Abstract:<正>Objective: To construct mammalian cell expression vectors for Mac-1 with CFP and YFP and apply FRET to study the dimerization and function of CD11 b( Mac-1 α subunit) and CD18(Mac-1 β subunit). Methods: The mammalian cell expression vector for CD11b fused with CFP at the carboxyl terminal was constructed to create recombinant plasmid of pCD11b-CFP. Then pCD11b-CFP was co-transfected with pYFP-CD18 into CHO cell, a fibroblast like cell line, as a target cell within which there are some signal pathways involved in inflammatory stimulation but without endogenous Mac-1. Then CHO cells stably expressing both CD11b-CFP and YFP-CD18 fusion proteins were selected by Western blot and laser scanning confocal microscope. Results: The cyan and yellow fluorescence in co-transfected positive CHO cells were observed under a fluorescence microscope. Abstract:<正>Objective: To study the effect of simulated ischemia-reperfusion (I/R) on If of sinoatrial node (SAN) cells and the intervention of KATP channel opener Pinacidil. Methods: The SAN cells of the neonatal rats were detached and purified 2 d before the experiment. Abstract:<正>Objective: To establish the rat model with myocardial hypoxia/reoxygenation (H/R) injury, and investigate the protective effect of EPO pretreatment on the myocardium. Methods: Sixty male adult Wistar rats were randomly divided into 3 groups: control group, H/R group, and EPO group, 20 in each group. The rats in EPO group accepted injection of 5 000 U/kg recombinant human erythropoietin (RHuEPO) through vein, and the other rats accepted the injection of the same volume of saline. Twenty-four hours after the injection, rats in the EPO and H/R groups were put into the hypoxia environment for 12 h and then returned to the normoxic environment for 2 h, and then the samples of blood and myocardium were collected. Abstract:<正>Objective: To investigate anti-tumor effect of bufalin on the orthotopic transplantation tumor model of human hepatocellular carcinoma in nude mice. Methods: BEL-7402 cells of human hepatocellular carcinoma were inoculated to form subcutaneous tumors in nude mice by subcutaneous injection. Then the subcutaneous tumors were implanted into the liver of nude mice, and the orthotopic transplantation tumor models of human hepatocellular carcinoma were established. Seventy-five models were randomized into 5 groups ( n = 15) . Bufalin was injected intraperitoneally into the 3 groups at dose of 1.5,1 and 0.5 mg/kg for day 15 - 24, respectively. NS group were injected equal volume saline as above and adriamycin were injected intraperitoneally into ADM group at dose of 8.0 mg/kg for day 15. Abstract:<正>Objective: To test whether the novel tumor-suppressor candidate gene-ndr2 is also differentially expressed between osteoarthritis synovium cells (OASC) and rheumatoid arthritis synovium fibroblasts (RASF), and whether ndr2 can suppress the growth of RASF in vitro. Methods: Dot blotting, cell culture and gene transfection, cell cycle analysis techniques were applied to investigate the effect of ndr2 on the cell phenotype and cell cycles. Results: ndr2 is expressed in OASC but absent in RASF. Transient transfection of ndr2 into RASF can suppress the growth of RASF from phenotype observation. Cell cycle analysis showed that apoptotic peaks can be detected in RASF cells transfected with ndr2 gene. Conclusion: Novel tumor suppressor candidate ndr2 is not only differentially expressed between OASC and RASF but also can induce the apoptosis of RASF in vitro.

Methods: The eukaryotic expression vector pcDNA3. 1/V5-His-TOPO-HNP1 was cotransfected with plasmid pDCH1P11 carrying dhfr gene into dhfr- negative CHO (CHO-dhfr- ) cells and recombinant protein was verified by ELISA; G418 selective medium was used to screen the stably expressing cell clones followed by serial passages in 5×10-8 mol/L and 5×10 mol/L methotrexate (MTX) for gene amplification. Finally 4 cell clones with high expression level were obtained and confirmed by ELISA, RT-PCR and IFA. The bacteriastatic activity of concentrated supernatants was tested in vitro as well. Abstract:<正>Objective: To evaluate cyclooxygenase-2 (COX-2) expression induced by insulin in cultured rat vascular muscle smooth cells (VSMCs). Methods: The rat VSMCs were isolated and cultured in 60 mm plates, pre-treated with insulin 10, 100, 500, 1 000 mU/L for 6 h, and 100 mU/L for 3, 6, 12, and 24 h, respectively. RT-PCR was performed to measure COX-2 mRNA induction, and Western-blot was used to measure protein expression. In regard to the COX-2 enzyme activity, after stimulation for a specified duration, the culture medium was collected; Prostaglandin E2 (PGE2) released by VSMCs was measured with an EIA kit. Results: Three hours after insulin (100 mU/L) stimulation, the induction of COX-2 mRNA was detectable and increased with the time and dosage of insulin. Insulin induced COX-2 protein expression occurred in a time- and dose-dependent manner. In rat VSMCs, COX-2 induced by insulin resulted in PGE2 production, which was increased with insulin action. Abstract:<正>Objective: To investigate the cytotoxicity and cytocompatibility of chitin fiber reinforced polycaprolactone composite in vitro in order to provide useful scientific basis for clinical application. Methods: Cell morphology observation, MTT and DNA assay were used to evaluate the influence of the composite on the morphology, growth and proliferation of cultured L-929 cells. Results: The composite did not impair the morphology of cultured cells in vitro. MTT and DNA assay demonstrated that the growth and proliferation of the cultured cells were not significantly inhibited by the composite. Conclusion : The composites have fine cytocompatibility and are safe for clinical use of reconstruction of chest wall defects.
Abstract:<正>Objective: In order to study the role and significance of transforming growth factor beta-1 (TGF-β1 )mRNA in transplanted renal fibrosis(TRF). Methods: Renal pathologic changes and expression of TGF-β1 mRNA were observed using in situ hybridization technique. The normal renal tissue as a. control group. Results: Expression of TGF-β1, mRNA in the renal fibrosis increased, compared with that in the control group. The expression rate were co-related to the stage of TRF. Conclusion: TGF-β1 is related to the pathogenesis and development of TRF.

Abstract:<正>Objective: To study the effect of CO2 pneumoperitoneum and the special 30 degree head-down tilt position on the venous hemodynamics in the lower extremity. Methods: Color doplex ultrasound was adopted to evaluate the diameter and blood flow velocity of the right femoral vein of 18 patients undergoing gynecologic laparoscopy under the same pressure of pneumoperitoneum of 12 mmHg. The diameter of femoral vein and the flow velocity were measured; the blood flow volume was calculated based on the equation of Q = vπr2 . Result: After establishment of pneumoperitoneum, the dilation of the femoral vein and the decrease in the velocity and volume can be observed (P < 0.05). And the 30 degree head-down position could increase the flow velocity and volume of the femoral vein and decrease the diameter of the vessel ( P < 0.05). Abstract:<正>Objective: To explore the serum levels of adiponectin in patients with essential hypertension and the relation between adiponectin and blood pressure. Methods: Forty-five cases with essential hypertension and 43 healthy control subjects have been taken fasting blood samples for measurements of plasma glucose, plasma lipids, insulin, C-peptide, thyroid hormones, TNF-α, leptin and adiponectin. Glucose tolerance was assessed by 75-g oral glucose tolerance test. Abstract:<正>Objective: To study the causes and value of the pseudo-occlusion of the anastoraotic pathway between celiac and superior mesenteric arteries. Methods: 15 patients including 12 patients with hepatocellular carcinoma and 3 patients with hepatic hemangioma underwent superior mesenteric arteriography (SMA) and celiac arteriography (CA) in interventional therapy. The angiographic features of the 15 cases were dynamically observed by 2 experienced radiologists in double-blind manner. Results: Similar interpretations were given by the 2 radiologists. In CA, pseudo-occlusion occurred at the bifurcation of the common hepatic artery and the celiac artery in 8 cases, at the bifurcation of the common hepatic artery and the gastroduodenal artery in 6 cases and at the bifurcation of the right hepatic artery and the celiac artery in 1 case. Abstract:<正>This study was undertaken to analyze the perioperative factors and complications that influence the final outcome after deep hypothermic circulatory arrest and superficial hypothermia during operations on aortic aneurysms from Jan 2000 to Jun 2004. The results show that deep hypthermic circulatory arrest group provided a high risk of pulmonary insufficiency-hypoxemia and temporary neurological dysfunction in complication but not increased the risk of hospital mortality.
