    • Study and Application of Monoclonal Antibodies against Virus of Haemorrhagic Fever with Renal Syndrome

      Abstract:<正> This paper summarizes the study and application of monoclonal antibodies (McAbs)against haemorrhagic fever with renal syndrome (HFRS) virus in our department over the past sever-al years. The following six points are discussed: (1) establishment and characterization of thehybridoma cell lines secreting McAbs against HFRS virus and hemagglutinin of the virus: (2) theantigenic analysis of HFRS viruses in China by McAbs: (3) purification and application of theMcAbs: (4) purification and characterization of HFRS virus 50K stnactural protein by McAba-affini-ty chromatography and the McAbs possessing different characteristics: (5) detection of HFRS virusantigen in peripheral blood lymphocytes from HFRS patients by the McAb-IFAT; and (6)development of McAb-ELISA indirect sandwich methods, and detection of HFRS virus antigen andIgM, IgG and/or HI antibodies in human and animals. The results of the studies show that theMcAbs can be used for early diagnosis, epidemiological investigation, preparation of vaccine andimmunotherapy of HFRS.

      Abstract:<正> The lipopolysaccharide (LPS, endotoxin) isolated from Bacteriodes fragilis NCTC 9343 ex-hibted an endogenous heterogencity consisting of R (rough) and SR (semi-rough ) LPS fractionsupon sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The LPS was chemically consti-tuted by D-glucosamine, D-glucose. L-rhamnose, D-galactose. fatty acids and phosphate in amolar ratio of 2: 1: 1: 4: 5. 4.: 2.7 The fatty acids were predominated by 3-hydroxypentadecanoic(3-OH-15: 0), 3-hydroxyhcxadecanoic (3-OH-16: 0), 3-hydroxyheptadecanoic (3-OH-17: 0), 3-hydroxy-15-methyl hexadecanoic (3-OH-15-Me-16: 0) and 13-methyl-letradecanoic acid (13-Mc-14:0). Approximately 1 mol of 2-Keto-3-deoxy-D-mannooctonatc (KDO) was detected after treat-ment of LPS with 48% of hydrofluoric acid (HF), indicating it was present as a phosphorylatedmolecule.

    • The Role of Ca2+ and Cyclic AMP in the Modulation of BenzodiazepineReceptor on Aldosterone Secretion

      Abstract:<正> PK11195, a ligand of peripheral-type benzodiazepine receptor can stimulate thealdosterone secretion of isolated adrenal glomerulosa cell: this effect could be abolished by nifedipinewhich mainly blocks the calcium channel in plasma membrane, but could not be abolished bydantrolene, a selective blocker of mitochondria calcium channel. Even under the condition of themaximum stimulative effects on aldosterone secretion, PK11195 could not change the cyclic AMP(cAMP) content in isolated glomerulosa cells. These results indicated that in the modulatory mecha-nism of benzodiazepine receptor on aldosterone secretion, the intracellular messenger might be theCa2+ from extracellular Ca2+ pool, but not the Ca2+ from mitochondria Ca2+ pool or cAMP.

    • Effects of Sympathetic Denervation and Dexamethasone on Pial Microvascular Pressure in Acute Hypoxic Rabbits

      Abstract:<正> To determine the effects of sympathetic denervation and dexamethasone on the individualcerebral vascular pressure profile during hypoxia, the authors measured the pial microvascular pres-sure in 24 anesthetized rabbits with a servo-null system. At a simulated altitude of 4000 meters, inthe intact group, the mean pressure in pial arterioles of 50-70 μm in diameter decreased markedly.The ratio of pressure drop between the upstream and downstream of the arteriole measured in-creased markedly, and the blood flow in the internal carotid artery was nearly doubled of the nor-mal. The pial arteriolar pressures of both the denervated and dexamethasone groups were significantlyhigher than that of the intact group, but were not different from their prehypoxic values. The ratioof pressure drop between the upstream and downstream of the denervated and dexamethasonegroups remained unchanged, and much lower than that of the intact group, These results lead tothe following conclusions: (1) Sympathetic nerves and dexamethasone exert no effect on thelongitudinal distribution of cerebro-vascular pressure in rabbits during normoxia, normocapnia andnormotension.(2) During hypoxia, pressure drop increases in the upstream arteries and/or decreasesin the downstream vessels of the microvascular pressure measured site (arterioles 50- 70 μm in diame-ter) partly due to the activation of the sympathetic nerve. (3) Dexamethasone can prevent theredistribution of cerebrovascular pressure induced by hypoxia, which can partly explained the ac-tion of dexamethasone in preventing high altitude sickness.

    • Evaluation of α-L-Fucosidase in the Diagnosis of Primary Hepatic Carcinoma

      Abstract:<正> α-L-fucosidasc (AFU) activity was determined by referring to Troost’s method in sera, liv-er tissues and lesions from patients with hepatoccllular carcinoma (HCC), other liverspace-occupying lesions (SOL) and diseases. The results showed that the sensitivity and specificityfor the diagnosis of HCC were 81.2% and 83. 3% respectively with the assay of serum AFU activi-ty, and sensitivities in the diagnosis of AFP-negative and early HCC were 76.1% and 70. 8%,respectively. The AFU activity in the HCC tissues and their pericancerous liver tissues was twice asmuch as controls, and consisted with their serum AFU activity (r=0. 742, P<0.01). It is sug-gested that AFU could be a new biologic marker of HCC and have an important value in thediagnosis of HCC.

      Abstract:<正> The mean length of the stria vascularis (SV) and the basilar membrane of four normalguinea pigs measured by means of surface preparation was 21.11 mm and 19. 90 mm respectively,and the mean width of their apical turns of SV and the turns of the cochlear basis was 0. 44 mmand 0. 86 mm respectively. The ultrastructure of SV, spiral ligament (SL) and spiral prominence(SP) of both ears of eight guinea pigs were observed by scanning electron microscopy(SEM) andtransmission electron microsoopy(TEM). There were a large number of coated vesicles in thesupranuclear region of SV, some opened to the endolymph. A great number of folding cytoplasmicprojections were filled with small vesicles The capillary network was extremely rich indicating thatthe endolymph could be secreted and absorbed by SV. The cellular conjunction in the SV. SL andSP formed an effective barrier. It has not been proved in this study that the melanin offers resistanceto acoustic stimulation.

      Abstract:<正> A quantitative method using fluorescence spectro-photometer to detect theinterleukin 2 reocptor (IL 2R) on the surface of mononuclear cells in normal humanperipheral blood is described. For expression of IL 2R, the optimum conoentration of PHA andCon A were 100 μg/ml and 10 μg/ml respectively. The PHA induced effect on mononuclear cellswas better than Con A induced one. The peak value of IL 2R cxpression was at the 24th h afterstimulation The optimum dilution of the first antibody (anti-Tac) was 1: 50, while the dilution ofthe second antibody(fluorescein conjugated rabbit anti-mousc IgG) was 1: 32 Their optimumincubation period was 20 and 10 min repectively. The concentration of fluorcscein-conjugatedantibody per 1×106 mononuclear cells from the same normal human peripheral blood was5.22±0.45×10-10 mol, and the coefficient of variation was 8. 6%. The concentration offluorescein-conjugated antibody per 1×106PBMC from five healthy individuals was 4. 8±0. 97×10-10 mol Therefore this assay system is stable and quite reproducible.

      Abstract:<正> Surgical treatment of tongue tumors usually results in a tongue defect half or more thanhalf of its original size which not only interferes with speech, swallow and mastication functions butalso creates contour deformity and induces mental suffering In order to restore the contour and func-tions of the tongue 21 cases of tongue defects were repaired and reconstructed with forearm radialor ulnar cutaneous or myocutaneous flaps All the transplanted flaps survived and the restored con-tour and functions were satisfactory. The surgical procedure of tongue reconstruction was intro-duced and how to have a satisfactory recovery of the tongue functions and raise the rate of successof the operation were discussed.

      Abstract:<正> This report describes for the first time the effects of hyperbaric oxygen therapy with 253kPa on patients(n=24) with post - trauma acute compartment syndrome, and the relationship be-tween the prognosis of the syndrome and the changes of plasma fibronectin. Plasma fibronectin wasmeasured using single radial immunodiffusion both before and after therapy with hyperbaric oxygen,with 30 healthy people as control for plasma fibronectin. The results showed that the clinical symp-toms and signs of all 16 patients with acute compartment syndromes in early stage were significantlyimproved after 3 times of hyperbaric oxygen therapy and disappeared after 5 times withoutfasciotomy. Six patients who were treated with hyperbaric oxygen after fasciotomy showed reductionin the infection, and, edema of the injured limb However, the effects were less favorable for 2 pa-tients with necrosis although their clinical symptoms and signs were partially improved. Besides,we found that these patients’ plasma fibronectin concentrations were significantly afterhyperbaric oxygen therapy. The mean increase was 43 (18. 9%; P<0. 01), 61( 24. 7%, P<0. 01),49 (17. 2%, P<0. 01), 57 (17. 4%; P<0. 05) and 55 mg/L ( 16. 6%; P<0. 05) after 1, 2, 3, 4and 5 times of therapy, respectively. Plasma fibronectin concentrations returned to the level of thecontrols after 3 times of therapy (P>0. 05). After fourth and fifth therapy plasma fibronectin lev-els of the patients rose higher than those of the controls (P<0.05). The results suggest that hyperbaric oxygen therapy has a beneficial effect on patients withacute compartment syndrome in the early stage Hyperbaric oxygen therapy can reduce edema andskeletal muscle necrosis The method can be used to treat patients with acute compartmentsyndrome in early stage and as an effective adjunctive treatment after fasciotomy.

      Abstract:<正> A hybridoma cell line secreting monoclonal antibody to idiotope of the monoclonalantibody 94D1 specific for asexual erythrocytic stages of Plasmodium falciparum was established byfusion of SP2/0 mouse myeloma cells with spleen cells of Wistar rats immunized with monoclonalIgG of 94D1 purified from ascitic fluid of BALB/C mouse by affinity chromatography on ProteinA- Sepharose CL- 4B. Specificity of the anti - idiotope antibody (anti - Id), 41RF5, was deter-mined with indirect enzyme-linked immunosorbent assay (ELISA). The results showedthat 41RF5 reacted only with 94D1 IgG2b, not with normal mouse IgG and other seven mousemonoclonal antibodies - five specific for erythrocytic stages of P. falciparum, one for erythrocyticstages of P. inui, and one for Dengue fever virus type III, which indicates that 41RF5 does recognizespecifically the idiotope of 94D1 monoclonal antibody. In 41RF5 heterohybrid culture supernatant,anti-Id titre measured by ELISA was above 1:1 280. Up to the present, the heterohybrid cell linehas been cultured stably for 16 months.

      Abstract:<正> The levels of plasma renin activity(PRA), (?) II(ATII), serum sodiurnand urinary sodium excretion within 24 h m heart failure (?) were (?), in onder to studythe effect of sodium on the PRA and ATII levels in heart (?) patients The (?) showed thatthe PRA and ATII levels were higher left heart falure patents than those in nonnal persons andright heart failure patients, but the serum sodium and urinary sodium excretion left heart failurepatients was lower than that of normal persons and right heart failure patients that patients withleft heart failure were of high renin activity type and patients with right heart failure were of low ornormal renin activity type and that in heart failure patients the PRA and ATII level and urinary so-dium excretion were inversely conelated We suggest that it may be useful in selecting drugs forheart failure (?) when the patients are divided into subgroups by PRA and sodium index.

    • Histological Changes and Differential Diagnosis of Mesenchymal Chondrosarcoma

      Abstract:<正> Eight cases of mesenchymal chondrosarcoma either of skeletal(5 cases) or extrasketetalorigin (3 cases) are reported. According to histopathological and diagnostic criteria,mesenchymal chondrosarcoma were classified into two types cartilage isiand cell type andundifierentiated small cell type It is believed that the application of this classification in the study ofmesenchymal chondrosarcoma is helptul the estimation of its malignancy and choice of treatmentas well al in the prediction of its prognosis. Mesenchymal chondrosarcoma is a highly malignant neoplasm with poor prognosis. None ofthe patients in this series survived more than five years after they were diagnosed. The diffentialdiagnosis of malignant lymphoma, chondrosarcoma, hemangiopericytosarcoma, etc. was discussed.My data support the assumption that mesenchymal chondrosarcoma originates from the secondarymesenchyme rather than the primary mesenchyme.

    • Acoustic Trauma of Guinea Pigs Caused by 60 Cobalt γ-Ray

      Abstract:<正> Auditory cortical cvokod rcsponse(ACER) and auditory brainstom cvoked response(ABR) were recorded in guinea pigs before and after irradiation of 60Co γ-ray. A single dose ofirradiation on head was 90Gy and 70Gy in two groups, respectively. Two h after irradiation. thethreshold shift was 12. 5 dB and 9 dB, and the ACER (?) post-irradiation amplitude ex-ceeded the maximum pre-irradiation value by 20% and 37%. Six h after irradiation, the thresholdshift increased to 37 dB and the maximum amplitnde decreased to the level of pre-irradiation in70Gy-group. In another group, right auditory bulla arca was irradiated with a total dose of 45Gy ina course of six fractions/two weeks. Fourteen d after irradiation, the threshold shift of ABR was 118dB and the latency of wave I was longer. The amplitude-intensity curve was (?) afterirradiation. Histological observation using scanning (?) showed that only outer hair-cells in the basal coil of the cochlea were (?) 6 h after irrdiation 50Gy-group. Bul 16 hlater, outer, hair cells in all coils of the cochlea were found to be destroyed extensively and inner (?)cells were destroyed slightly. In 45Gy-group, 14 d after irradiation, outer hair cells in all coils of thecochlea were damaged and no inmer hair cell was injured severely.
