      Abstract:Objective: Single nucleotide polymorphisms (SNP) of ATP-binding cassette transporter A1 (ABCA1) gene are related to plasma lipid and susceptibility to coronary artery disease (CAD). Our first goal was to screen all 50 coding regions of ABCA1 to find new SNPs. Our second goal was to investigate the frequency distribution of R1587K and M883I polymorphisms of ABCA1 gene, which are the variant occurred most frequently, in Chinese people and to evaluate their association with the CAD phenotype and plasma lipids. Methods: Single-strand conformation polymorphism (SSCP) and DNA sequence were used for confirming new SNP of ABCA1, and restriction fragment length polymorphism (RFLP) were applied for confirming genotypes of R1587K and M883I in 112 CAD cases and 108 healthy people. Results: We discovered a new ABCA1 SNP in Chinese population, which converse 233 amino acids from Methionine to Valine (M233V). This new ABCA1 SNP located in exon7, and might potentially modulate the biological function of lipid metabolism. For R1587K and M883I SNPs, the K allele and I allele frequency was 28.9%and 31.1%, respectively. The K allele at R1587K conferred lower mean values of HDL-C in a dose-dependent manner in both CAD patients and healthy people. However, 883I allele was not associated with plasma lipid level. Neither 1587KK nor 883II associated with increased risk of CAD. Conclusion: Our study finds a potential functional ABCA1 SNPs and revealed K allele of R1587K associated decreased HDL-C level in Chinese population.

      Abstract:Objective:To gain purified recombination protein rGPI-vWF A3 which can guide endothelial progenitor cells (EPCs) gathering to the internal surface of the injured vessels theoretically and analyze its bioactivity in vitro. Methods: The plasmid vector of vWF A3 fusion protein which contained GPI structure was constructed and recombination protein rGPI-vWF A3 was expressed and its bioactivity was analyzed by ELISA and FCM. Results: Fusion protein vWF A3-GPI was expressed and purified by immunoaffinity chromatography and it could stably anchored on the surface of EPCs and showed its biological activities of collagen conjunction. Conclusion: These findings represent a novel strategy in EPC transplantation treatment for vessel injury.

    • A versatile cloning vector facilitates target gene expression in prokaryotic and eukaryotic cells

      Abstract:Objective: To facilitate manipulation of gene expression in different host cells, we used pEGFP-N1 as backbone to construct a versatile vector that can drive foreign gene expression in prokaryotic and eukaryotic cells. Methods: A cloning and expression vector, pEGFP-N1-lac, was constructed by inserting the prokaryotic lac promoter of pUC19 into the eukaryotic expression vector, pEGFP-N1, between the eukaryotic PCMV promoter and enhanced green fluorescent protein (EGFP) open reading frames. To assess the function of pEGFP-N1-lac, the nucleotide sequence encoding the hepatitis C virus (HCV) core protein was cloned into the multiple cloning sites. Western blotting analysis was used to detect the expression of the HCV core protein in Escherichia coli DH5α and HepG2 cells. Results: Restriction enzymedigestion and sequence analysis indicated that pEGFP-N1-lac was successfully constructed and the HCV core gene was cloned into this vector. The Western blotting results showed that pEGFP-N1-lac promoted expression of HCV core gene in prokaryotic E. coli DH5α and eukaryotic HepG2 cells. Conclusion: The pEGFP-N1-lac vector has been successfully constructed and functions in both prokaryotic and eukaryotic cells. The EGFP reporter can be used as an insert-inactivation marker for clone selection or as an expression tag. This vector can be used for cloning and expression of genes in both prokaryotic and eukaryotic cells, making gene cloning, expression and functional studies convenient as well as time- and labor-efficient.

      Abstract:Background: Laparoendoscopic single-site surgery (LESS) may serve as a potential alternative to conventional laparoscopy and is developing quickly, but still in its infancy. The study is to present our two-year experience in transumbilical LESS simple nephrectomy (LESS-SN) for non-functioning kidney, in an effort to evaluate its feasibility, clinical outcomes and potential advantages. Methods: From December 2008 to December 2010, a total of 11 patients with body mass index (BMI) ≤30 underwent transumbilical TriPort TM LESS-SN by a single experienced urologist at our institution. The indications for nephrectomy included nonfunctioning kidney associated with ureteropelvic junctionstricture (n=1), ureteral calculi (n=6), tuberculosis (n=3), and ureteral stricture (n=1). Patient demographics, perioperative and follow-up data were prospectively collected and analyzed. Results: Ten procedures were successfully completed with one patient converted to open surgery due to uncontrollable bleeding. The mean operative time was 189.2 (ranging 100320 min) with an estimated blood loss of 204.5 (ranging 50-1 000 ml). There were two complications of bleeding (1- intra-, 1- post-). The mean hospitalization after surgery was 7.9 d (ranging 4-17 d). With a regular follow-up of 1, 6, 12, and 24 months after surgery, all patients remained symptom-free with an intra-umbilical scar. Conclusion: Transumbilical LESS simple nephrectomy for nonfunctioning kidney can be accomplished with favorable surgical outcomes and a superiority of cosmesis. However, cases with chronic inflammation are not suitable for initial up-take and should only be attempted by the very experienced laparoscopist.

      Abstract:Objective To evaluate the effect of carotid artery stenting combined with nimodipine on cerebral hemodynamics by comparing and analyzing the change of anterior cerebral blood flow before and after treatment through transcranial Doppler (TCD). Methods Twenty patients with ischemic cerebrovascular disease were detected in blood flow velocity of bilateral middle cerebral arteries (MCA) and anterior cerebral arteries (ACA) and artery pulsatility index with TCD before and two weeks after treatment. Results After treatment, TCD examination showed dynamic changes in blood flow significantly. Ipsilateral MCA flow velocity significantly increased; ipsilateral ACA flow velocity did not change significantly; there was no significant increase in the contralateral MCA flow velocity; flow rate decreased significantly in contralateral ACA. Conclusion Carotid artery stenting combined with nimodipine treatment can cause anterior cerebral hemodynamic changes in ipsilateral hemisphere and significantly improve blood flow.

    • Prunella vulgaris L. extract improves cellular immunity in MDR-TB challenged rats

      Abstract:Objective: To study the effect of the extract of Prunella vulgaris L. on multiple drugs resistant bacillus tuberculosis (MDR-TB). Methods: Experimental animal model in rats was induced by MDR-TB. Normal group, model group and Prunella vulgaris L. group were set up. The contents of IFN-γ, IL-4, IL-10 and IL-12 were examined by ELISA. Their genome mRNAs were extracted, the target genes were amplified by PCR. RT-PCR was used to detect the mRNA levels of them. Results: The content of IFN-γ of the extract of Prunella vulgaris L. group was 1.98±0.67 pg/ml, IL-4 was 6.47±1.46 pg/ml, IL-10 was 12.13±3.43 pg/ml and IL-12 was 3.02±0.86 pg/ml. Compared with the model group, Prunella vulgaris L. group was notable difference in serum IFN-γ, IL-12 and IL-10 (P<0.05). The mRNA levels of IFN-γ, IL-12 increased and IL-10 decreased obviously, the differences were quite significant (P<0.05), but IL-4 had no obvious change. Conclusion: The extract of Prunella vulgaris L. can enhance the cellar immunological function in rats from up-regulation of the level of genetic transcription, accordingly provide the theory basis of healing of tuberculosis with it.

      Abstract:Peutz-Jeghers syndrome (PJS), also known as hereditary intestinal polyposis syndrome, is an autosomal dominant genetic disease characterized by the development of benign hamartomatous polyps in the gastrointestinal tract and hyperpigmented macules on the lips and oral mucosa. Here, we reported 5 cases of PJS and our experience about its diagnosis and treatment.
