    • Studies on chemical constituents of Polygala japonica Houtt.

      Abstract:<正>Objective: To investigate the chemical constituents of Polygala japonica Houtt. . Methods: The constituents were separated and purified by column chromatographies with silical gel, RP-silical gel and Sephadex LH-20. Their structures were elucidated on the basis of spectral data (IR, MS, 1H-NMR, 13 C-NMR, HMQC and HMBC). Results: Totally 15 compounds were obtained from Polygala japonica Houtt. and their structures were identified as castaprenol-12 ( I ),β-sitosterol ( II ), stigmasterol ( I ), stigmasta-7, 22-di-ene-3-one (IV ), n-dotriacontanol (V ), n-hexadecanol (VI), arachidic acid ( VI ), ero-gosterol ( VIIII ), kaempferol ( IX ), quercetin ( X ), daucosterol ( XI ), p-hydroxybenzoic acid ( XI ), coumaric acid ( XIII ), rhamnocitrin 3-O-β-D-galactopyranoside ( XIV ), quercetin 3-O-β-D-glucopyranoside ( XV ). Conclusion: Compounds I , I , IV , V , VI , VII , VIII, XII , XIII and XIV were obtained from this plant for the first time.

    • Inhibition of germ tube’s formation of Candida albicans by lactobacillus spp.

      Abstract:<正>Objective:To investigate the effects of 6 strains of lactobacilli and their main culture metabolites on the morphogenesis of Candida albicans (C. albicans) so as to screen medicinal strains as antifungal agents. Methods: Spent culture supernatant (SC’S) at different culture time points, live lactobacilli, heat-killed bacteria and bacterial short-chain fatty acids (SCFA) with different molarities were incubated with C. albicans respectively. Crystal violet-based germ tube assay was used to detect the germination of C. al-bicans and the inhibitory efficiency of each addition was tested by germination rate. Results: SCS of lacto-bacillus spp. decreased the germination significantly and live bacteria of L. rhamnosus. GG, L. acidophilus 1.050103-12 as well as L. johnsonii JCM1022 could partially inhibit the conversion, however, all the heat-killed bacteria failed to control the germ tube formation. In addition, only butyric acid blocked the conversion of yeast to hypha among all the SCFA. Conclusion: The excellent fungistasis activities make L. rhamnosus. GG. L. acidophilus L050103 12 and L. johnsonii JCM1022 potential strains as antifungal drugs and the inhibition seems to have direct correlation to the metabolites.

    • High-yield expression of recombinant mouse coagulation factor Ⅶ in methylotrophic yeast Pichia pastoris

      Abstract:<正>Objective: To explore high yield secretory expression of rccombinant mouse coagulation factor VII (rmFVII ) protein in Pichia pastoris (P. pastoris). Methods: The fragment of mFVII cDNA was amplified by PCR from a pcDNA3 mFVII plasmid. Then the cDNA fragment was suhclonecl intoα-factor secretion signal open reading frame of pPIC9K secretory expression vector. The mutagenesis of mFVII was performed by Site-Direct Mutation and then verified by DNA sequencing. The yeast expression vector of rmF VII. named as pPIC9K-rmF VII . was linearized with Sac I and transferred into GS115 strains(his Mut+ ) by electroporation. The rccombinanls were identified by direct PC’R and selection on MM and MD plates. rmFVII was expressed in recombinant strains (his Mut ) for 4 d. The expression level and activation of rmFVII in the BMMY medium were detected by SDS PAGE and Western blot respectively. Results: pPIC9K-rmF VII was constructed and transferred to GS115 strains successfully. 48 hour post induction by methanul rmFII protein was secreted into the culture supernatant. The molecular weight of the expressed produets was shown to he about 46 kD by SDS PAGE analysis. Western blot showed that the expressed rmF VII exhibited specificity and antigenicity. Conclusion: Since mF VII is considered as a tumor-targeting molecule , this study may provide a basis for further ami tumor strategy on rmFVII.

      Abstract:<正>Objective: To compare renal sodium transport, using fractional excretions of lithium(FELi) as a marker of proximal tubule sodium reabsorption, between hypertensive and non-hypertensive ouabain-treated rats and further to elucidate the role of ouabain in pathogenesis of hypertension. Methods: Thirty male Sprague-Dawley rats weighting 180-200 g were randomly divided into normal control group and ouabain treated group. Rats were infused with 1 ml/kg·d normal saline or 27. 8μg/kg·d ouabain in-traperitoneally once a day respectively. Systolic blood pressure (SBP), heart rate and body weight were recorded weekly. Rats were sacrificed 6 weeks after treatment. Blood and 24-hour urine sample were collected to measure the serum and urinary concentration of sodium, trace lithium and creatinine. Endogenous creatinine clearance rate(Ccr), fractional excretions of sodium (FENa), fractional excretions of lithium (FELi) and fractional reabsorption of sodium in the postproximal tubules (FDRNa) were calculated. Ouabain levels of plasma and renal tissue, plasma renin activity, angiotensin I and aldosterone concentration were determined. Results: 65% of the ouabain-treated rats achieved significantly higher SBP after 4 weeks, compared with that of the saline control groups or self baseline (P<0. 01). But in the other 35% of the ouabain-treated rats, their SBP was similar with control group during the experiment (P>0. 05). The body weight, heart rate and food intake between the 3 groups were no significant differences (P> 0. 05). FELi and FDRNa were significantly lower in ouabain-hypertensive group compared with ouabain-non-hypertensive group and control group(P<0. 01 and P<0. 05). The FEu and FDRn, of ouabain-nonhyper-tensive groups were similar with control group(P>0. 05). Ccr and FENa were comparable between the 3 groups (P>0. 05). Plasma and renal tissue ouabain levels, plasma renin activity, angiotensin I and aldosterone contents in ouabain-hypertensive rats were comparable with ouabain-nonhypertensive rats. Conclusion: Increase of proximal tubule sodium reabsorption play an important role in the pathogenesis of ouabain-hypertensive rats. The change of renal sodium transport may result from regulation to renal Na+ , K + -ATPase by ouabain.

    • Expression of metallothionein-3 gene associated with lung adenocarci-noma

      Abstract:<正>Objective:To explore the role of metallothionein-3 gene in lung adenocarcinoma. Methods: cDNA microarray and northern blots analysis were used to study the expression of metallothionein-3 in lung adenocarcinoma. Results: The results of cDNA microarray showed that metallothionein-3 gene was expressed highly to normal lung tissues at the average ratio of 7. 8 (Cy5/Cy3) in lung adenocarcinoma tissues. Northern blots confirmed the ratio changes detected from the microarray. Conclusion: Metallothionein-3 may play an important role in the lung adenocarcinoma development.

      Abstract:<正>Objective:To analyze the p16INK4a genomic alteration and expression status in 3 human pancreatic carcinoma cell lines with different potential of metastasis. Methods: Using PCR-SSCP, Dot-blot and immunohistochemistry. the p16INK4a genomic mutation and expression were analyzed on DNA, mRNA and protein levels in 3 human pancreatic carcinoma cell lines Patu8902, Patu8988 and SW1990, which had different potential of metastasis. Results :(1) On DNA level: there was no deletion of p16INK4a Exon I in 3 cell lines; p16INK4a Exon II was only deleted in Patu8902 while no deletion in Patu8988 and SW1990. No insertion, microdeletion and point mutation were found in the 3 cell lines. (2) On RNA level: the expression of p16INK4a protein was negative in Patu8902. low expressed in SW1990, but highly expressed in Patu8988. (3) On protein level: P16 protein was strongly stained in Patu8988, much lower in SW1990, but not stained in Patu8902. Conclusion :The genomic type and expression of p16INKa4 are quite different in 3 pancreatic carcinoma cell lines which have different potential of metastasis. It is suggested that genomic ho-mozygous deletion and low expression of mRNA might relate to the potential of metastasis of pancreatic cell lines. In other words, dysfunction of p16INK4a might be an important mechanism in the metastasis of pancreatic carcinoma.

    • The effects of antisense PTEN gene transfection on the growth and invasion of glioma cells

      Abstract:<正>Objective:To study the effects of antisense PTEN gene on the growth and invasion of glioma cells. Methods: A pcDNA3. 1/Hygro(-) recombinant plasmid containing antisense PTEN gene fragment was constructed. Glioma cells of primary culture were transfected with antisense PTEN gene vector and stably transfected clones were selected. Then, the different growth and invasion abilities and the different MMP9 mRNA expressions of three kinds of cells were observed, including the transfected cells, untrans-fected cells and the cells transfected with empty vector. Results:The abilities of growth and invasion of the transfected cells and the expressions of MMP9 mRNA were obviously enhanced. Conclusion: Antisense PTEN gene could have a negative impact on the growth and invasion of primary culture glioma cells.

      Abstract:<正>Objective: A murine model of mixing syngeneic and haploidentical major histocompatibility complex (MHO matched bone marrow cells transplant was used to evaluate the effect of splenocytes in graft-versus-host disease (GVHD) and host-versus graft reaction (HVGR). Methods: BALB/C recipient mice were lethally conditioned with 8. 5 Gy and injected with different grafts which consisted of syngcneic bone marrow cells plus splenocytes (SPLCs) and haploidentical MHC matched hone marrow cells (BMCs) plus different closes of splenocytes. Recipient mice were detected for the percentage of haploidentical MHC matched mouse origin cells in the peripheral blood cells and checked daily for the appearance of GVHD symptoms. Histopathological examination of multiple organs from moribund mice was used to evaluate the grades of GVHD. Results: Recipient mice infused with 10×106 haploidentical MHC matched SPLCs and 5×106 syngeneic splenocytes showed a higher level and more stable chimerism with GVHD II degree histopathological alterations. Histopathological results of GVHD in other group’s hosts were not obvious, and the levels of chimerism were unstable. All of the mice survived over 150 d. Conclusion:The proportion and dose of syngeneic and haploidentical MHC splenocytes are of importance for inducing stable en-graftment on the basis of nonlethal GVHD and to balance GVHD and HVGR.

      Abstract:<正>Objective:To evaluate the changes of brainstem auditory evoked potential (BAEP) in patients with delayed encephalopathy after acute carbon monoxide poisoning. Methods : BAEPs were performed in 32 controls and 40 patients. Wave I , II , III , IV , V latencies and I - III , IIII - V , I - V interpeak latencies were measured, respectively. Results: Abnormalities of BAEPs in 13 patients (13/40, 32%). Among the 13 abnormal BAEPs. 3 displayed prolongation of latency to waves in one side, no potential in another side; 5 displayed a similar abnormality which was bilateral prolongation of latency to waves;and another 5 displayed unilateral latency delay. Compared wave I , II , III , IV , V latencies and I - III , III - V , I - V interpeak latencies in the patients and the controls, there were no significant differences (P>0. 05). Conclusion: BAEPs can be used for evaluating the diagnostic and prognostic values in the cases of delayed encephalopathy after acute carbon monoxide poisoning.

    • Expression of caspase-3 and TRAIL receptors in CD4+ and CD8+ T cells of SLE patients

      Abstract:<正>Objective: To study the expression of caspase-3 and tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) receptors in the CD4 + and CD8+ T cells of systemic lupus enythematosus (SLE) patients. Methods: RT-PCR was used to analyze the expression of caspase-3 and TRAIL receptors in CD4+ and CD8+ T cells of SLE patients and normal subjects. Results: The death domain-containing TRAIL-R1/R2 as well as "decoy" TRAIL-R3/R4 were co-expressed in majority of CD4+ and CD8+ T cells in both SLE patients and normal subjects. The CD8+ T cells from SLE patients showed significantly higher expression of caspase-3 and TRAIL-R2 than those from normal subjects,and the expression was correlated with the activity of the disease. Conclusion: The TRAIL-R2 signal pathway might contribute to the apoptosis of T cells in SLE.

      Abstract:<正>Objective: To evaluate the application of two-dimensional electrophoresis in the research of differentially expressed proteins in the human asthenospermia. Methods: Two-dimensional gel electrophoresis was performed on 4 normal sperm samples from healthy men and 4 sperm samples from 4 asthenospermia patients. After silver staining, the differential expression proteins were analyzed by PDQuest 2D analysis software. Results: Six differential protein spots were identified. Four spots showed increased expression in the control gels compared with the patient gels. Conclusion: The protein profiles of differential expression between the normal spermatozoa and idiopathic asthenospermia were established and some differential proteins were found. The data of this study would establish the better fundament for further isolation and identification of differentially expressed proteins in human asthenospermia sperm.

      Abstract:<正>Objective: To study the relationship between the polymorphism of drug resistant gene rpoB and drug resistance against rifampicin(RFP) of M. tuberculosis L-forms, and to evaluate its clinical application. Methods: A total of 52 clinical isolated strains of M. tuberculosis L-forms were collected. rpoB gene polymorphism was analyzed by polymerasc chain reaction and single-strand conformation polymorphism (PCR-SSCP) and conventional antimicrobial susceptibility test (AST). Their results were compared. Results: AST results showed that 38 of 52 clinical isolated strains were drug resistance (73. 08%), while PCR-SSCP indicated 65. 38% (32/52) rpoB gene polymorphism. There was no statistic significance (X2 = 2. 4914) between the 2 methods. Conclusion:Combined the application of PCR-SSCP with AST in detecting rpoB drug resistant gene polymorphism of M. tuberculosis L-form from pneumoconiosis patients with tuberculosis may have advantages at earlier diagnosis and guidance of clinical medications.

    • The application of island myocutaneous flap for challenging wounds on cervico-thoracic region

      Abstract:<正>Objective: To introduce the experiences in the application of island myoculaneous flap for challenging wound on cervico-thoracic region. Methods: Different myocutaneous flaps were selected according to the location, peculiarity and etiological factor of wound. There were 28 cases of island pectoralis major island myocutanuous flaps, 34 cases of latissimus dorsi island myocutaneous flaps. 19 cases of trapizius island myocutaneous flaps and 17 cases of rectus abdominis island myocutaneous flaps in this report. Results: All 98 patients with challenging wound on cervico-thoracic region were successfully treated with this method without complications, and obtained functional and cosmetic effectiveness. Conclusion : Challenging wounds in cervico-nuchal region can be repaired with pertoralis major island myoculaneous flap, latissimus dorst island myocutaneous flap and trapizius island myocutancous flap, while challenging wounds in thoracic region can be repaired with latissimus dorsi island myocutaneous flap and rectus abdominis island myocutaneous flap. Satisfactory functional and cosmetic results can be obtained.

    • Clinical application of sirolimus in renal post-transplant patients

      Abstract:<正>Objective:To investigate the clinical application of sirolimus (SRL) in renal post-transplant patients. Methods: From January 2003 to December 2004, we adopted a protocol for renal transplantation which included sirolimus in place of mycophenolale mofetil. Their clinical course was evaluated during the first 6 months after surgery. Maintenance immunosupprcssion included sirolimus. corticosteroid and cy-closporine. Sirolimus dosing was initiated at 6 mg on the first day, from then on 1. 2-1. 5 mg as a single daily dose and adjusted to maintain the levels at 5-15 ng/ml. 25 cases were treated with SRL combined with low dose of CsA (4. 5-5 mg/kg·d) and prednisone(SRL group). Another 35 cases were treated with mycophenolate mofefil (MMF 1. 5-2. 0 g/d)combined with CsA (4. 5-5 mg/kg·d) and prednisone(MMF group). Results: Patients’ graft survival rate was 100%. There was no significant difference in average serum creatinine level and incidence of acute rejection between SRL group and MMF group[10. 0% (2/20) vs 11. 4% (4/35), P>0. 05], During the follow-up period, the incidence of side effect was similar in SRL group or MMF group, except for hyperlipidemia in SRL group. Sirolimus was discontinued in 5 patients for adverse events predominantly for numbness of oral lip, delayed renal allograft function, poor wound healing, liver or kidney function injury and pneumonitis. Conclusion: Early outcomes with sirolimus were acceptable with 100% graft survival and 10. 0% incidence of acute rejection. However, because of adverse events including poor wound healing and pneurnonitis, the immunosuppression regimen of SRL combined with low dose of CsA has been limited to clinical application in some degree in early transplant recipients. As one of therapeutical choices, it has been a long way to investigate SRL in clinical extension.
