Abstract:<正>Objective: To explore the possible mitochondrial DNA (mtDNA) polymorphism in Han Chinese. Methods: The complete mitochondrial genome of 26 unrelated healthy Han Chinese were extracted and sequenced. Results:The mtDNA nucleotide sites (2 706, 7 028, 8 860, 11 719, and 15 326) were found totally different from the Revised Cambridge Reference Sequence (rCRS). These single nucleotide polymorphisms (SNPs) were 2 706 A→G, 7 028 C→T, 8 860 A→G, 11 719 G→A, 15 326 A→G. Conclusion: These findings provide new insights into the characteristics of Han Chinese mitochondrial genetic diversity.
Abstract:<正>Objective: To assess the inhibitory effects of local injection of liposomal adriamycin (LADR) on the proliferation of lymph node metastases in rabbits bearing VX2 carcinoma in the mammary gland. Methods:Thirty female New Zealand white rabbits were divided into 3 groups, with 10 in each. VX2 tumor mass suspensions were injected into the breast tissues of rabbits. Treatment initiated once the axillary lymph node reached 5 mm in the maximum diameter. Group 1 received a sham treatment. Group 2 received a subcutaneous injection of LADR adjacent to tumor. Group 3 received an intravenous injection of free ADR (FADR) at the same dose and concentration to group 2. The breast tumors and axillary lymph nodes were resected after the treatment was repeated 3 times. The tumor and node sizes before and after treatment were measured. PCNA mRNA expressions in breast tumors and axillary nodes were determined using RT-PCR. Results: The mean growth ratios of lymph nodes after treatment were 3. 70, 1. 55, and 2. 89,respectively, in groups 1,2, and 3. The slowest node growth was observed in animals of group 2, with significant differences from group 1 (P<0. 001) and group 3 (P = 0. 002). The relative values of PCNA mRNA expression in lymph nodes were 0. 541, 0. 329,and 0. 450, respectively, in groups 1,2, and 3. Group 2 exhibited a significantly reduced PCNA mRNA expression in metastatic lymph node, as compared to group 1 (P<0. 001) and group 3 (P = 0. 004). Intravenous FADR injection effectively lowered the mRNA expressions of PCNA in breast tumors, which were not apparently altered after local LADR injection. Conclusion: Local injection of LADR holds a strong inhibitory effect on the proliferation of metastatic tumor cells in lymph nodes and appears to be an effective method for the treatment of lymphatic metastases of breast cancer.
Abstract:<正>Objective:To study curcumin’s growth inhibitory effects and morphology changes on sarcoma graft’s of S180 mouse, with further inquiry into the possible mechanisms. Methods: Thirty S180 mouse were randomly assigned into 3 groups : saline group (blank control group), Cytoxan group (positive control group) and curcumin group. The tumor inhibitory rates, the index of thymus and spleen.the growth of tumor and the change of pathology-morph,the index of apoptosis cells and morphology changes of apop-tosis cells in the different groups were observed. Results: (1) Tumor’s inhibitory rate in curcumin group and cytoxan group was 68.32% and 70. 43%, respectively. Compared to blank control group, the 2 groups had significant elevated tumor inhibitory rate (P<0. 01). (2) Thymus index of curcumin group did not have significant decrease compared to blank control group (P>0. 05). (3) Under electroscope,curcumin group and positive control group had significant decrease in terms of growth of tumor, degree of infiltration of tumor, the number of nucleus fission, and blood vessels number compared to saline group (P<0. 05). However, the degree of cell necrosis, the number of splenic segments and macrophage are increased significantly compared to negative group. (4) Accumulative score of apoptosis cell in curcumin group was significantly higher than other two groups(P<0. 05). Conclusion: Internal organ study and cell morphology observation show curcumin can effectively inhibit the growth and cause the death of sarcoma graft of S180 mouse without interference with thymus.
Abstract:<正>Objective: To investigate the nature of the restoration of cell polarity and bile excretion function in Sandwich-cultured hepatocytes. Methods: Freshly isolated hepatocytes from male Sprague-Dawley rats were cultured in a double layer collagen gel Sandwich configuration. Morphological changes were observed under a inverted microscope. The domain specific membrane associated protein DPPⅣwas tested by immunofluorescence. and the bile excretion function was determined by using fluorescein diacetate. Hepatocytes cultured on a single layer of collagen gel were taken as control. Results: Adult rat hepatocytes cultured in a double layer collagen gel sandwich configuration regained its morphological and functional polarity and maintained polygonal morphology for at least 4 weeks. Immunofluorescence studies using antibodies against DPPⅣshowed polarity restoration as early as 48 h. After cultured in the double layer collagen gel Sandwich configuration for 96 h the hepatocytes began to excrete bile; while hepatocytes cultured on a single layer collagen gel had no bile excretion. Conclusion: Hepatocytes cultured in a double layer collagen gel Sandwich configuration are able to regain their morphological and functional polarity given certain conditions. Hepaotcyte culture is a useful tool for the study of polarity restoration.
Abstract:<正>Objective:To study the effect of iodine-125 interstitial brachytherapy on liver cancer. Methods: Animal model of human liver cancer was established by injecting SMMC-7721 cells cultivated in vitro subcutaneously into the flank of BALB/c nude mice. Nude mice with tumor of 5 mm in diameter were randomly divided into 2 groups (n = 10). One iodine-125 seed of apparent activity 0. 8 mCi was implanted into the center of tumor in treatment group, whereas an inactive seed was implanted in control group. The other 20 nude mice with tumor reaching 10 mm in diameter were also treated as above. The size of tumor was determined weekly after implantation, and pathological examination and blood routine were taken on the 28th day. Results: Tumor growth was obviously inhibited in treatment group of tumor of 5 mm in diameter, and there was statistically significant difference in tumor volume between treatment and control groups (P<0. 01). Around iodine-125 seed, apparent necrosis of tumor was shown in treatment group, accompanied by karyopyknosis and reduced plasma in residual tumor cells microscopically. Tumor growth was not inhibited in either treatment or control group of tumor of 10 mm in diameter. There was no obvious adverse effect except for decreased white blood cells in treatment groups. Conclusion: There is certain effect of iodine-125 interstitial brachytherapy on liver cancer, which is associated with the size of tumor.
Abstract:<正>Objective: To explore the expression of nuclear factor-kappa B (NF-κB) in Schwann cells (SCs) and its effect on motor neuron apoptosis in spinal cord following sciatic nerves injury in adult rats. Methods: Thirty-six adult Sprague-Dawley (SD) rats were divided randomly into normal control group (n=6), and sciatic nerves crushing group (n=30). and the later was further equally randomized into 5 subgroups: 1. 3. 7. 11. and 21 d post-injury groups. The expression of NF-κB of normal and injured nerves were examined by immunohistochemistry staining, and the apoptosis of motor neurons in spinal cord of lumbar 4 to lumbar 6 (L4-L6) was investigated by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling (TUNED assay. Both were quantitated by image analysis. Results: In crushing group, except 21 d post-injury group, the expression of NF-κB was markedly higher than that in the normal control group (P<0. 05,P<0. 01). At 1 d after sciatic nerves crushing, the expression of NF-κB was obviously up-regulated, reached peak at 3 d. and recovered at 21 d. The same trend was observed in the time-course on motor neuron apoptosis after sciatic nerves injury. Correlation analyses revealed that motor neuron apoptosis was significantly and positively correlated with the expression of NF-κB following sciatic nerves injury (r=0. 976 0,P<0. 01). Conclusion: After injury of sciatic nerves, the presence and up-regulation of NF-κB in SCs may be involved in motor neuron apoptosis in L4-L6 spinal cord.
Abstract:<正>Objective:To study the protective mechanisms of neurotrophin-3 (NT-3) on the spinal cord injury. Methods: Totally 105 SD rats were randomly divided into 3 groups: control group, experimental group and sham operation group. Rats from the former 2 groups were inflicted to animal model of acute spinal cord injury according to Allen’s (WD) by situating a thin plastic tube in the subarachnoid space below the injury level for perfusion. Rats in experimental group received 20μl NT-3 (200 ng) from the tube at 0, 4, 8, 12, 24 h and 3, 7 d after injury, and those in control group got an equal volume of normal saline at the same time. The animals in sham operation group only received opening vertebral plate and tube was put in subarachnoid space. The rats were sacrificed at 4, 8, 12, 24 h and 3, 7, 14 d post injury (n = 5). The expression levels of Bcl-2 and Bax proteins in spinal cord of rats were detected by immunohis-tochemistry assay. Results: The level of Bax protein in control group significantly increased as compared with those in sham operation group, and the peak reached at 8 h after spinal cord injury. The Bcl-2 proteins were always weakly positive. The Bax proteins in NT-3 group significantly decreased but the Bcl-2 proteins obviously increased as compared with those in control group. Conclusion: NT-3 can protect spinal cord from injury in vivo. One of the mechanisms is that NT-3 can inhibit abnormal expression of Bax protein, and increase the expression of Bcl-2 protein, then inhibit apoptosis after spinal cord injury.
Abstract:<正>Objective: To study the expression of △Np73, an isoform of the p53 homologue p73, in the different stages of human non-small-cell lung cancer (NSCLC) and the association of△Np73 expression with in patient survival. Methods: Semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) was used to study the expression of△Np73 mRNA in 51 resected NSCLC tissues. Its relation to clinicopathological factors and survival outcome were analyzed. Results: The positive rate and expression level of△Np73 mRNA in the cancer tissues were significantly higher than that in the matched non-cancer lung tissues. The incidence of positive expression of△Np73 was 50. 0% , 52. 6% , and 87. 5% in patients with stageⅠ,Ⅱ, andⅢ, respectively. Positive expression of△Np73 was associated with pathological TNM stage (P = 0. 046), while not with age, gender, histological type and differentiation status. Survival rate of patients with high△Np73 mRNA was significantly poorer than those with low△Np73 mRNA levels (P<0. 001). Multivariate analysis revealed that△Np73 mRNA levels were a significant prognostic factor, independent of the other conventional prognostic factors. Conclusion: NSCLC has over-expression of△Np73 mRNA, which is closely related to TNM stages and prognosis of patients with NSCLC. These results suggest that measurement of△Np73 mRNA levels in tumor tissues might be useful as a promising predictor for the prognosis of patients with NSCLC.
Abstract:<正>Objective: To investigate the effects of static magnetic field(SMF) on the viability, adhesion molecule expression of human umbilical vessel endothelial cell. Methods: Magnetic flux intensity was 0. 1 mT, 1 mT, 10 mT. Cell viability and proliferation were measured with 3H-TdR and MTT methods; and apoptosis of human umbilical vein endothelial cell (HUVEC) was studied by flow cytometry and transmission electric microscopy. ELISA was used to measure the expression of ICAM-1 and VCAM-1 on endothelium. Results: 0. 1 mT SMF had no effects on the growth of HUVEC. however,SMF of 1 mT, 10 mT attenuated growth of HUVEC. 10 mT static magnetic field could induce apoptosis and necrosis of HUVEC. 10 mT SMF enhanced the expression of ICAM-1 and VCAM-1 on endothelium. Conclusion: The effect of SMF depends on the intensity of SMF. 10 mT SMF has adverse effects on human umbilical vessel endothelial cell.
Abstract:<正>Objective: To evaluate the changes of serum matrix metalloproteinase-9 (mmp-9) in patients of incipient diabetic nephropathy with or without macrovascular disease and to analyze the factors associated with homocysteine(hcy), interleukin-6(IL-6), tumor necrosis factor-alpha (TNF-α), highly sensitive C-reactive protein (hsCRP), HbA1c and lipid profile in those patients in order to know whether this marker or other factors are more important to induce diabetic macrovascular disease. Methods: Type 2 diabetes mellitus(T2DM) subjects with incipient diabetic nephropathy with or without macrovascular disease were selected for participation and divided into 2 groups. The patients in group 1(n= 38) used insulin, and patients in group 2 (n = 34) were treated with an oral antidiabetic drug. Then serum mmp-9, hcy, IL-6 and TNF-αin these patients were measured, and compared to the healthy subjects as control (n= 16). The results were analyzed by SPSS13. Results: Serum mmp-9 and hcy of the patients having incipient diabetic nephropathy with macrovascular disease were higher than that of patients without macrovascular disease (P<0. 01). For insulin-injected patients, whether they accompanied with macrovascular diseases or not, the serum levels of mmp-9, hcy, IL-6 and TNF-αwere all lower, but no significant statistics compared with non-insulin used patients or the healthy subjects. The serum level of mmp-9 was more correlated with the serum hcy in antidiabetic drug used patients. (P<0. 000) Conclusion: The serum level of mmp-9 plays an important role of pathogenesis in the macrovascular disease in the incipient diabetic patients, and the serum level of hcy also can reflect the severely degree of macrovascular disease in these patients, insulin can reduce these markers.
Abstract:<正>Objective:To introduce a key-vertebral-screws technique(KVST) in the surgical treatment of scoliosis extending to main thoracic levels, and to find the role of fulcrum bending in predicting the result of surgical treatment for scoliosis by this technique. Methods: Seventeen consecutive patients with scoliosis extending to main thoracic spine,who underwent pure posterior fusion without anterior or posterior release by KVST between January 2004 and July 2005 were evaluated for fulcrum bending flexibility, surgical correction rate, fulcrum bending correction index (FBCI) in main thoracic curves. Universal Spine System (USS) instrumentation was used in 15 cases,Monarch in another 2 cases. The severity of the curves was measured by Cobb’s method using Rad Work 6. 0 software. Preoperative standing AP radiographs, preoperative fulcrum bending anterioposterior (AP) radiographs, postoperative standing AP radiographs, and most recent follow-up standing AP radiographs for spine were measured and recorded. All the data were analyzed with two-sample paired t-test by Origin 7. 0 software. Results: Infection and neurological complications were not noted. No major complications were found. Just one case had some axial back pain, which got a full recovery from physiotherapy for 2 weeks. In the X-ray, there was an average correction of 71. 5% of the fused main thoracic curves, which had no significant lose of correction in final follow-up. For the whole fused main thoracic curves, the fulcrum bending flexibility were lower to operation correction rate (P = 0. 013). The average FBCI was 123%. From the data, the more rigid curves (especially fulcrum bending flexibility <50%), the more correction rate operation could get, compared with fulcrum bending flexibility. Conclusion: (1) KVST is a good method in the surgical treatment of thoracic scoliosis, which can get satisfying result with lower medical cost. (2) Fulcrum bending flexibility is lower than operative correction rate by KVST in main thoracic curves (P<0. 05). In the more rigid curves assessed by fulcrum-bending radiograph, the operative corrective could be gained, especially in the curves which FBCI is lower than 50%.
Abstract:<正>Objective:To evaluate the electrophysiological characteristics and radiofrequency catheter ablation of atrial tachycardia (AT) in patients with repaired atrial septal defects(ASD). Methods: In 76 consecutive patients with AT who underwent the electrophysiological study and radiofrequency catheter ablation (RFCA). 4 patients (one male and three female aged 35. 5±11. 5 years) had AT-related myocardial scar or incision. Earliest activation combined with entrainment mapping was adopted to determine a critical isthmus. Results: Re-entry related to the lateral atriotomy scar was inducible in 3 of 4 patients. With en-trainment mapping, the PPI-TCL difference was <30 ms when pacing at the inferior margins of the right lateral atriotomy scar. Among them, 2 patients had successful linear ablation between scar area to inferior vena cava, and 1 patient between scar areas to tricuspid annulus. Re-entry involving an ASD patch was demonstrated in 1 of 4 patients. PPI-TCL differences <30 ms were found when entraining tachycardia at sites near the septal patch. But linear ablation failed in terminating AT. There was no complication during procedure. No recurrence of incision-related AT was found during follow-up except for the failed patient. Conclusion: Under conventional electrophysiological mapping, adopting linear ablation from scar area to anatomic barrier, successful ablation also can be obtained in patients with IRAT related to myocardial scar or incision.
Abstract:<正>Objective: To investigate the gene polymorphism in a pedigree of congenital monilethrix. Methods: Genomic DNA of affected members, the normal members of the pedigree and 50 unrelated normal members who came from different regions were extracted with a whole blood genomic DNA extraction kit and used as a template for the polymerase chain reaction (PCR)-mediated amplification of hHB1 and hHB6 genes. Results: In the pedigree, DNA analysis of patients and normal persons revealed C(447th) in exonl of hHB1 gene and the 52th codon was CCA encoding arginine. But it was a heteropeak of G or C in 50 unrelated normal members, which encodes glycine or arginine. It showed that this change was a single nucleotide polymorphisms (SNP). Conclusion:A genetic heterogeneity of monilethrix exists in Chinese population. SNP which can result in the change of amino acid sequence is found in a pedigree of congenital monilethrix, and a genetic heterogeneity of monilethrix existed in Chinese population.

The clinical manifestation of CMCJ is nonspecific, which includes swelling, pain, mobile teeth and bleeding, etc. To develop the diagnosis and treatment of CMCJ. this article presents 2 cases of CMCJ treated in our hospital. Their clinical data, treatment and prognosis were analyzed retrospectively.
