Abstract:<正>Objective: To study whether the anti-AS effect of ginkgo biloba extract (GbE) was related with inhibitory effects on the expression of IL-1β, TNF-αand up-regulation of IL-10 and IL-10R in the heart of atherosclerotic (AS) rats. Methods : The experimental model of AS rats were established by in-traperitioneal injection of vitamin D3 with high fat and cholesterol diet. All rats were divided into 3 groups: control,AS and GbE. GbE (100 mg/kg) was administered to rats by ig. After 8 weeks, the expression of IL-1β, TNF-α, IL-10, and IL-10R in the heart of AS rats were detected by enzyme-linked immunosorbant assay, reverse transcriptase polymerasechain reaction and Western blotting. Results:The protein and mR-NA expressions of IL-1β,TNF-α,IL-10 and IL-10R were markedly higher in AS group than those in control group (P<0. 01). The protein and mRNA expressions of IL-1βand TNF-αwere markedly lower in GbE group than those in AS group; while the protein and mRNA expressions of IL-10 and IL-10R were markedly higher in GbE group than those in AS group (P<0. 01). Conclusion: GbE has significant inhibitory effects on proinflammatory cytokine IL-1β, TNF-α. The up-regulation of anti-inflammatory cy-tokine IL-10,IL-10R that may be partially responsible for its anti-AS effects.
Abstract:<正>Objective:To evaluate the antiviral activity of the alcohol extract of Styela plicata on DHBV (duck hepatitis B virus) in vivo. Methods: Guangzhou-Sheldrake ducklings congenitally infected with DHBV were assigned to receive the alcohol extract of Styela plicata or lamivudine for 30 consecutive days. The DHBV DNA of sera was detected by RT-PCR. and the histological analysis of duckling liver was evaluated. Results:Thirty days after therapy,histological analysis of duckling liver showed that the ducklings receiving the alcohol extract of Styela plicata or lamivudine exhibited catabatic status in the degree of liver cell degeneration and inflammation compared with the ducklings receiving normal diet. DHBV DNA of sera from alcohol extract of Styela plicata-treated ducklings and lamivudine-treated ducklings all produced significantly lower levels compared with ducklings receiving normal diet (P<0. 01 ). Although these treatment groups all exhibited a rebound phenomenon 10 d after withdrawal of medication, they still exhibited a significant lower level of serum DHBV DNA compared with the control group responded to normal diet (P<0. 05, P<0. 01). Conclusion:Styela plicata may be an effective antiviral medicine in treating chronic hepatitis B. The data of this experiment will be valuable in studying the therapeutic role and the potential therapeutic mechanism of Styela plicata.
Abstract:<正>Objective: To evaluate the differentiation of human umbilical cord blood cells into hepatocyte-like cells. Methods: Mononuclear cells (MNCs) derived from human umbilical cord blood were isolated using Ficoll. The experiment was derived into 3 categories:(1) MNCs co-cultured with 50 mg minced liver tissue separated by a trans-well membrane and then collected at 0 h,24 h,48 h and 72 h; (2) MNCs cultured along supplemented with 100 ml/L FBS. 100μ/ml penicillin, 100μg/ml streptomycin. 4. 7μg/ml linoleic acid, 1×ITS, 10-4 mol/L L-Ascorbic acid 2-P and a combination of FGF4 (100 ng/ml) and HGF (20 ng/mL). Cells were then collected at 0 d and 16 d to examine the expression profile of hepatocyte correlating markers;(3) 0. 2-0. 3 ml of MNCs with a cell density of 2×107/ml were transplanted into prepared recipient mice [n = 12, injected with 0. 4 ml/kg (20%) CCl4 and 150 ng/kg 5-fluorouracil (5-Fu) prior the transplant 24 h and 48 h, respectively] via injection through tail vein. Mice were sacrificed 4 weeks after transplantation. The hepatocyte correlating mRNAs and proteins were determined by RT-PCR, immunohistochemical analysis and immunoflurence technique. Results: (1) After 72 h. a number of glycogen positive stained cells were observed with MNCs co-cultured with damaged mouse liver tissues. The expression of hepatocyte markers, human albumin (ALB),α-fetal protein (AFP) and human GATA4 mRNA and proteins were detected by RT-PCR and immunohistochemistry as well. For the confirmation, the DNA sequencing of PCR products was performed. In control groups, MNCs co-cultured with normal mouse hepatocytes or MNCs cultured alone, all markers remained negative. (2) In growth factor supplemented culture system, MNCs developed into larger volume with richer cytoplasm and binucleation after 16 d. Positive expression of ALB, AFP, CK18 and CK19 mRNA were detected with RT-PCR, and ALB positive staining was observed by immunocytochemistry as well. In contrast. MNCs cultured without exogenous growth factors scarcely attached to the culture dish and ALB mRNA was not detected. (3) In transplantation experiment, both of ALB and AFP mRNA were detected by RT-PCR and HSA, PCNA and ALB positive staining were observed in the livers of recipient mice by immunocytochemistry. Conclusion: MNCs from human umbilical cord blood could convert into hepatocyte-like cells in 3 different ways, indicating their potential use in the clinic applications for the treatment of human liver diseases.
Abstract:<正>Objective: To determine whether attenuated Salmonella typhimurium producing Helicobacter pylori (Hp) urease subunit B(UreB)can elicit specific immune responses against Hp in mice tested by a lavage technique. Methods: Attenuated Salmonella typhimurium producing Hp UreB immunized orally Balb/c mice twice at a 3-week interval. After 12 weeks,mice intestinal secretions were obtained without harm by administering a lavage solution intragastrically. The mice intestinal secretions of immune group were also directly washed out after the mice were killed. The antibody responses were evaluated by using serum and intestinal fluid with ELISA assay. Results: The multiple oral immunizations with SL3261/ pTC01-UreB induced significantly Hp-specific mucosal IgA response as well as serum IgG response. The IgA was also consistently higher in the intestinal fluid obtained by the lavage solution than by direct washout. In addition, no obvious side effects and changes in gastric inflammation were observed in mice. Conclusion: The attenuated Salmonella typhimurium expressing Hp UreB may be used as an oral vaccine against Hp infection. And the lavage technique is an ideal method in the study of mucosal immune responses.
Abstract:<正>Objective: To investigate the activation (phosphorylation) and subcellular localization of extracellular signal-regulated kinase (ERK1/2). as well as the possible mechanism, following cerebral ischemia and ischemia/reperfusion in rat hippocampus. Methods: Transient brain ischemia was induced by the four-vessel occlusion method in Sprague-Dawley rats. Western blot analysis. Results: During cerebral ischemia without reperfusion ERK1/2 activation immediately increased with a peak at 5 min and then decreased in the cytosol fraction, which was paralleled by the increase of ERK1/2 activation in the nucleus fraction. During reperfusion, ERK1/2 was activated with peaks occurring at 10 min in the cytosol and at 30 min in the nucleus, respectively. Under those conditions, the protein expressions had no significant change. In order to clarify the possible mechanism of ERKl/2 activation, the rats were intraperitoneally administrated with N-methyl-D-aspartate (NMDA) receptor antagonist dextromethorphan(DM), L-type voltage-gated Ca2+ channel (L-VGCC) antagonist nifedipine (ND) 20 min before ischemia, finding that DM and ND markedly prevented ERKl/2 activation of nucleus fraction induced by reperfusion. not by ischemia. Conclusion: These results suggested that the nuclear translocation mainly occurred during ischemia, while ischemia-reperfusion induced ERK1/2 activation both in the cytosol and the nucleus. Two type calcium channels contributed, at least partially, to the activation of ERK1/2.
Abstract:<正>Objective:To observe the effects of different doses of acitretin on the transcription of FGF10 mRNA and the translation of FGF10 protein in cultured HaCaT cells. Methods: HaCaT cells were treated with different doses of acitretin for 48 h, then the changes on the transcription of FGFfO mRNA and the translation of FGF10 protein in these cells were detected by immunofluorescence and in situ hybridization assay. Results: Compared with the control group, the transcription of FGF10 mRNA and the translation of FGF10 protein were gradually decreased along with the increasing dose of acitretin. There were significant differences between different groups (P<0. 01). Conclusion: Acitretin could inhibit the transcription of FGF10 mRNA and the translation of FGF10 protein in HaCaT cells. With the dose of acitretin increased, the stains of both FGF10 mRNA and FGF10 protein in HaCaT cells are reduced.
Abstract:<正>Objective:To study the role of intracellular calcium signal pathway in the down-regulation of aggrecan induced by cyclic tensile strain in the annulus fibrosus cells. Methods:The expression of aggrecan mRNA and core protein were respectively detected with RT-PCR and western blot after the channels transmitting calcium ions were blocked with EGTA, gadolinium and verapamil. Results :EGTA. gadolinium and verapamil partially prevented the effects of cyclic tensile strain on the expression of aggrecan in annulus fibrosus cells. Conclusion:The calcium signaling is involved in the down-regulation of proteoglycan resulting from cyclic tensile strain in the annulus fibrosus cells.
Abstract:<正>Objective: To study extracellular multi-neuron activity in the nervous system based on wavelet-fractal technique. Methods: The wavelet transform was employed to decompose the original signal and obtain 4 sub-patterns. The dividing fractal dimensions of these sub-patterns were computed. A knn-classier was used to classify feature vectors. Results: Not all the elements in feature vector DimDC were very powerful for this pattern recognition problem through the empirical study of noise signals. The most effective feature vector was defined as DimDC= (d3:d4) above. Conclusion:Wavelet fractal algorithm has high accuracy and provides a powerful tool for clinical application.
Abstract:<正>Objective: To explore the fundamental theory, methods and data of the quality of life (QOL) of soldiers and its influence factors in the first response army. Methods:Totally 215 soldiers in the first response army of a military area in China were inquired by using self-regulating questionnaire and WHOQOL-BREF (Chinese Edition). Results:WHOQOL-BREF had good acceptability, reliability and validity in the first response army. QOL of soldiers in the first response army was middling. The influencing factors of QOL of the soldiers in first army includes self-report health, different areas where the army men come from, different arms of services, whether only child in family. And the difference was statistically significant. Conclusion:WHOQOL-BREF is fit for evaluating the quality of life of soldiers in the first response army. There are differences of the quality of life among the soldiers in the first response army. So we should take appropriate measures to improve the soldiers’ quality of life.
Abstract:<正>Objective: To retrospectively study the effects of epinephrine on blood pressure, heart rate as well as renal function in the patients with severe acute pancreatitis complicated with septic shock. Methods :Twenty-five patients with severe acute pancreatitis complicated with septic shock were divided into 3 groups according to the biggest infusing rate of epinephrine used, with the infusing rate of 0. 01-0. 05, 0. 06-0. 10, >0. 10μg/kg·min in group A (n = 9), B (n = 8) and C (n = 8), respectively. Mean arterial blood pressure (MAP), heart rate (HR), urine output, blood urea nitrogen (BUN), creatinine (CRE), urine albumin (U-ALB) and urineβ2-microglubulin (Uβ2-MG) as well as APACHE III scoring were recorded in all the patients. Results: Before anti-shock therapy was given, hypotension, tachycardia, olig-uria as well as the abnormal levels of CRE, BUN, U-ALB, Uβ2-MG and APACHE III scoring occurred in all the 25 patients. With anti-shock therapy, MAP, HR, urine output and BUN, CRE in the patients from the 3 groups gradually returned to normal (P<0. 01 vs before anti-shock therapy), and U-ALB, Uβ2-MG output and APACHE H scoring also restored but still remained abnormal (P<0. 01 vs before anti-shock therapy). Conclusion: The first goal to treat the patients with severe acute pancreatitis complicated with septic shock should be restoring the organ blood supply. Based on volume resuscitation, epinephrine and other vasoactive drugs could be combined to maintain circulatory stability and also could benefit the restoration of the renal function.
Abstract:<正>Objective: To examine the changes of mu-opioid receptors (MORs) expression in human chronic inflamed knee joint synovium tissue. Methods:Knee joint synovium tissues were taken from 21 patients with chronic arthritis (inflamed group) and 6 fresh bodies with normal knee joints (control group). And the expression of MORs was detected by using immunohistochemistry. flow cytometry(FCM) and reverse-transcription polymerase chain reaction (RT-PCR). Results: The expression of MORs in the inflamed group was significantly higher than that in the normal group by using the 3 techniques(P<0. 05). Conclusion: Chronic inflammation enhances the up-regulation of MORs in human knee joint synovium tissue.
Abstract:<正>Objective: To assess the value of MR spectroscopy (MRS) in the diagnosis of prostate cancer by meta-analysis. Methods: Prospective studies were selected from the MEDLINE, Ovid, Embase databases. Springer, Elsevier, China National Knowledge Infrastructure. According to the suggestion, results were determined by the ratio of (Cho + Cr)/Cit . If this ratio was less than 0. 75, negative result was respectively determined, and the definitive tumor was diagnosed as this threshold. According to the assessment standard of Evidence-based Medicine, English and Chinese literature in Grade A and B on MRS imaging was included. According to homogeneity test, different effect models were chosen to calculate different pooled weighted values of sensitivity, specificity and the corresponding 95% confidence intervals (95% CD. Summary receiver operating characteristic (SROC) curves were used to assess the results. Funnel plot was used to analyze publication bias. Results: According to the assessment standard of Evidence-based Medicine, only 5 papers in Grade B were included in this research. The pooled weighted sensitivity and its 95% confidence interval is 82%(73%, 89%) and the pooled weighted sensitivity and its 95% confidence interval is 68% (58%, 76%). The AUC (area under curve) is 83. 40%. An asymmetric funnel plot suggested two missing studies leading to publication bias. Conclusion: If the ratio of (Cho + Cr)/Cit is regarded as the diagnostic criteria in detecting prostate cancer by MRS, meta-analysis suggests this method has a better diagnostic value to detect the malignant prostate mass but the sensitivity needs to be improved. We hope to support a method and requirement about diagnostic test. Performing perspective register and improving quality of study design is the only way to reduce the bias and get real information of disease.
