Abstract:The aim of this study is to investigate epigenetic mechanism of ABCG2 induced drug-resistance.It is not only expatiate for drug-resistance regulation mechanism in all-round,but also to provide scientific experimental basis for selecting target to reverse its drug-resistance.Apply methylation-specific PCR(MSP) to have tested methylation of ABCG2 promoter region -359 to -353 specific positions in breast cancer tissues and paired adjacent tissue of 22 cases,and test their methylation positions with MSP products for sequencing;and adopt fluorescent quantitation RT-PCR to test expression level DNMT1,DNMT3A,DNMT3B and ABCG2;to make analysis on relationship between them with statistical spearman correlation.Specific positions of ABCG2 gene promoter region of 18 cases among the 22 cases with breast cancer(18/22,82%) existed high methylation(P<0.05),MSP products sequencing proved methylation of the specific position,and mRNA expression level was relative higher in remarkable positive correlation(P<0.05).ABCG2,DNMT1,DNMT3A,DNMT3B mRNA expression levels in breast cancer tissues were obviously higher than adjacent tissues(P<0.01),and DNMT3B expression level was obviously higher than DNMT1 and DNMT3A(P<0.01) in negative correlation with ABCG2 gene expression(P=0.001).-359 to -353 positions of promoter regions of ABCG2 gene existed high methylation capable to push expression of this gene in beast cancer tissue.DNMT3B is involved in expression regulation in ABCG2 gene,and provides new scientific basis for drug-resistance target as reverse ABCG2 induction

Abstract:The purpose of this study was to explore the change of telomerase in passage from human endometrial stromal stem cells isolated from human endometrium.Telomerase activity of cultured endometrial stromal cells was assessed at mRNA and protein levels using RT-PCR and immunohistochemistry technique.Telomerase mRNA and protein levels were higher at early passages,and then had a gradually decreased trend of immunoreactive intensity and gradually weakened positive cells with progressive passage in endometrial stromal stem cells.These results suggest that telomerase is contributed to the insenecence of endometrial stem cell.

Abstract:Acute leukemia is one of the most common hematologic malignancies and its exact mechanism of development is unknown yet.In clinical,acute leukemia always accompany with abnormal iron balance.Ferritin,folic acid and vitamin B12 are important components of blood cells.In this study,we measured variations of serum levels of ferritin(SF),folic acid and vitamin B12 in the acute leukemia patients under different disease stages including first diagnosed stage,complete remission(CR) stage and not remission(NR) or relapse stage.It demonstrated that serum SF levels in patients at the first diagnosed stage and NR or relapse stage were significantly higher than the CR stage in acute myelocytic leukemia(AML) patients and acute lymphoblastic leukemia(ALL) patients(P<0.05).The serum folic acid levels in patients at the first diagnosed stage and NR or relapse stage were significantly lower than the CR stage in AML patients and ALL patients(P<0.001).Whereas,serum vitamin B12 levels in AML patients were significantly higher at the first diagnosed stage and NR or relapse stage than the CR stage(P<0.001).And it was significantly higher in ALL patients at the NR or relapse stage than at the first diagnosed stage and CR stage(P<0.05).There are obvious variations of serum levels of SF,folic acid and vitamin B12 in acute leukemia patients under different stages.Increased serum levels of SF and vitamin B12 and decreased levels of folic acid may correlate to the active degree of acute leukemia as well as tumor load.

Abstract:Objective:To investigate the association of the expression of estrogen receptor α,estrogen receptor β in placenta with intrahepatic cholestasis of pregnancy(ICP) susceptibility.Methods:In 14 cases of mild ICP,14 cases of severe ICP and 14 cases of normal cases(control group) with corresponding age and gestation weeks,the expressions of ERα and ERβ were detected by means of immunohistochemical method S-P.Results:The mean grey numbers of ERα in each group mentioned above were 151.68±3.76,149.85±3.69,153.18±3.18,without significant difference(P>0.05).The mean grey numbers of ERβ in each group mentioned above were 146.51±3.81,139.43±9.97,149.87±4.17,with significant difference(P>0.05);the expression of ERβ of severe ICP group was significantly higher than that of the mild ICP group and the control group(P<0.05).The expression of ERβ in every group was higher than that of ERα(P<0.05).Conclusion:ERβ maybe play an important part in the etiology and development of ICP.

Abstract:LaparoEndoscopic Single-site(LESS) renal surgery emerging as a potential alternative to conventional laparoscopy,is technically challenging and the major vascular anomaly may increase the risk of intraoperative haemorrhage.Herein,we present a case of right transumbilical LESS radical nephrectomy which was successfully performed in the presence of double inferior vena cava and duplicated the standard laparoscopic techniques.Most importantly,to bring such an aberrant vascular anatomy to the attention of laparoscopic,especially LESS surgeons with high resolution pictorial illustrations.

Abstract:Recently, chitosan nanoparticles have been an attractive biomaterial for their unique properties. Chitosan nanoparticles are mainly used as drug delivers. However, they are seldom used in wound healing after severe bleeding. In the present paper, chitosan nanoparticles with an average size of 60.2 nm were prepared through ionic gelation and characterized by X-ray diffraction and FT-infrared spectrometry. A new animal model of severe bleeding was made to evaluate its effects on wound healing besides hemostasis. The results showed that it can achieve well hemostatic effects and also induce wound healing after severe bleeding. Blood cells can be absorbed by chitosan nanoparticles and the bleeding is treated. Furthermore, the chitosan nanoparticles sustains longer than other materials in wound healing. At the 15th day after operation, the chitosan nanoparticles with the size of 58.3 nm can still be seen in the new-regenerated hepatic tissue. It can induce the formation of a clear granulation around the wound site. Through histological observation, we find out that chitosan-TPP nanoparticles can induce wound healing through migration of the inflammatory cells and synthesis of collagen. So, chitosan nanoparticles show us both hemostasis and the ability of wound healing for parenchymal organs.

Abstract:The interrupted aortic arch (IAA) is rare congential heart defect and entails a very poor prognosis without surgical treatment. We describe a young adult patient with IAA associated with severe pulmonary hypertension and discuss the perioperative anesthetic management of single-stage surgical correction in this patient. Preoperative anesthetic preparation with better understanding of the malformation, intraoperative hemodynamic monitoring, and adequate alveolar ventilation in the perioperative period were all important factors contributing to a successful outcome.

We present such a case who received a successful long-term single intravenous prograf administration to transitionally maintain the immunosuppression.
