    • Profiles of hippocampal neuron activity during auditory discrimination cognition in guinea pigs

      Abstract:<正>Objective: To clarify the firing characteristics of the hippocampal pyramidal cells and in-terneurons in the auditory discrimination cognition. Methods: Thirteen guinea pigs were studied by the paired (active cognition group. n=10) or unpaired (passive cognition group. n = 3) training with 1 kHz (CS+)and 500 Hz tones (CS- ) and the air puff (US) applied 250 ms after the CS+ onset. Results: In active group. 32 pyramidal cells showed exciting response to the CS+ tone. 16 cells inhibited response and 4 cells revealed no response to the high frequency tone and 18 interneurons almost unchanged. In passive group, the pyramidal cells responded to the tone casually and 10 out of the 13 interneurons remained invariably. Conclusion: The result suggests that the pyramidal cells play a major role in coding auditory information by the networks, and the interneuons may modulate it via forward and feedback.

    • Differential expression of βig-h3 in human hepatoma cell lines

      Abstract:<正>Objective:To observe the differential expression of TGF-β-induced gene human clone 3 (βig-h3) in human hepatoma cell lines. Methods: Human hepatoma cells HHCC. 7721. T7721 constructed by stably transfecting HAbl8G/CD147 cDNA into 7721 and normal human liver cell QZG were cultured as previously. RT-PCR and western blot were used to investigate the differential expression ofβig-h3 in human hepatoma cell lines and normal liver cell. Results:The results of RT-PCR suggested that the expression ofβig-h3 mRNA in human hepatoma cells was higher than that in normal human liver cell QZG(P< 0. 01), and its expression level in human hepatoma cells in turn was HHCC>T7721>7721. Moreover, the similar results ofβig-h.3 protein expression were testified by western blot. Conclusion: This study demonstrates that the expression ofβig-h3 in hepatoma cell lines is higher than that in normal liver cell QZG, which provides a sound basis for exploring the function ofβig-h3 in processes of adhesion and metastasis of human hepatoma cells.

    • Optimization of the construction of recombinant plasmids PPARγ-pSUPER-EGFP for RNA interference

      Abstract:<正>Objective: To construct and identify the recombinant plasmids PPARγ-pSUPER-EGFP for RNA interference. Methods: The pSUPER-EGFP vectors were used to transcribe functional short interfering RNA (siRNA). Four pairs of 64 nt PPARγsiRNA encoding sequences were inserted into the downstream of the H1 promoter. The recombinant plasmids were confirmed by double digestion with the enzymes and sequencing. Western blotting was used to examine the silencing effect of PPARγgene in RAW264. 7 cells. Following procedures were used to optimize the experiments: the oligonucleotides were incubated 5 min at 95 C and cooled automatically in boiled water bath to anneal, and then phosphorylated oligonucleotides, pSUPER-EGFP plasmids was digested with Bgl II and Hind III . and the product was ligated into digested pSUPER-EGFP plasmids, and transforming the ligation products followed by screening and identifying positive clones. Results:Four kinds of positive clones producing 285 bp fragments were selected. Sequencing further proved their correctness. Four recombinant plasmids containing corresponding PPARγgene-specific target sequences induced the silencing of its target gene more or less. Conclusion: The optimizing method in constructing these recombinant plasmids serves other plasmid-based RNA interference research. The final plasmids PPARγ-pSUPER-EGFP established the basis for research on the function of PPARγgene.

      Abstract:<正>Objective:To investigate the expression of CXCR, on HL-60 cell line and the proliferation, apoptosis of HL-60 cell line cocultured with bone marrow stromal cells, so as to assess the possibility of 12G5. an anti-CXCR4 monoclonal antibody, in eradicating the minimal residual disease. Methods:The activity of SDF-1 was inhibited by 10μg/ml 12G5. After treatment with 12G5. the status of adhesion was observed, and the adhesion rates, apoptosis and cell cycles were detected after 24 h of treatment. Cell growth rates were measured by trypan blue exclusion. Cell growth curve was plotted, and the expression of PCNA and apoptosis related protein including PCNA, Bcl-2 and Fas were detected with immunohis-tochemical technique. Results :(1) There was middling degree expression of CXCR4 on HL-60 membrane. From 0 h to 6 h, as the time of 12G5 incubation along, the expression of CXCR4 decreased gradually. (2) After treatment for 24 h, the adhesion rates in the experiment group and the control were (39. 4±7. 9)% and (51. 4±5. 9)%, respectively. (3)After treatment for 24 h, the percentage of HL-60 cells in G0/G1 phase were (55. 21±4. 9)%, and that in S phase and G2/M phase were (30. 40±4. 1)% and (14. 39±5.2)%, respectively, with the corresponding proportions being (44. 67±2. 2) % , (45. 30±3. 7)% . and (10. 03±2. 6)% in the control. (4) The percentage of apoptotic HL-60 cells was (8. 95±1. 7)% in the experiment group, compared to (3. 97±2. 4)% in the control. (5)The survival rates of HL-60 cells decreased markedly at 48 h to 96 h, and the proliferation slowed down at this time duration. (6)The expression of PCNA and Bcl-2 down-regulated significantly, but the Fas protein expression was up-regulated. Conclusion :12G5 could inhibit the capability of adhesion and proliferation of HL-60 cells and it can induce more cells to enter G0/G1 phase and promote apoptosis. It may be helpful by inhibiting the bioactivity of SDF-1 with 12G5 in the therapy of marrow residual disease.

    • Inhibitory effect of adenoviral vector-mediated AT2R gene transfection on neointimal hyperplasia

      Abstract:<正>Objective: To investigate the effect of adenoviral vector-mediated AT2R gene transfection on neointimal hyperplasia after rat carotid artery balloon injury. Methods :AT2R gene was transferred into rat carotid arteries by recombinant adenovirus pAd-AT2R after the establishment of rat carotid balloon injury restenosis model. The arteries were harvested on the 14th day after gene transfer. The efficiency of trans-gene delivery was measured by the expression of adenovirus-encoding green fluorescent protein (GFP) under fluorescent microscope. The expression of AT2R and PCNA (proliferating cell nuclear antigen) was e-valuated by RT-PCR, immunocytochemistry, immunofluorescence staining, confocal microscopy, respectively. The ratio of intimal to medial area (I/M) was quantified with images and determined by an image analysis system. Results: GFP-positive area in adventitia, media and the forming neointima was about 40%. Adenoviral delivery of rat AT2R gene up-regulated AT2R expression in balloon-injured rat carotid and reduced PCNA expression and I/M significantly in neointima(P<0. 01). Double immunofluorescence labeling of AT2R and PCNA also showed that AT2R gene transfer inhibited VSMCs proliferation in neointima. Conclusion: AT2R gene transfer may be a novel promising therapy to limit neointimal hyperplasia.

    • Effect of neurotrophin-3 on SOD and MDA in rats after acute spinal cord injury

      Abstract:<正>Objective:To investigate the effect of neurotrophin-3 on the expressions of SOD and MDA in the injured spinal cord of rats. Methods: Totally 105 SD rats were randomly divided into 3 groups (n = 35): sham group, control group and experimental group. Animal model of acute spinal cord was inflicted with Allen’s method by a thin plastic tube situated in subarachnoid space below the injury level for perfusion. Rats in experimental group received 20μl NT-3 (200 ng) from the tube at 0, 4. 8. 12. 24 h and 3. 7 d after injury, and those in control group got the equal volume of normal saline at the same time points. The animals in sham group only received opening vertebral plate and putting tube in subarachnoid space. The rats were sacrificed at 4, 8. 12. 24 h, and 3. 7, 14 d postinjury (n = 5). And the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in blood were observed with colorimetric method. Results: The serum level of SOD reduced obviously and the level of MDA raised obviously in rats after the injury, and the activity of SOD reached the lowest on day 3 and the concentration of MDA reached peak at the 7 d. In the experimental group, the SOD level was obviously higher (P<0. 01). and MDA level was lower than the control (P<0. 01). Conclusion:NT-3 can mitigate secondary injury of spinal cord in vivo. One of mechanisms is that inhibits abnormal expression of MDA and elevates the activity of SOD. thus the injury of free radical and lipid peroxidation is attenuated.

    • Expression and its clinical significance of HLA-G in HCMV-infected placenta] villi at early pregnant stage

      Abstract:<正>Objective:To study the expression and its clinical significance of HLA-G in HCMV intrauterine infected placental villi at early pregnant stage. Methods:PCR (polymerase chain reaction) was used to screen the peripheral blood for HCMV-DNA in 462 women who had willingly undergone induced abortion. Then immunohistochemistry was also used to detect expressions of mouse anti-HCMV early antigen (HCMV-EA) and mouse anti-HLA-G in HCMV-DNA positive cases’ placental villi. The difference of HLA-G expressions between the intrauterine infection group (HCMV-EA positives), the intrauterine infection-free group(HCMV-EA negatives) and the normal control group (SO cases of healthy early placental villi) was compared. Results: Of the 78 cases, which were detected HCMV-DNA positive, 11 (14. 10%) were HCMV-EA positive. Compared with the other two groups, HLA-G expressions in the intrauterine infection group were both obviously decreased (both P<0. 001). HLA-G expression positions in all three groups were mainly located in the cytotrophoblast. Conclusion:Intrauterine HCMV infection at early pregnant stage is closely related to HLA-G expression at the maternal-fetal interface. The virogenetic products may affect the expression of HLA-G at the maternal-fetal interface and that of its immunological function, thus leading to different clinical outcomes.

      Abstract:<正>Objective:To introduce a new better contrast-enhanced MR angiographic method, named 3D time-resolved imaging of contrast kinetics (3D-TRICKS). Methods: TRICKS is a high temporal resolution (2-6 s) MR angiographic technique using a short TR(4 ms) and TE(1. 5 ms), partial echo sampling, in which central part of k-space is updated more frequently than the peripheral part. TRICKS pre-contrast mask 3D images are firstly scanned, and then the bolus injecting of Gd-DTPA. 15-20 sequential 3D images are acquired. The reconstructed 3D images, subtraction of contrast 3D images with mask images, are conceptually similar to a catheter-based intra-arterial digital subtraction angiographic series (DSA). Thirty patients underwent contrast-enhanced MR angiography using 3D-TRICKS. Results: Totally 12 vertebral arteries were well displayed on TRICKS, in which 7 were normal, 1 demonstrated bilateral vertebral artery stenosis, 4 had unilateral vertebral artery stenosis and 1 was accompanied with the same lateral carotid artery bifurcation stenosis. Four cases of bilateral renal arteries were normal, 1 transplanted kidney artery showed as normal and 1 transplanted kidney artery showed stenosis. 2 cerebral arteries were normal, 1 had sagittal sinus thrombosis and 1 displayed intracranial arteriovenous malformation. 3 pulmonary arteries were normal, 1 showed pulmonary artery thrombosis and 1 revealed pulmonary sequestration’s abnormal feeding artery and draining vein. One left lower limb fibrolipoma showed feeding artery. One displayed radial-ulnar artery artificial fistula stenosis. One revealed left antebrachium heman-gioma. Conclusion: TRICKS can clearly delineate most body vascular system and reveal most vascular abnormality. It possesses convenience and high successful rate, which make it the first choice of displaying most vascular abnormality.

    • The value of 18-FDG-PET for diagnosing and evaluating lymph node metastasis in primary breast cancer

      Abstract:<正>Objective: To analyze the result of 18F-2-deoxy-2-fluoro-D-glucose-positron emission tomography (FDG-PET) in suspicious primary breast cancer patients and to evaluate its value for the surgery therapy. Methods: Total 36 patients suspected of breast neoplasm were enrolled into the research. The result was compared with the pathology result. The rate of missed diagnosis, the rate of mi.sdiagnosis. the sensitivity and specificity were calculated and analyzed. Results:Compared with the pathology results, the misdiagnosis rate. the rate of missed diagnosis. the sensitivity and specificity of FDG-PET for breast cancer were 0% , 36. 36%. 63. 63% and 100%, respectively. To those who had a neoplasm no more than 2 cm in diameter, the rate of missed diagnosis was as high as 41. 67%. To 33 breast cancer patients, the misdiagnosis rate, the rate of missed diagnosis, the sensitivity and specificity for lymph node metastasis were 18. 75%, 41. 18%, 58. 82% and 81. 25%, respectively. Conclusion:FDG-PET has a perfect specificity and a considerable sensitivity to the primary breast neoplasm and similar to the lymph node metastasis diagnosis. It is an ideal choice for those patients with suspected breast cancer but reluctantly to receive a vulnerarious examination.

    • A comparison of GlideScope videolaryngoscope with Macintosh laryngoscope for laryngeal views

      Abstract:<正>Objective:To describe the use of the GlideScope in comparison with direct laryngoscopy for elective surgical patients requiring tracheal intubation. Methods:Two hundred patients. ASA I - I scheduled for elective surgery under general anesthesia requiring orotracheal intubation were selected. Information was collected identifying the patient demographics and airway assessment features (Mallampati oropharyngeal scale, thyromenta distance and mouth opening). In a random crossover design, after induction of anesthesia and neuromuscular block, the laryngoscopes were inserted in turn, and the views of the glottis at laryngoscopy (Cormack and Lehane scores) were compared. The tracchea was intubated using either the standard Macintosh laryngoscope or GlideScope after the second grading at laryngoscopy was done. Complications associated with intubating were recorded. Results: There were 200 patients including 107 males and 93 females, with mean age being 52±13 years, height 164. 8±11. 3 cm, weight 64. 0±11. 5 kg, thyromental distance 6. 9±1. 1 cm, and mouth opening 5. 7±0. 5 cm. There was a significant association between the preoperative view of the oropharynx (Mallampati score) and the view of the glottis at laryngoscopy for both the direct Macintosh laryngoscope (P<0. 001) and the GlideScope (P<0. 001). A-mong 200 patients, 106 patients had the same C&L grade, 91 of remaining patients showed improvement in the C&L grade (P<0. 001) obtained with GlideScope compared with the direct Macintosh laryngoscope. 3 of remaining patients showed better view of the glottis(C&L grade) with the direct Macintosh laryngoscope (grade 1) than with GlideScope (grade 2). There were no cases of failure to be intubated. There were no cases of dental or mucosal injury in all patients. Conclusion: GlideScope videolaryngoscope yielded comparable or superior laryngeal view compared with Macintosh laryngoscope. The new type of laryngoscope may have potential advantages for managing the difficult airway.

    • Anteroposterior cricoid split interposition costal cartilage grafts for laryngotracheal stenosis in children

      Abstract:<正>Objective: To investigate the effects of anteroposterior cricoid spliting interposition costal grafting for moderate and severe laryngotracheal stenosis in children. Methods: From 1995 to 2005, 87 children (aged 1. 2 to 14 years) with moderate and severe glottic and subglottic stenosis were retrospectively studied. They were operated with cricoid spliting laryngotracheal reconstruction in our hospital. All of 87 patients were tracheostomy-dependent before surgery. Results: Eighty-five patients (95%) were successfully decannnulated and got an effective phonation. The follow-up time was more than 5 years. The effect of operation was satisfactory and the growth and development of children was normal. Conclusion: Anteroposterior cricoid split interposition costal cartilage graft is a safe and effective treatment method for moderate and severe glottic and subglottic stenosis in children.

      Abstract:<正>Objective: To analyze the inductive agents, post-surgical managements and precautions of the serious complications caused by transcatheter arterial chemoembolization (TACE) for the treatment of hepatocellular carcinoma. Methods: Clinical data of 2 375 cases of TACE for 832 patients with middle-stage or advanced hepatocellular carcinoma from January 1994 to December 2002 were analyzed. Results: There were 7 cases of serious complications, one of which was liver carcinoma rupture and another was acute renal failure. Totally there were 4 cases of paraplegia and one case of conduit knotted. Conclusion: Deadly complications may happen after TACE and it needs close surveillance and management in time.
