stage.

      Abstract:<正> Male mice were subjected to 6 Gy total body irradiation,20% TBSAfull-thickness burns,or combined radiation-burn injury and lipid peroxides(LPO),vita-min E,sulfhydryl group,respiratory control ratio(RCR),ADP/O ratio,and cytochromeoxidase activity of the liver mitochondria were determined in the first 9 d postinjury.Theresults are as follows:(1)LPO level increased in the early postinjury stage after combinedradiation-burn injury,on the 5th-7th day after irradiation and on the 7th day postburn.(2)Vitamin E level decreased significantly in the two groups of radiation and burn inju-ries but showed no significant decrease after combined injury.(3)The sulfhydryl groupshowed a tendency to increase in all the 3 groups.(4)The activity of cytochrome oxidaseincreased significantly on the 7th day after radiation but decreased considerably in theburn and combined injury groups.(5)RCR and ADP/O ratio decreased more significantlyin the combined injury group than in either the radiation group or the burn group.These facts suggest that the respiratory dysfunction of the liver mitochondria results mostprobably from the damage on the mitochondrial membrane due to lipid peroxidation.
