    • Lung mechanics in canine oleic acid-induced respiratory distress syndrome model

      Abstract:<正> In mechanically ventilated anesthetized dogs (pentobarbital sodium,30mg/kg) with acute lung injury induced by intravenous infusion of oleic acid(OA),changes in airflow,lung volume,and tracheal (Pao),transpulmonary (Ptp)and esophageal (Pes) pressures were measured.Changes in lung mechanics werestudied before and after OA infusion at intervals,during an observation periodlasting 4h,using curve-fitting method,interrupter technique and pressure-volume(PV) loops measurement.The main findings are listed as follows:(1)Complianceand functional residual capacity (FRC) showed a marked decrease,while Pao andPtp showed a marked increase within 2h after OA.(2) Airflow resistance atexpiratory phase estimated by the curve-fitting method showed a marked increaseafter OA.(3)PV loops of the lungs or total respiratory system showed similarchanges 4h after OA,characterized by the presence of an inflexion point at theinflation limb,increased hysteresis and rightward and downward shift of the loop.It is suggested that use of the curve-fitting method in combination with the PVloop for the total respiratory system would be more appropriate for monitoringlung mechanics during mechanical ventilatory support of ARDS patients.

    • Effect of berberine on reperfusion arrhythmias in the isolated rat heart

      Abstract:<正> Effect of berberine on reperfusion arrhythmias was studied using a10min left anterior descending coronary artery occlusion followed by reperfusionin the isolated rat heart.Berberine when given 10min before coronary ligation re-duced the incidence of reperfusion arrhythmias in a dose-dependent manner.With10-5,3.16×10-5 and 10-4mol/L of berberine,ventricular fibrillation was reducedfrom its control incidence of 100% to 90%,70% and 40% (P<0.05) respectively.Heart rate was also reduced in a dose-dependent manner,falling from its controlvalue of 282±11 to 180±11 beats/min with the highest concentration of berberine.Coronary flow was increased in all berberine treated groups.With anantiarrhythmic dose of berberine (10-4mol/L),hearts were paced (5 Hz) duringischemia period;under these conditions the anti-arrhythmic effect of berberinewas lost.When berberine (3.16×10-5 and 10-4mol/L) was administered just beforereperfusion,no anti-arrhythmic effects were observed.The α-adrenoceptor antago-nist phentolamine (2.6×10-5mol/L) prevented reperfusion arrhythmias and ismuch more effective than berberine.These results suggest that the prophylatic ef-fects of berberine on reperfusion arrhythmias in the isolated rat heart are due toits negative chronotropic properties.

    • Measurement of individual hypoxic sensitivity by means of mouth occlusion pressure

      Abstract:<正> mouth occlusion pressure (P0.1) rather than inspiratory ventilation (V1)and mean inspiratory airflow ((?)) was used for expounding the mechanism of indi-vidual hypoxic sensitivity.Eighteen young healthy male subjects participated inthe experiment of progressive isocapnic hypoxia produced by rebreathing method.The results showed that there were significantly linear relationship (P<0.01) be-tween V1 and P0.1,as well as (?) and P0.1 during the hypoxic loading of twoend-tidal carbon dioxide pressure (PETCO2 levels,4.3 and 5.9kPa.Ventilation in-creased with progressive hypoxia.Therefore,they all represent the useful indexesof inspiratory drive.P0.1 is more sensitive than V1 and (?) because it isindependent of pulmonary mechanics.

    • Down-regulation of glucocorticoid receptor mRNA by glucocorticoids in rats

      Abstract:<正> The effect of glucocorticoids on the down-regulation of glucocorticoidreceptor (GR) mRNA was studied in intact rats.GR mRNA was characterized byNorthern blot hybridization and quantitated by dot blot hybridization using a hu-man GR cDNA fragment as a probe.Administration of hydrocortisone (F) inpolyvinyl alcohol (PVA) resulted in a rapid increase in plasma glucocorticoidswhich maintained at stress levels (20 to 40μg/dl) for about 3 d.HepaticGR mRNA decreased significantly to 73.5±6.3% of control values 6h followingF treatment,after which the decline of GR mRNA was gradual,reaching a mini-mum of 44.0±5.0% of control levels 3d after the treatment.The effect of F onthe down-regulation of hepatic GR mRNA lasted up to 11 d.In contrast,F treat-ment had no effect on GR mRNA in rat brain.These results are consistent withthe changes in GR in rats as reported previously,except that even though thehepatic cytosol GR decreased markedly,no significant changes in hepatic GRmRNA were found 1h after F treatment,strongly suggesting that thedown-regulation of GR by its ligands in vivo occurs at both transcriptional andposttranscriptional levels and is of tissue-specific fashion.

      Abstract:<正> using a WZD—1 microcirculation microscope,we observed the tongue tipand nail-bed microcirculation of uremic patients with deficiency of kidney-yangbefore and after blood dialysis.The results showed that the pathological basis ofuremia with deficiency of kidney-yang may be microcirculation disturbance,in-crease of metabolites and water retention.The effusion from capillary loops intongue tip may be one of the pathogenetic factors of a swollen and jaggedtongue.The flow rate of blood in capillary loops in the nail-bed was apparentlyabnormal and markedly improved after blood dialysis.The weighted integral ofnail-bed microcirculation decreased from moderately abnormal (4.99±1.52) toapproximately normal (1.98±0.97) (P<0.01).Microcirculation seems to be a sensi-tive criterion for reflecting the pathologic changes of uremia with deficiency ofkidney-yang.

      Abstract:<正> Thirty feet of Chinese adult cadavers were dissected under an operativemicroscope to observe the extensor hallucis brevis and extensor digitorum brevisand their vessels and nerves in order to design myocutaneous flaps.It was be-lieved that the lateral tarsal artery and the lateral branch of the deep peronealnerve are the best choice to be included in the pedicle when muscle flaps ormyocutaneous flaps are to be obtained from the dorsum of foot.The size of themuscles may be enlarged both laterally and medially as the clinicalcondition requires.

    • Antibody-mediated targeting of doxorubicin-containing immunoliposomes to human gastric cancer

      Abstract:<正> Doxorubicin (DXR) was entrapped in liposomes coated with themonoclonal antibody,MGb2,against human gastric cancer.Negative-stain electronmicroscopy showed the diameter of liposomes was 105.6±21.1nm.The drugentrapment efficiency was 47.6±8.1%.There were about 5667 drug moleculesentrapped and 48 antibody molecules incorporated per liposome.It was confirmedby ELISA and binding assay that the immunoliposomes could bind selectively andcarry their contents to human gastric cancer cell line,BGC-823.Theimmunoliposomes showed specific cytotoxicity to BGC-823,2.3-fold more effectivethan free DXR and 56.7-fold more effective than non-specific liposomes,but only1/25 of toxicity to normal human embryonic lung cell line,SL7,as comparedwith free DXR.This study indicates that the immunoliposomes could deliverDXR selectively to the target cells and might be useful in targeting chemotherapyof human gastric cancer.

    • Functional changes of pulmonary surfactant in rats with lung injury induced by endotoxin

      Abstract:<正> We studied the functional changes of pulmonary surfactant (PS) in acutelung injury models produced by endotoxin injection (E.coli O55B5) in rats.The sur-face properties of the lung lavage liquid and the total phospholipids (TPL) ex-tracted from it were assessed on a modified Wilhelmy film balance.γ-A isothermof the lavage liquid revealed an increase in minimum surface tension and a de-crease in hysteresis area,recruitment index and stability index,whereas that ofTPL extracted from it did not show any change except for hysteresis area.Thesurface activity correlates positively with the TPL content but negatively with thetotal protein content in the lavage liquid.The findings indicated that there was adysfunction of PS in rats with the lung injury induced by endotoxin,suggestingthat the function deficiency of PS might be caused by decreased phospholipidsand increased proteins in the alveoli.

    • Experimental study on the method of efficient heat acclimatization in macaques

      Abstract:<正> Twenty-eight adult macaques were divided into five groups for differenttreatments.Of these five groups,groups 1 to 3 were exposed to heat once dailyfor 10d until the rectal temperature (RT) rose to 40℃,40.8℃ and 41.6℃,respectively.The other two groups (groups 4 and 5) were exposed to heat twicedaily and every other day,respectively,until the RT of both groups reached41.6℃.The method of heat exposure to obtain the required RT was to keep themacaques in a hot chamber (Td 41℃,Tw 38℃).Before and after theacclimatization period,the macaques were given a standard high temperature testin the same environment as the heat acclimatization (HA) period until the RTrose to 41.6℃.The results revealed that the animals in groups 3 to 5 whose RTin HA increased to 41.6℃ showed a significant HA effect by prolonging the dura-tion of exposure (P<0.05~0.001) and decreasing the RT (1.14~1.83℃),especiallyin the daily exposed group.Therefore,one of the most effective ways for devel-oping full acclimatization in macaques is to expose to heat once daily for 10duntil the RT rises to 41.6℃.

    • Mechanism of palliative effect of pethidine on tracheal hot air inhalation injury in rats

      Abstract:<正> The increases of tracheal vascular permeability and malondialdehyde(MDA) content 1,2 and 3 h after introduction of hot air into the rat tracheaswere markedly inhibited by pethidine.The decreases of tracheal superoxidedismutase (SOD) activity 2 and 3h after hot air injury were elevated bypethidine.However,the inhibitory effect of pethidine on tracheal vascularpermeability was not antagonized by naloxone.This implies that opiate receptorsensitive to naloxone is not involved in the inhibitory effect of pethidine ontracheal vascular permeability.Pethidine could decrease MDA content and in-crease SOD activity.These antioxidative effects are beneficial to decreasingvascular permeability.

      Abstract:<正> The purpose of these experiments was to investigate the possible ionicbases for ranitidine (Ran) in isolated papillary muscles of the guinea pig.Ran,when the concentration was below 300μmol/L,had no influence on contraction ofnormal muscles and only above 0.3 mmol/L,it showed weak negative inotropic ef-fect.The contractile activity elicited by histamine in potassium depolarized mus-cles was depressed by Ran markedly and dose-dependently.The IC50 of Ran was0.273μmol/L.This depressant effect was reversed by an elevation of externalCa2+concentration to 7.7mmol/L.The contraction induced by isoproterenol inpotassium depolarized preparations was not altered by Ran.Ran at theconcentrations of 1,10 and 100μmol/L failed to show influences on fast actionpotentials.However,Ran at 0.5μmol/L time-dependently inhibited slow action po-tentials.It is concluded that the ionic mechanism of Ran might be attributed toantagonized H2 receptor of myocardium which in turn decreases Ca2+ inward cur-rent.

      Abstract:<正> Rapid debridement and 1% chlorhexidine iodine complex (CIC)wereused for treating badly cdntaminated and/or infected missile wounds 72 h after in-jury.The result showed that the combined use of the two methods proved to behighly effective in clearing out the necrotic tissues,bony debris and pathogenicbacteria,and in achieving primary healing.The problems about the extent andthe method of debridement in the early débridement of the fire-arm injury are dis-cussed in this paper.

    • Ototoxicity induced by kanamycin in iron-deficient young rats

      Abstract:<正> Using auditory electrophysiological,histopathological,enzymehistochemical and scanning electron microscopic techniques,we observed thechanges of function and structure of cochlea in iron-deficient growing rats receiv-ing kanamycin intramuscularly.The results showed that at an equal dose ofkanamycin,hearing loss and hair cell damage were more serious,and incidence ofhearing loss was much higher in iron-deficient rats than in normal rats.Theinhibition of succinate dehydrogenase activity of cochlea induced by interaction ofkanamycin and iron deficiency was more obvious than that by either one of them.Iron deficiency can increase the sensitivity of cochlea to kanamycin.The resultsindicated that iron deficiency may be one of the reasons for high incidence ofototoxic deafness in children,and it should be cautious when using such drugs inchildren with iron-deficient anemia.

      Abstract:<正> Urea-soluble antigens of Trichinella spiralis infected larva were preparedand the efficiency of the antigens in diagnosis of trichinosis by ELISA wasstudied.Specific IgG antibodies were detected by ELISA using urea-solubleantigens in 91.67% (22/24) of 24 serum samples taken from patients withtrichinosis and 91.67% (11/12) of 12 blood spots on filter paper.Specific IgMantibodies were seen in 95.24% (20/21) of 21 serum samples from patients withtrichinosis using the antigens.Sera of patients infected with Schistosoma japonicumhad some cross teactions to the antigens,but the sera of mice infected withSchistosoma japoni(?) Ancylostoma caninum larva and Plasmodium yoelti were allnegative in the test.

      Abstract:<正> Estrogen and progesterone receptor (ER and PR) contents of 40 breastcancer patients were determined with immunofluorescent staining.Thedifferentiation of cancer cells was evaluated on the basis of histological andultrastructural changes.It was found that there was a positive correlation of ERand PR contents with the cellular differentiation of breast cancer,and the lattercould be used to estimate the status of hormone receptors.Breast cancer is a malignant disease with heteropathological characteristics.In handling a breast cancerpatient,both the status of hormone receptors and the cellular differentiationshould be considered and the treatment varied with each pa(?)ent.For those witha well-differentiated cancer and rich hormone receptors,endocrine therapy shouldmainly be administered,while for those with a poorly-differentiated cancer and afew hormone receptors,chemotherapy would be of choice.

    • The design and application of NiTi shape memory alloy bow staple in cranioplasty

      Abstract:<正> The NiTi shape memory alloy (SMA) bow staple consists of an arched body,one front arm and one rear arm.It has the function of shape memory and manyadvantages,i.e.,high strength,low specific gravity,strong retrieving force,highresistance to fatigue and erosion,nontoxicity,nonmagnetism and goodhistocompatibility.It can be widely used and make the fixation convenient and ef-fective.The staple has been used clinically in 152 cases of cranioplasty.Resultshave showed that duration of the operation is shortened and harm during the op-eration is reduced,because dura mater needn’t be seperated and the skull needn’tbe penetrated through.Some complications can be avoided and the operationbecomes simpler,safer and more reliable.The bow staple is suitable for fixationwith any kind of repairing material in cranioplasty and does not interfere withthe CT and MRI reexaminations.
