Abstract:<正>Objective:To evaluate the effects of curcumin on regulating the proliferation,cell cycle distribution,apoptosis and relevant mechanisms in keratinocyte cell lines.Methods:The human immortalized human keratinocyte lines(HaCaT cells) were treated with different doses of curcumin.The effects of curcumin on cell viability were measured by MTT assay,and the cell cycle distribution and apoptosis determined by flow cytometry.The mRNA expression changes of proliferating cell nuclear antigen(PCNA),cyclin D1 and Bcl-xL were from real-time PCR analysis and the protein levels were detected by Western blotting.Results:Data obtained in the study showed that curcumin could cause significantly inhibitory effect on proliferation in HaCaT cells in a time- and dose-dependent manner.Cell arrest at G1/S phase and significant apoptosis were observed after being treated with curcumin for 24 h.In association with these,the expression of PCNA,cyclin D1 and Bcl-xL were decreased both at mRNA and protein levels for the same treatment.Conclusion:Curcumin can inhibit proliferation,induce cell arrest at G1/S phase and cause apoptosis in HaCaT cells.The decreased expression of PCNA,cyclin D1 and Bcl-xL induced by curcumin contributes to the above effects in vitro.

Abstract:<正>Ubiquitous on earth,bacteriophages are the most abundant entities in every ecosystem,but human knowledge about them is still limited compared with that about other forms of organisms.To enrich human knowledge and promote the utilization of bacteriophages,it is necessary to isolate and characterize as many as possible different bacteriophages.Here we describe the isolation of a T4-like bacteriophage IME08 and a rapid method for its genetic characterization.With this method we easily cloned a few random fragments of the bacteriophage genome.Sequence analysis of these random clones showed that bacteriophage IME08 shared the highest sequence similarity with T4-like Enterobacteria phage T4(94%identity),JS98(95% identity),JS10(95%identity) and RB14(94%identity) respectively,which suggested that IME08 belonged to T4-like bacteriophage genus.

Abstract:<正>Objective:To ascertain the variations of mitochondrion DNA(mtDNA) in mouse tumors and to inquire into the relationship between mutations of mtDNA and carcinogenesis.Methods:The variations of D-loop,ND3 and tRNAMet+Glu+Ile gene fragments of mtDNA from six tumor cell lines of mice were analyzed by PCR technology with restriction fragment length polymorphism analysis(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP) and single strand conformation polymorphism analysis(SSCP-PCR) method.Results:ND3 and tRNAMet+Glu+Ile gene fragments of mtDNA from the tumors showed no variation in 27 endonuclease sites;D-loop of mtDNA from Hca-F had an additional endonuclease sites of HinfⅠin contrast to that of the inbred mouse.Deeply analyzed by PCR-SSCP,the D-loop of mtDNA was found to possess mutations in 4 of 6 tumors.Conclusion:D-loop is the hot spot of tumor mtDNA mutation which can act as contributors to the carcinogenic process.

Abstract:<正>Objective:To establish an experimental model of urethral stricture in rabbits.Methods:A total of 21 adult male New Zealand rabbits were included into group.After intravenous anesthesia,urethroscopy was performed with a pediatric resectoscope(F13).Fifteen animals were randomly selected as the study group.A 1 cm-long circumferential electrocoagulation of the bulbar urethra was performed to these animals until ulceration of the mucosa.The remaining 6 animals underwent video urethroscopy without electrocoagulation,serving as controls.On the 30th day,retrograde urethrogram and urethroscopy were performed to evaluate urethral stricture formation,histological examinations(HE and Sirius Red staining) were done to assess urethral pathological change.Results:Two rabbits in study group died and no death occurred in controls.Based on urethrogram and urethroscopy,no rabbits in control group developed urethral stricture,while significant stricture formation was observed in every case of the study group.Histological examination showed a normal urethra in control cases,while at stricture site of the study group extensive fibrosis of muscle and submucous tissue was observed with a large number of fibroblasts infiltration. High collagen expression in fibrosis tissue was assayed by Sirius Red staining.Conclusion:A 1cm-long endoscopic electrocoagulation can successfully induce urethral stricture formation in rabbit models.This method offers an ideal animal model for the fundamental and clinical study of urethral stricture.

Abstract:<正>Despite the considerable mortality and morbidity in metastasis cardiac mass,antemortem diagnosis is unusual.Here we report a case of a left atrial mass echocardiographically mimicking myxoma,in a patient with lung carcinoma.The mass was pathologically confirmed to be metastatic carcinoma,which had entered the left atrium via direct invasion of left superior pulmonary vein,illustrating the potential for unusual routes of tumor spread into the heart and the importance of obtaining a tissue diagnosis in such cases.
Abstract:<正>A 27 year-old male patient was diagnosed with Beh(?)et’s disease(BD) and posterior visual pathway affection(visual cortex),which resulted in reduced visual acuity and field of vision.This case demonstrates the effect of tumor necrosis factor (TNF) alpha blocker therapy on Beh(?)et’s cerebral vasculitis.

Abstract:<正>Hepatocellular carcinoma is a highly malignant neoplasm and frequently involves extrahepatic organs but decidedly rarely occurs in brain.We describe 3 cases of brain metastases in patients suffering from post-HBV hepatocarcinoma.The"stroke-like"presentation of the cerebral localization of the disease can be explained by both the important vascularization of the tumor and the frequent hemocoagulative alterations caused by the cirrhosis.The importance of diagnostic neuroradiology is briefly addressed,with reference to the fundamental role played by MRI.Surgery of these lesions does not present any particular technical problems as long as they are located in accessible areas and the patient’s general and neurological conditions allow it.Postoperative radiotherapy seems to improve the quality and quantity of residual life,although the number of patients described in the literature was too small to draw any definite conclusion in this regard.

Abstract:<正>Drug-induced acute pancreatitis(DAP) is defined as pancreatitis with corresponding clinical manifestations resulted from drug-induced pancreatic secretion dysfunction and pancreatic tissue damage.One case of DAP resulted from chemotherapeutics (Nimustine) was diagnosed and treated our in hospital in 2009.This patient belonged to pancreatitis induced by anticancer drugs,and the toxicity of anticancer drugs acted directly on pancreatic cells,leading to the occurrence of pancreatitis.After treatment,the pancreatitis was effectively treated in this patient,but the final the patient and his family eventually gave up the treatment due to aggravated primary diseases.
