Abstract:<正>Objective:To investigate whether the rabbit serum after radiofrequency ablation to liver tumor can induce mesenchymal stem cells (MSCs) differentiating into hepatocyte-like cells in order to find a new source and culture process for repairing liver injury.Methods:A tumor piece of 1 mm×1 mm×1 mm was transplanted into a tunnel at right liver of rabbits.The model of liver tumor was established after 2-3 weeks.The serum was collected from rabbits 72 h after being subjected to radiofrequency ablation of the liver tumor.Mesenchymal stem cells were isolated from rabbit bone marrow and cultured in DMEM containing autologous rabbit serum.Three kinds of media (L-DMEM) were tested respectively:①containing 10% fetal calf serum (FCS);②containing 30% rabbit autologous serum after radiofrequency ablation of the liver tumor (ASRF);③containing 30% rabbit autologous serum (AS).MSCs were cultured on 12-well plates until passage 2 and examined under the light and electron microscopy at indicted intervals. The expression of albumin and CK18 was detected using immunofluorescence to identify the characteristics of differentiated cells.Results:MSCs performed differently in the presence of fetal calf serum,rabbit autologous serum and rabbit autologous serum after radiofrequency ablation of the liver tumor.Induced by the serum after radiofrequency ablation to liver tumor for 7 d,the spindle-shaped MSCs turned into round shaped and resembled hepatocyte-like cells. The reactions were not found in MSCs cultured in FCS and AS groups.After induction for 14 d,slender microvilli, cell-cell junction structure and cholangiole emerged,and the differentiated cells expressed albumin and CK18.All those could not been observed in 10% FCS and 30% autologous serum groups.Conclusion:Mesenchymal stem cells differentiate into hepatocyte-like cells in the serum after radiofrequency ablation of liver tumor,providing us a potential cell source and culture process for clinical application in liver injury repairing.

Abstract:<正>Objective:To explore the role of activated liver X receptor a (LXRα) on the expressions of interleukin-1 receptor associated kinase-4 (IRAK-4) and NF-kappaB (NF-κB) in the inflammatory response which induced by LPS in the Kupffer cells and to investigate the possible mechanisms of LXRαnegative regulation of inflammatory response. Methods:The Kupffer cells were isolated from male Kunming mice by collagen perfusion in situ.And these cells were divided into 4 groups:normal control group,LPS treatment group,LXRαagonist T0901317 treatment group,LPS and T0901317 combined treatment group.The LPS treatment group were treated with a final concentration of 1μg/ml LPS in RPMI 1640 and cultured for 6 h,the T0901317 treatment group were treated with a final concentration of 5μg/ml in RPMI 1640 and cultured for 24 h,and the combined treatment group received pre-culture for 24 h with a final concentration of 1μg/ml T0901317 in RPMI 1640 and then cultured for 6 h with a final concentration of 5μg/ml LPS in RPM! 1640.All groups were cultured for 30 h.The expression of LXRα,IRAK-4 and NF-κB at mRNA and protein levels were detected by real-time PCR and Western blotting,and the TNF-αand IL-1βlevels were detected by ELISA. Results:The levels of LXRαmRNA and protein were highest in T0901317 group,and lowest in LPS group (P<0.05). The level of IRAK4 and NF-κB mRNAs and proteins were evidently lower in the Combined-treated group than in LPS group (P<0.05).And the level of TNF-αand IL-1 were observed highest in LPS group (P<0.05),but no difference among the Control group,T0901317 group and Combined-treated group (P>0.05).Conclusion:These date suggest that the LXR agonists can effectively up-regulate the expressions of LXRαmRNA and protein and inhibit the inflammatory response.This may be via down-regulating the expressions of IRAK4 and NF-κB at mRNA and protein levels.

Abstract:<正>Objective:To explore the role of transcription-coupled repair (TCR) pathway in the UVC-induced SupF gene mutation in Tet-on 293 cell line,we designed and constructed a Tet-responsive plasmid DNA pTCR-C1,and utilized this pTCR-C1 plasmid to obtain the mutation frequency of SupF reporter gene induced by UVC in Tet-on 293 cell line. Methods:SupF gene was cloned into a Tet-responsive plamid pBI-L,which include a bidirectional Tet-responsive promoter,and was named pTCR-C1.The pTCR-C1 plasmid was transfected into Tet-on 293 cell line,and the mutation frequency of SupF reporter gene was detected in the presence and absence of DOX.Results:The pTCR-C1 plasmid was identified with the methods of restriction digestion and DNA sequencing.The mutation frequency of SupF reporter gene in the presence of DOX was higher than in the absence of DOX.Conclusion:The TCR pathway takes part in the UVC-induced SupF gene mutation in Tet-on 293 cell line.

Abstract:<正>Objective:To screen the proteins interacting with the Treg specification factor forkhead box protein P3 (FOXP3) by yeast two-hybrid system.Methods:Human FOXP3 gene was amplified by nest RT-PCR from peripheral blood mononuclear cells (PBMC) and inserted into plasmid pGBKT7 to construct the bait vector,then the self-activation and toxicity of the bait vector in host yeast strain AH109 were observed.Thereafter,a human liver cDNA library was screened by the bait vector.The positive clones were selected out by nutrient-deficient culture and back-hybridizing.The sequences from the candidate positive clones were blasted and analyzed by bioinformatics methods.Results:The constructed bait vector encoding FOXP3 was found no self-activation and toxicity in yeast AH109.Three proteins which interacted with FOXP3,including tumor protein D52,splicing factor 3b subunit 1 and hypothetical protein,were identified.Conclusion:Three new candidate proteins interacting with FOXP3 are selected out by this yeast two-hybrid system and library,which may facilitate the further study of FOXP3 in Treg.

Abstract:<正>Objective:To study the anti-fatigue effects of salidroside in mice.Methods:Totally 120 normal male Kunming mice were randomized into 5 groups (4 salidroside intervention groups and the control group) based on body weight. The control group was given distilled water and the 4 intervention groups were given various doses of salidroside (60, 180,360,720 mg/kg) for 15 consecutive days,respectively.The levels of lactate,serum urea nitrogen,muscle and liver glycogen,the longest swimming time and hemoglobin were determined before and after swimming test.Results: Different doses of salidroside significantly lengthened the swimming time and increased the contents of hemoglobin and muscle and liver glycogen,while reducing that of lactate in blood significantly compared with control group, especially in the 180 mg/kg salidroside group.Conclusion:Salidroside has noticeable anti-fatigue effect on mice. These effects were dose-dependent,and the strongest effect on most biomarkers was seen with an intermediate dose.

Abstract:<正>Objective:To clone and construct the recombinant plasmid containing ATP synthase lipid-binding protein-like protein gene of Schistosoma japonicum,(SjAslp) and transfer it into mammalian cells to express the objective protein. Methods:By polymerase chain reaction (PCR) technique,SjAslp was amplified from the constructed recombinant plasmid pBCSK+/SjAslp,and inserted into cloning vector pUCm-T.Then,SjAslp was subcloned into an eukaryotic expression vector pcDNA3.1(+).After identifying it by PCR,restrictive enzymes digestion and DNA sequencing,the recombinant plasmid was transfected into HeLa cells using electroporation,and the expression of the recombinant protein was analyzed by immunocytochemical assay.Results:The specific gene fragment of 558 bp was successfully amplified.The DNA vaccine of SjAslp was successfully constructed.Immunocytochemical assay showed that SjAslp was expressed in the cytoplasm of HeLa cells.Conclusion:SjAslp gene can be expressed in eukaryotic system,which lays the foundation for development of the SjAslp DNA vaccine against schitosomiasis.

Abstract:<正>Objective:To evaluate the lumbar stability and the primary clinical results of unilateral facetectomy, transforaminal lumbar interbody fusion (TLIF) and unilateral pedicle screw fixation by X-Tube system.Methods: Five human lumbar cadaveric functional spine units(FSU) were obtained and graded facetectomy by 0,1/4,1/2,3/4 and 4/4 were performed respectively on the left articular process of them.The stability of these 5 models was evaluated at flexion,extension,lateral bending and axial rotation.After a serial of biomechanical researches,23 patients from June 2004 to March 2006 in our department underwent unilateral facetectomy,transforaminal lumbar interbody fusion (posterior lumbar interbody fusion) and unilateral pedicle screw instrumentation by X-Tube system.After general anaesthesia,with the guide of fluoroscopy and using X-Tube system,procedures of unilateral endoscopic facetectomy,spinal nerve root decompression,autologus spongy bone transplantation,one cage oblique insertion and unilateral pedicle screw instrumentation were performed.Results:There was no significant difference in flexion,extension,lateral bending and axial rotation of lumbar motion range after unilateral graded facetectomy.The stability of left/right axial direction was greatly affected when the range of graded facetectomy exceed 1/2.According to the Nakai criteria,for the 23 patients,the clinical result was excellent in 15 (65.2%), good in 6 (26.1%) and fair in 2 (8.7%).The fusion rate was 95.6% in excellent and good cases.Although partial absorption of bone grafts was observed in 1 case which might indicate a unsuccessful fusion,there was no loosing and replacement of instrument and no clinical symptoms occurred.Conclusion:The lumbar stability will be affected significantly when the range of graded facetectomy exceeds 1/2.Procedures of unilateral facetectomy, transforaminal lumbar interbody fusion and unilateral pedicle screw fixation is an optional strategy for microsurgical reconstruction,though the indications of the procedure should be carefully restricted.

Abstract:<正>Objective:To study the efficacy and safety of autologous transplantation of bone marrow mesenchymal stem cells on diabetic patients with lower limb ischemia.Methods:Fifty Type 2 diabetic patients with lower limb ischemia were enrolled and randomized to either transplanted group or control group.Patients in both group received the same conventional treatment.Meanwhile,20 ml bone marrow from each transplanted patient were collected,and the mesenchymal stem cells were separated by density gradient centrifugation and cultured in the medium with autologous serum.After three-weeks adherent culture in vitro,7.32×108-5.61×109 mesenchymal stem cells were harvested and transplanted by multiple intramuscular and hypodermic injections into the impaired lower limbs.Results:At the end of 12-week follow-up,5 patients were excluded from this study because of clinical worsening or failure of cell culture. Main ischemic symptoms,including rest pain and intermittent claudication,were improved significantly in transplanted patients.The ulcer healing rate of the transplanted group (15 of 18,83.33%) was significantly higher than that of the control group (9 of 20,45.00%,P=0.012).The mean of resting ankle-brachial index (ABI) in transplanted group significantly was increased from 0.61±0.09 to 0.74±0.11 (P<0.001).Magnetic resonance angiography (MRA) demonstrated that there were more patients whose score of new vessels exceeded or equaled to 2 in the transplant patients (11 of 15) than in control patients (2 of 14,P=0.001).Lower limb amputation rate was significantly lower in transplanted group than in the control group (P=0.040).No adverse effects was observed in transplanted group. Conclusion:These results indicate that the autologous transplantation of bone marrow mesenchymal stem cells relieves critical lower limb ischemia and promotes ulcers healing in Type 2 diabetic patients.

Abstract:<正>Objective:To describe the MR1 findings and discuss the pathogenesis formation mechanism of syringomyelia in the patients after spine surgical operation.Methods:Totally 13 patients were been enrolled in the study.Before operation,none of our patients presented with spinal cord syringomyelia.The mean follow-up duration was 6 years (range 2-10 months) by MRI.Four of the 13 patients after spinal cord tumor removed operation,3 patients after spinal trauma operation,2 cases had scoliosis before,2 cases were cervical spondylotic myelopathy and 2 cases had tethered cord syndrome before operation.MR features of pre- and post-operation on all patients were been studied in contrast with surgical results and clinical symptoms.The radiology diagnostic was made by 3 different radiologists respectively. The characteristics of length,width,signals,shape of cavity and spinal cord position as well as subarachnoid shape were focused on.Results:All the patients had no syringomyelia on MRI before operation.The spinal cord of 7 patients showed persist compression and 2 patients had tethered cord before operation.MRI features of syringomyelia after operation in our cases showed longitudinal cavity with syrinx fluid had T1 and T2 relation characteristics of cerebrospinal fluid (CSF).The mean length was 5.5 spinal segment,4 cord of 13 patients presented cord compressed from anterior materials,4 occurred postoperative adhesion to the back of lumbar spinal canal and spinal cord of 1 cases clung to the front wall of cervical spinal canal.Conclusion:The mechanism of syringomyelia in the patients after spinal surgical intervention may be the persisting compression or intention of the spinal cord in the period of pre- or post-operation.The edema,cyst,malacia of spinal cord are the most important lesions and risk factors resulted in the syringomyelia.

Abstract:<正>Papillary fibroelastoma,a rare primary benign tumor,can be found anywhere in the heart,but most commonly involves the cardiac valves.Most papillary fibroelastomas do not cause symptoms and are usually incidental found by routine echocardiograpby or at autopsy.However,with the advent of echocardiography,more and more patients are diagnosed in life.Early diagnosis of this condition becomes very important since it represents a surgically correctable cause for systemic emboli,stroke,myocardial infarction,and sudden cardiac death.The echocardiographic findings should be confirmed by histology because the clinical differential diagnoses includes myxoma,vegetation,thrombi, lipoma,and pseudopapillary fibroelastoma.We reported a case of papillary fibroelastoma of the aortic valve and presented a comprehensive review of the literature.
