Abstract:Objective:To address how genistein sensitizes the chemotherapy-resistant ovarian carcinoma cells and promotes apoptosis in the respect of cell cycle and the regulation of survivin expression in the process.Methods:Ovarian SKOV-3 carcinoma cell line was treated with genistein or cisplatin either alone or in combination.Cell viability was showed by MTT method.Cell cycle and apoptosis were detected by flow cytometry.Survivin mRNA and protein were revealed by RT-PCR and immunocytochemistry,respectively.Results:Genistein could reduce the cell viability in a dose-dependent manner,while cisplatin did so at a much higher level.In contrast,if the two agents were treated in combination,half growth inhibition(IC50) value for cisplatin was reduced remarkably and the effect was synergistic as analyzed by isobologram.In particular,the reduced cell viability was exhibited by a switch in cell cycle progression,as the cells were arrested in G2/M phase and the G0/G1 phase-fraction was significantly decreased.The reduced cell viability appeared to involve apoptosis,based on our results from flow cytometry and Hoechst 33258 staining.In the meanwhile,genistein performed the inhibitory effect on cisplatin-induced survivin expression.Conclusion:Genistein can sensitize ovarian carcinoma cells to cisplatin therapy with the inhibition of survivin expression as the potential mechanism.

Abstract:Objective:To investigate the alternations of proteins in the colonic mucosa of chronic slow transit constipation(STC) rats with a 2-DE-based proteomic method and analyze the function of these down-regulated proteins so as to provide theoretical basis for the pathogenesis of intestinal mucosa of chronic STC rats.Methods:STC model was established by feeding rats with 8 mg/(kg·d) diphenoxylate for 120 d.An experimental model of chronic STC rat was used for separation of proteomics from colonic mucosa using two-dimensional electrophoresis(2-DE).Proteins altered in expressional level were identified by Image Master 2DElite,mass spectrometry,and bibliometrics were applied to identify the differential protein expression and their clinical significance and function were analyzed.Results:Obvious differential protein expression was observed in the pathogenesis of STC,including mast cell protease(A1),non-specific dipeptidase(A2) and chondrosome succinate dehydrogenase precursor(A3).The expressions of A1,A2 and A3 were down-regulated in the gel graph of STC rats.Conclusion:The down-regulation of chondrosome succinate dehydrogenase,mast cell protease as well as non-specific dipeptidase in rat colon suggests the functional impairment of the oxidoreduction of mitochondrion is very important in the genesis and development of STC.The immunological reaction of STC rats is weakened,and the function of digesting and absorbing protein may be damaged to some extent.

Abstract:Objective:To analyze the antifungal effects of Chinese herb monomers,i.e.berberine,baicalin,eugenol and curcumin,on Candida albicans.Methods:After Candida albicans strain Y01-09 was incubated for 48 h in YEPD broth which contained different concentrations of Chinese herb components,the cell cycle,fluorescent intensity and the size of cell volume were detected by flow cytometry.Results:The 4 Chinese herb monomers could affect the cell cycle of Candida albicans in different ranges.The ratio of cells in S-G2-M period decreased as the agents concentration increased,indicating that the cell division was inhibited.The fluorescent intensity of Candida albicans cells became weaker after being incubated,which reflected the loss of DNA fragments.The higher the concentration was,the weaker the fluorescent intensity became.The cell size,cell diopter and particle size changed much as the agents concentration increased.Conclusion:Chinese herb monomers play the antifungal role in inhibiting cell division.FCM could be used to determine the susceptibility of antifungal agents.

Abstract:Objective:To explore the effects of Res on the proliferation and apoptosis of human cervical cancer cell lines C33A,SiHa and HeLa.Methods:Inhibition rates by different concentrations of Res were calculated using MTT method.Apoptosis rates and cell cycles were measured and examined by flow cytometry(FCM).Morphological configuration of apoptotic cells were observed under the fluorescence microscope.Results:The growth of cancer cells was inhibited by Res of varied concentrations in a time-and dose-dependent manner(P<0.01).The cells showed characteristic apoptosis morphologic changes under fluorescence microscope.Res exerted no effects on cell cycles.Conclusion:Res inhibits the growth of cervical cell lines C33A,SiHa and HeLa by inducing cell apoptosis in a time-and dose-dependent manner.

Abstract:Objective:To investigate the therapeutic effects of dexamethasone,anisodamine and rhubarb(DAR) on endotoxin,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and pancreatic damage in rat models of acute pancreatitis(AP).Methods:The AP rat models were prepared and randomly assigned to AP group(n=10) and DAR group(n=10),while other healthy rats were assigned to the sham-operated group(n=10).The rats were euthanized at 6 h after operation,and then the serum levels of endotoxin,TNF-α,IL-6 and histology of pancreas were determined as the indexes of therapeutic effects.Results:At 6 h after operation,serum levels of endotoxin,TNF-α and IL-6,and pancreatic damage were significantly increased in AP group compared with those in sham-operated group(P<0.01).Compared with the AP group,DAR therapy remarkably attenuated the endotoxin,TNF-α,IL-6 levels and reduced pancreatic damage(P<0.05).Conclusion:The inhibition of pancreatic damage by DAR in rats with AP might contribute,in part at least,to the amelioration of pancreatic inflammation.The present study provides beneficial evidence that DAR may be useful in the treatment of AP model of rats.

Abstract:Objective:To assess the inhibitory effects of liposomal adriamycin(LADR) locally injected into mammary glands of VX2 tumor-bearing rabbits on proliferation of lymph nodal metastatic cells.Methods:Twenty-one VX2 tumor-bearing rabbits were randomly and equally divided into 3 groups.Rabbits were randomized to receive sham treatment(Group I),subcutaneous LADR around tumor(Group II) and intravenous free adriamycin(Group III),respectively.Breast tumor and axillary lymph nodes were harvested after 3 repeated treatment.Nodal sizes of both pre-and post-treatment were measured.Proliferating cell nuclear antigen(PCNA) mRNA in both tumor and lymph nodes were determined by RT-PCR.Results:The mean size of axillary lymph nodes in Group I,II and III increased by 3.70%,1.55% and 2.89%,respectively,with significant difference between Group III and I(P=0.004) and between Group II and III(P=0.002).Relative expression values of PCNA mRNA in breast tumors of Group I,II and III were 0.486,0.513 and 0.396,respectively.For Group III,PCNA mRNA was significantly less expressed than that in Group I(P=0.023) and II(P=0.005).Relative expression values of PCNA mRNA in axillary lymph nodes of Group I,II and III were 0.541,0.329 and 0.450,respectively.Compared with Group I,Group III showed a markedly decreased expression of PCNA(P=0.021).The least level of PCNA mRNA was found in Group II,with a significant difference from that in Group III(P=0.004).Conclusion:Local injection of LADR was an effective therapeutic regimen for lymphatic metastases from breast cancer,regardless of its little effect on primary tumor.

Abstract:Objective:To make up a portable,economic drainage device to prevent the development of pocket hematoma and avoid the additional therapies of pocket hematoma.Methods:Between 2003 and 2006,a total of 265 devices were implanted at our institution.The 89 high-risk patients were determined by the predictors of hematoma occurrence(marasmatic elder,inevitable oral antiplatelet/anticoagulation therapy,venous pressure increased by other comorbidity,the deficiency of the clotting mechanism for hepatic diseases,or incognizable severe intraoperative bleeding),and other 186 patients were included in non-high-risk group.The 89 high-risk patients were randomized into treatment and control subgroups by sortition.Surgical procedures differed only by the application of the portable,economic drainage device prior to wound closure in treatment subgroup.Results:The incidence of pocket hematoma was 4.3% in treatment subgroup,18.6% in control subgroup and 2.7% in non-high-risk group,leading to 2,6 and 3 patients’ prolonged hospitalization,respectively.The additional cost due to pocket hematoma was lower(1.5 times) in the treatment group compared to the control group.There wasn’t antidromic infection and delayed cure of the skin incision with the use of our drainage device within 6 months.Conclusion:Our portable drainage device was made up easily and quickly.It could decrease the total cost of hospitalization,did not increase the other adverse events and seemed to be suitable for such patients with a tendency to develop pocket hematoma undergoing the implantation of pectoral pacemakers,implantable cardioverter defibrillator,or cardiac resynchronization therapy.

Abstract:Objective:To explore the construction of metastatic spinal cancer(MSC) tissue microarrays and validate its value in immunohistochemical study of MSC.Methods:Paraffin-embedded specimens from 71 MSC cases and 6 primary tumor cases were selected as donor blocks and prepared into MSC tissue microarrays by tissue array arrangement,the steps of which included location,punching,sampling,sample seeding,and re-diagnosis by hematoxylin-eosin(HE) as well as MMP-9 and MMP-14 immunohistochemical staining.Results:The MSC tissue microarrays thus constructed were intact and crackless,containing 154 complete and well arranged microarray points.None of the sectioned tissue microarrays was lost,and the results of HE staining was consistent with the primary pathologic diagnoses.Immunohistochemical staining was also good without non-specific or marginal effect.Conclusion:The MSC tissue microarrays have a high value in the immunohistochemical study of MSC.

Abstract:Objective:Uterus-to-abdominal-wall fistula is a very rare complication after uterine-incision delivery over the last decades.It can even lead to death.Mainly,the fistula occurs when big tension and critical infection exist within the incision of uterus and abdominal wall.Methods:The authors described the clinical presentation,pathology of uterus-to-abdominal-wall fistula,and reported their experience in 6 cases who underwent Z-plasty operation for this rare complication from January 1998 to January 2008.Results:All flaps survived completely and all the wounds in abdominal walls healed very well.The six cases were followed up for 1 to 5 years,and no fistula recurrence occurred.Conclusion:Z-plasty technique is a very simple and efficient approach to repair uterus to abdominal fistula after uterine-incision delivery.
