Abstract:<正>Objective:To study the characteristics of latency of somatosensory evoked potential (SEP) and motor evoked potential (MEP) and the expression of nestin in subventricular zones (SVZ) after per- sistent focal cerebral ischemia in rats.Methods:The model of cerebral ischemia in rats was made by mid- dle cerebral artery occlusion (MCAO).All animals of ischemia were sacrificed after 12h,1d,3d,7d, and 14d to observe the changes of latency of SEP and MEP and to detect the expression of nestin,with an immunohistochemieal approach.Results:The latencies of P1 (positive wave 1),N1 (negative wave 1)and P2 (positive wave 2) in SEP were significantly prolonged after MCAO.The latencies of N1 and N2 waves in MEP were postponed gradually and no statistical difference of latency of N1 wave was found in rats at 7 d and 14d after MCAO.The expression of nestin increased at 12h,and showed a significant augmentation at 3d and peaked at 7d,then declined slightly at 14d after MCAO.Conclusion:The cerebral ischemia prolonged the latency of EP waves and the expression of nestin was up-regulated and reached the peak at 7 d,showing the ischemia induced the proliferation of nervous stem cells.The SEP and MEP may evaluate the proliferation in SVZ after brain isehemia.
Abstract:<正>Objective:To detect the expression of glial fibrillary acid protein (GFAP) and taurine trans- porter (TauT) in the retinal Müller cells in high glucose culture with taurine and to explore the influence of glucose on the taurine transporting,and the possible protective effects of taurine on Müller cells in early diabetic retinopathy.Methods.The Müller cells from the rat retina were cultured in high glucose,and GFAP and TauT expressions were detected in the cells treated with different doses of taurine by immuocy- tochemical fluorescein staining and Western blotting.Results:High glucose enhanced the expression of GFAP and decreased the expression of TauT in Müller cells.Taurine decreased the up-regulation of GFAP in the cells which was induced by high glucoses 0.1-10mmol/L taurine increased the expression of TauT in Müller cells.Conclusion:Taurine can inhibit the changes in Müller cell resulted from high glucose.
Abstract:<正>Objective:To evaluate the inhibitory effect of heparanase antisense oligodeoxynucleotide (AS-ODN) on the angiogenesis and metastasis of human mammary carcinoma cell xenografts in nude mice. Methods:The AS-ODN complementary to the start codon region of heparanase mRNA and its control, scrambled nonsense oligodeoxynucleotide (NS-ODN) were designed and synthesized.A subcutaneous growth model and an acute hematogenous metastasis model of human mammary carcinoma were estab- lished in nude mice and were treated with ODNs.The heparanase expression in tumor was evaluated by RT-PCR and Western blot.The microvessel density (MVD) was measured by immunohistochemistry for factorⅧ.The tumor volume was calculated and lung metastatic nodules were counted.Results:The hep- aranase expression,MVD,tumor volume and lung metastatic nodules in AS-ODN treated group were sig- nificantly decreased compared with that in NS-ODN treated group and that in PBS group (P<0.01).Con- clusion:Heparanase AS-ODN has significant inhibitory effect on the angiogenesis and metastasis of human mammary carcinoma cell xenografts in nude mice.
Abstract:<正>Objective:To investigate the susceptibility of strains separated from probiotic products for medical purpose to 14 antimicrobial agents.Methods:The single aerobic strains were isolated from these products respectively and disc agar diffusion assay was proceeded to determine the susceptibility.Results: Probiotics tested in the study mostly showed multiresistant to the agents.Lactobacillus acidophilus LAP, LAB,Lactobacillus bulgaricus LBJ and Streptococcus thermophilus STJ were resistant to vancomycin.Con- clusion..Drug resistance exists in most of commercial probiotics.The evaluation and monitoring of safety of probiotic products for medical purpose should be paid great attention.
Abstract:<正>Objective:To investigate the changes of intestinal mucosa tight junctions (TJs) claudin-1, -3,-4 proteins and mRNA changes in patients with irritable bowel syndrome (IBS) and to elucidate their possible roles in the changes of bowel evacuation habit and formation.Methods:Claudin-1,-3,-4 proteins and mRNA were evaluated in intestinal mucosa in control group,D-IBS (diarrhea IBS) group and C-IBS (constipation IBS) group with immunohistochemical assay and Realtime-PCR.Results:It was observed that claudin-1,-3,-4 proteins were localized in the membranes of epithelial cells along the entire length of the plasma membrane including the apical end of the epithelial cells.The claudins were concentrated at the site of TJs only.Claudin-1,-3,-4 mRNA and claudin-1 protein in small intestinal mucosa and colonal mu- cous in D-IBS group were significantly downregulated (P<0.05).Claudin-1,-3,-4 mRNA and proteins in small intestinal mucosa and colonal mucous in C-IBS group were significantly upregulated (P<0.05). There was no significant difference in the expression of claudin-3 protein in both small intestinal mucosa and colonal mucous between D-IBS group and control group(P>0.05).Similarly,no significantly differ- ent expression of claudin-4 protein in colonal mucous in D-IBS group was found compared with control group (P>0.05).Otherwise,the expression of claudin-4 protein in small intestinal mucosa decreased in D-IBS group (P<0.05).Conclusion:Claudin-1,-3,-4 may play a potential important role in the changes of bowel evacuation habit and formation in patients with IBS.It is not due to the localization changes of claudin proteins in T J,but may be caused by the quantitative changes of the expression of TJ proteins and mRNA.
Abstract:<正>Objective:To explore the serum levels of resistin and adiponectin in patients with overweight and obesity.Methods:Fifty-eight cases with normal weight and 24 patients with overweight and obesity have taken fasting blood samples for measurements of plasma glucose,plasma lipids,insulin,C-peptide, thyroid hormones,C-response protein,interleukin-6,TNF-α,leptin,adiponectin and resistin.Results: The concentrations of resistin in cases with overweight and obesity were significant higher than those in the normal weight cases (16.01±8.60 vs 11.63±9.05ng/ml,P=0.047).Pearson relation analysis showed that serum resistin concentrations were positively correlated with age (r=0.476,P=0.019),but negatively correlated with C-peptide (r=-0.45,P=0.024),and adiponectin concentrations were posi- tively correlated with HDL-c (r=0.463,P=0.023) and systolic blood pressure (r=0.409,P=0.047)in overweight and obesity cases.Conclusion:The concentrations of resistin in cases with over-weight and obesity are higher,and there is no correlation between resistin and blood glucose,blood lipids and insulin, while the serum adiponectin concentrations positively correlated with HDL-c and systolic blood pressure.
Abstract:<正>Objective:To examine and compare the synchronization of different brain regions during the Chinese and English Stroop tasks.Methods.Ten native Chinese speakers with a moderate command of English participated in this study,and event-related potentials were recorded while participants performed the Stroop task.Then wavelet-based estimation of instantaneous EEG coherence was applied to investigate the synchronization of different brain regions during Stroop task.Results:A greater negativity for the in- congruent situation than congruent situation appeared from 350ms to 600ms post-stimulus onset over frontal,central,and parietal regions in Chinese Stroop task,while the negativity was absent in English Stroop task.However,not only in Chinese Stroop task but also in English Stroop task was it found signif- icantly higher EEG coherences for the incongruent situation than congruent situation over frontal,pari- etal,and frontoparietal regions before 400ms post stimulus onset atβ(13-30 Hz) frequency band.Conclu- sion:This finding indicated that wavelet-based coherence is more exquisite tool to analyze brain electro- physiological signals associated with complex cognitive task than ERP component,and that functional syn- chronization indexed by EEG coherence is enhanced at the earlier stage while processing the conflicting in- formation for the incongruent stimulus.
Abstract:<正>Objective:To explore the prevalence rate of knee joint osteoarthritis and its pathogenetic fea- tures among the middle-aged and old aged people in the urban and rural area of Xi’an.Methods:From May to October 2005,1538 Han nationality above 40 years in Xi’an were investigated according to strati- fied and cluster random sampling.The same questionnaire was delivered to subjects who had been given normotopia and lateral position radiographic examination in both knees.Excluded from etiological arthri- tis,some subjects with clinical symptoms and a radiographic grade (beyond Kellgren & Lawrence gradeⅡ) were finally diagnosed as spontaneous knee joint osteoarthritis.All analyses were performed with SPSS 13.0 and t test,U test.Single factor analysis and multiple logistic regression analysis were used to analyze dates.Results:The total prevalence of spontaneous knee joint osteoarthritis was 12.1% with 2.5% and 2.1% for right and left knees,respectively.The prevalence of knee joint osteoarthritis in wom- en was 2.5 times of that in men (17.2% vs 6.8%,P=0.000).the osteophyte prevalence in women was 2.3 times of that in men (26.5% vs 11.4%,P=0.000).The prevalence of knee joint osteoarthritis and osteophyte increased with the growth of age.So did the symptomatic knee,except for the age group of 56- 60 years and 66-70 years.The prevalence of symptomatic knee joint for urban area was significantly higher than that for rural area (56.3% vs 45.3%,P=0.003).In women,the prevalence of knee joint os- teoarthritis increased significantly with the increasing of body mass index,which had not been observed in men.Female and obesity are the risk factors for osteoarthritis,but bean and its products and meat could prevent the osteoarthritis.Conclusion:Old age,female and obesity serve as risk factors for knee joint os- teoarthritis.
Abstract:<正>Objective:To explore the present situation and development of Medical Delivery System in China.Methods:The data in three Nation Health Service Investigations were compared and analyzed by applying literature meta-analysis,Delphi and cross-sectional investigation,etc.Results:The Medical Ser- vice resource develops fast,and public cost for medical treatment keeps increasing.Organization structure presents a vertical and flat trend.Inhabitant health level rises year by year,and it is hard to resolve the problem of potential medical demand transformed.Conclusion:Medical Delivery System in China has formed,but the development of Medical Service System still exists structural problems in the deeper lay- er.The development trend predicts that the supervision of the government is gradually reinforcing and community health service develops step by step.
Abstract:<正>Objective:To establish a scientific,objective and applicable index system for evaluating out- standing biomedical scientists for science foundation of Shanghai.Methods:According to the principal in- dices that have been used in the developed countries for evaluating their talented personnel and the reality of our country,an index system was set up to evaluate the outstanding biomedical scientists for Shanghai science foundation.The following parameters were used to simplify the indices.,correlation coefficient, multiple correlation coefficient,partial correlation coefficient,creditability,and discriminatory power. And analytic hierarchy process was used to determine the weights of each index.Results and Conclusions: The established index system is scientific and applicable;it is helpful for cultivating and evaluating out- standing biomedical scientists.
