    • Seven amino acid peptide mimic from HVR1 of HCV influences cytokine profile of immune cells

      Abstract:Objective:To investigate the actions of cytokine profile in the immune cells by a seven amino acid peptide mimic from HVR1 of HCV (GQTYTSG,named 7P).Methods:The peripheral blood mononuclear cells (PBMCs) from 17 healthy blood donors were stimulated with 7P,and the cytokine levels in CD4+CD8-,CD4-CD8+ T cells,NK,NKT cells were analyzed by the intracellular cytokine staining.Results:The frequency of cells which secreted interferon-γ (IFN-γ) was found to be significantly increased in all cells,interleukin-10(IL-10) was significantly increased in CD4+CD8-,CD4-CD8+ T cells but decreased in NK,NKT cells,and tumor necrosis factor-α (TNF-α) was decreased in CD4+CD8-,CD4-CD8+ but increased in NK cells.Conclusion:7P could induce a cytokine profile in different immune cells in vitro and there was some difference between the CD4+CD8-,CD4-CD8+ T cells which represented the adaptive immunity cells and NK,NKT cells which represented the innate immunity cells.This kind of variation of cytokine profile might contribute to anti-virus and anti-inflammatory immune reaction.

    • The effect of alphastatin peptide suppressing the hypoxia-induced vasculogenic mimicry formation of glioma

      Abstract:Objective:To investigate the vasculogenic mimicry formation induced by hypoxia in Ⅱ-Ⅲ human glioma cell and the effect of alphastatin peptide suppressing the hypoxia-induced vasculogenic mimicry formation and the mechanism.Methods:MTT,Transwell and three-dimentional culture were used to detect the proliferation,migration and tubule formation of SHG44.The expression of vascular endothelial growth factor-α(VEGF-α),erythropoietin-producing hepatocellular carcinoma-A2 (EphA2) and matrix metalloproteinases 2 (MMP2) was detected by RT-PCR and Western blotting analysis.Results:The OD 490 in hypoxia group was 0.60±0.06 and in control group was 0.46±0.05.The number of cell migration was 178.71±18.81 in hypoxia group and 85.86±17.92 in control group.The tubule formation was 56.80±12.21 in hypoxia group and 4.20±2.62 in control group.The proliferation,migration and tubule formation in hypoxia group were significantly higher than that in control group.The expression of VEGF-α,EphA2 and MMP2 was upregulated in hypoxia.When various concentrations of alphastatin (100,1 000,10 000 nmol/L) were added to hypoxia group,the numbers of cell migration were 142.57±12.12,92.71±17.68,30.00±7.72 and the tubule formation were 47.71±10.58,18.86±8.40,8.43±5.62.The cell migration and tubule formation were significantly suppressed by alphastatin in a dose-dependent manner.In alphastatin group,the phosphorylation of EphA2 protein (P=0.037,F=4.629) and activation of MMP2 protein (P=0.005,F=9.331) were significantly suppressed but there was no change in VEGF-α protein.Conclusion:Ⅱ-Ⅲ human glioma cell is able to form vasculogenic mimicry induced by hypoxia and alphastatin peptide can suppress the hypoxia-induced vasculogenic mimicry.VEGF-α induced EphA2 phospharilation and MMP2 activation maybe the key pathway to form vasculogenic mimicry.

    • P2X4 receptor and brain-derived neurotrophic factor in neuropathic pain

      Abstract:Objective:To investigate whether the activation of p38MAPK is involved in the neuropathic pain induced by P2X4 receptor,and the effects of activated P2X4 receptor and p38MAPK on expression of brain-derived neurotrophic factor (BDNF) in the chronic neuropathic pain.Methods:Lumbar intrathecal catheters were chronically implanted in male Sprague-Dawley rats.The right sciatic nerve was loosely ligated proximal to the sciatica’s trifurcation at approximately 1.0 mm intervals with 4-0 silk sutures.The microglia inhibitor minocycline,P2X4 antagonist (TNP-ATP) and p38MAPK inhibitor (SB203580) were intrathecally administered every 12 h,3 d post-chronic constriction injury (CCI).Mechanical nociceptive thresholds were assessed with the paw withdrawal threshold (PWT) to von Frey filaments.The expression of P2X4 and BDNF were assessed by both immunohistochemical analysis and RT-PCR.Results:Intrathecal injection of minocycline or TNP-ATP or SB203580 significantly attenuated CCI-induced mechanical allodynia.The time courses of P2X4 receptor and BDNF expression were increased at all points after CCI and reached a peak level on postoperative d 7.Intrathecal injection of minocycline or TNP-ATP or SB203580 markedly suppressed the increase of CCI-induced P2X4 receptor and BDNF expression in the spinal cord.Conclusion:The activation of P2X4 receptor BDNF pathways contributes to neuropathic pain in CCI rats,and the activation of p38MAPK is involved in the neuropathic pain induced by P2X4 receptor.

      Abstract:Objective:To evaluate the technical aspects,efficacy and safety in the treatment of cavernous dural arteriovenous fistulae (DAVF) by transvenous liquid n-Butyl cyanoacrylate (NBCA) injection combined with coils placement in the cavernous sinus.Methods:Eight patients with cavernous DAVF,treated by transvenous embolization with combination of detachable coils and glue between February 2006 and February 2009 in our hospital,were retrospectively analyzed,including the clinical presentations,patterns of angioarchitecture,methods of treatment and the results of follow-up.Results:In all 8 patients,9 transvenous and 1 transarterial interventions were carried out.A single endovascular procedure was performed in 6 patients and 2 patients had to be treated twice.Of the nine transvenous approaches,eight approaches were performed via inferior petrosal sinus (IPS) and 1 via the facial vein.Complete angiographic obliteration was achieved in all patients immediately after the embolization.There was no procedure-related morbidity except for mild headache for one to seven days after the embolization.Clinical cure was achieved in all cases during the clinical follow-up periods,ranging from 8 months to 3 years.The follow-up angiography periods averaged 6.6 months with a range of 5-9 months,in which no recurrence was observed.Conclusion:In this small series,embolization with combination of glue and detachable coils by transvenous approaches was a safe,effective and economical method for the treatment of part of symptomatic patients presenting with complex cavernous DAVF.

      Abstract:Objective:To determine whether maternal β-human chorionic gonadotropin(β-hCG) level in second-trimester may be associated with subsequent development of pregnancy-induced hypertension(PIH).Methods:Seven hundred and sixty-two women in mid-trimester were to have maternal urine β-hCG standardized concentrations and maternal serum β-hCG measurements.Their case histories were recorded and reviewed from mid-trimester to delivery.The relation was observed between maternal urine,serum markers and subsequent development of PIH.Results:Among 762 women,504 cases were normal pregnancies,42 cases had PIH,94 cases had premature rupture of membrane (PROM),69 cases had preterm delivery (PD),53 other cases were excluded by various reasons.The levels of maternal urine,serum β-hCG in PIH were (61.75±9.78) IU/L and (304.56±54.17) ng/mg respectively,which were higher significantly than normal pregnancy group ([20.65±7.61] IU/L and [146.34±47.81] ng/mg,P<0.05).When maternal serum,urine β-hCG levels ≥2 MOM(multiple of mean),the incidences of developing PIH were increased significantly as compared with those of β-hCG <2 MOM women.The incidence of PIH increased from 5.1% in pregnancies with urine β-hCG ≥2 MOM to 11.7% in cases with urine β-hCG ≥4 MOM.Conclusion:The elevation of maternal mid-trimester urine,serum β-hCG levels is not only an early signal for dysfunction of placenta but also a dangerous signal for development of PIH.Second-trimester maternal urine β-hCG measurement proves to be superior to serum marker in clinical prediction.

      Abstract:Objective:To evaluate the efficacy of entecavir and adefovir dipivoxil on HBeAg-positive nucleos(t)ide-naive patients with chronic hepatitis B with the method of Meta analysis.Methods:We searched PUBMED,EMBASE,CNKI (China National Knowledge Infrastructure),the Cochrane Central Register of Controlled Trials and the Cochrane Database of Systematic Reviews with reference to all data documented before May 2010.The dosage of entecavir and adefovir dipivoxil was 0.5 mg/d and 10 mg/d,respectively.Heterogeneity was examined by Chi-square test,the relative risk calculated and forest plot drawn.Rates of undetected serum HBV DNA,serum alanine aminotransferase (ALT) normalization,HBeAg clearance,HBeAg seroconversion and adverse effect occurrence were analyzed.Results:Six articles were included,which fit well in with this study.Meta analysis showed that the rate of undetected serum HBV DNA(P=0.000 2 at week 12,P=0.002 at week 48)and that of serum ALT normalization(P=0.04 at week 12,P=0.008 at week 48)in the entecavir group were higher than those in the adefovir dipivoxil group.However,no statistic significance existed between the two groups in the rate of HBeAg clearance (P=0.17),the rate of HBeAg seroconversion(P=0.53)or the rate of adverse effect occurrence(P=0.92)at week 48.Conclusion:Entecavir was superior to adefovir dipivoxil in decreasing serum HBV DNA and normalizing serum ALT in the HBeAg-positive nucleos(t)ide-naive patients with chronic hepatitis B.

    • Proline-rich tyrosine kinase 2 and its inhibitor PRNK

      Abstract:Proline-rich tyrosine kinase 2 (Pyk2) is a nonreceptor protein tyrosine kinase,which is also known as Ca2 +-dependent tyrosine kinase or related adhesion focal tyrosine kinase.Pyk2 activation exerts a critical regulatory mechanism for various physiological processes including cytoskeleton function,regulation of cell growth and death,modulation of ion channels and multiple signaling events.However,mechanisms underlying the functional diversity of Pyk2 are not clear.A Pyk2 isoform that encodes only part of the C-terminal domain of Pyk2,named as PRNK (Pyk2-related non-kinase),acts as a dominant-negative inhibitor of Pyk2-dependent signaling by displacing Pyk2 from focal adhesions.Research on functional PRNK probably provides new potential inhibitory tool targeting Pyk2 and makes it possible to explore more of Pyk2 pathological mechanism.PRNK is a promising candidate targeting Pyk2 modulation.This review focuses on the functional investigation of Pyk2 and its structure and localization,including recent research with inhibitory strategies targeting Pyk2 by the method of PRNK.

      Abstract:We describe a 70-year-old female patient with diffuse large B-cell lymphoma of the cerebellum;she received resection of a left atrial myxoma five months ago.To the best of our knowledge,to date no association of atrial myxoma with the malignant cerebellar diffuse large B-cell lymphoma has been reported in the literature,except for high level of interleukin-6 (IL-6) in the serum under both conditions.IL-6 as a pro-inflammatory cytokine has been shown to be associated with tumor progression,including inhibition of cancer cell apoptosis and stimulation of angiogenesis.IL-6 concentrations may provide possible aetiological links between atrial myxoma and cerebellar diffuse large B-cell lymphoma in our case.

      Abstract:Myelolipoma of the extra-adrenal gland is a rare,benign tumor comprising mature fat tissue and hematopoietic elements.Most myelolipomas are asymptomic and discovered incidentally.We presented a 44-year-old male who was admitted to our hospital for right kidney dystrophy and renal calculus.Ultrasound examination of right kidneys showed hydronephrosis.Intravenous pyelography showed the right renal contour was unclear,with a circular high-density shadow inside.The nephrectomy was performed with renal myelolipoma as a final pathologic diagnosis.
