Abstract:<正> A set of allele-specific oligonucleotide (ASO) probes used for detecting all 18 β-tha-lassemia mutations found in Chinese was immobilized on two strips of Biodyne C membrane;one containing 7 pairs of oligonucleotide probes specific for the most commonly found mutant al-leles,and the other containing the remaining 11 pairs of ASOs specific for the less commonlyfound.The membranes were hybridized with β-globin sequences amplified by polymerase chainreaction (PCR) with biotinylated primers,and then treated with Streptavidin-POD conjugateand substrates for color development.The method has been applied successfully to the detectionof all 18 Chinese β-thalassemia mutations and prenatal diagnosis of two high-risk pregnancies ofβ-thalassemia.Patients with homozygous,heterozygous and compound heterozygous alleles ofthese mutations and normal individuals could be easily distinguished by the present method.Us-ing the immobilized-probe format (reverse dot blot),it was able to screen simultaneously multi-ple β-thalassemia mutations of a DNA sample by performing hybridization only once.This assayis simple,rapid and independent of radio-isotopes and can be appplied for all 18 β-thalassemiamutations so far found in Chinese population.It is considered that this method may be usefulfor gene frequency investigation of large numbers of β-thalassemia DNA samples and used as aroutine method in the clinic laboratory.
Abstract:<正> The changes of 8 oncogenes and 2 oncosuppressor genes in the specimens of cancer-ous and juxtacancerous tissues of 42 cases of gastric cancer were studied with southern blot hy-bridization and PCR-RFLP method.The probes used were c-Ha-ras,K-ras,N-ras,N-myc,c-myc,hst,EGFR,c-erbB-2,p53 and Rb.Amplification,rearrangement and deletion of c-Ha-ras were detected in 8/33 (24.2%) cases of gastric cancer,amplification or rearrangement ofhst and c-erbB-2 in 11/42 (26.2%) and 12/41 (29.2%) cases respectively,amplification ofEGFR in 9/42 (21.4%) cases,and deletion or rearrangement of p53 and Rb in 9/30 (30.0%)and 2/15 (13.3%) cases respectively.Amplification or rearrangement of N-ras (0/33),K-ras(1/26),N-myc (1/26),and c-myc (1/35)was rarely encountered.These findings suggest thatc-Ha-ras,hst,c-erbB-2,EGFR and p53 may be important genes involved in the pathogenesisof gastric cancer.
Abstract:<正> In this study,the DNA probe pPF14 was nonradioactively labelled by sulfo-modifica-tion,and used in a dot blot hybridization assay for detection of P.falciparum.The resultsshowed that the sulfoprobe successfully detected 25pg purified DNA and 0.001% parasitemia ofcultured P.falciparum,and did not react with human leukocyte DNA.In the study of 179clinical blood samples of malaria patients from Yunnan province,the DNA probe results corre-lated well with blood smear ones.Of 107 P.falciparum samples determined by microscope ex-amination,99 were positive by sulfoprobe (92.5%);Of 72 P.vivax samples,1 was crosslyreacted;no cross reactions were found with any of 48 normal blood samples.It is indicated thatsulfoprobe may be useful in specific diagnosis and epidemiological investigation of P.falciparuminfection.
Abstract:<正> It was investigated that the localization and distribution of calcitonin gene-relatedpeptide (CGRP) in the rabbit lung by ABC-GDN immunohistochemistry.The results demon-strated that CGRP immunoreactivity was located in neuroendocrine cells (NECs) and neu-roepthelial bodies (NEBs),including all airways from bronchi,bronchioles,and alveolar ducts.The nerve fibers in walls of the intrapulmonary bronchi and blood vessels also had CGRP-immunoreactivity.Adjacent sections immunostained with antibodies to CGRP and 5-HT provedthe coexistence of CGRP and 5-HT in NECs and NEBs.This finding suggested the possibilityof functional interaction between the peptide and amine.
Abstract:<正> A technique of direct sequence analysis of β-globin gene with the products of amplifi-cation by polymerase chain reaction (PCR) was reported and a case of β-thalassemia with therare mutation in Chinese,‘codon 14/15 (+G)’ was detected by this method.After the se-quence of the mutation site was determined,an analysis of the restriction map of the gene anddot blot hybridization with radioactive allele specific oligonucleotide probe was designed to con-firm the result of DNA sequencing.
Abstract:<正> Arterial blood gases,intraerythrocytic pH (pHi),2,3-diphosphoglycerate,standardP 50 (P 50std) and in vivo P 50 (P 50iv) were determined in 54 patients with cor pulmonale and23 normal subjects.It was found that no significant change of pHi was observed but the differ-ence of pHi and extraerythrocytic pH was decreased.P 50std was significantly decreased whileP 50iv remained essentially unchanged in those cases showing type I respiratory failure.Thesefindings suggest that P 50std cannot accurately reflect the changes of P 50iv in patients of corpulmonale;P 50iv in cases of cor pulmonale usually keeps relatively stable and coincides withthe ‘optimal P 50’,which indicates the adaptation of the organism to chronic hypoxia;and rel-ative intraerythrocytic alkalosis plays an important role in the relief of hypercapnia in cases ofcor pulmonale.
Abstract:<正> 190-kilodalton glycoprotein (P190) of Plasmodium falciparum,precursor of themajor surface protein of merozoites,is considered a promising candidate for blood stage malarialvaccine.Six primers were designed according to the sequence of MAD20 strain,with a GCclamp and BamH Ⅰ site at the 5’-end of each one,and a GC clamp and Xba Ⅰ site at the 3’-endof each one.The primers were synthesized by phosphoramidite approach (User’s Manual ofABI Company) and purified using HPLC.Three fragments in the second,third and fourth con-served regions of P190 gene of Plasrnodium falciparum FCC1/HN strain isolated from theblood of patients in Hainan Province of China were amplified by the polymerase chain reaction(PCR).The amplified fragments were suhcloned into pUC18 vectors and sequenced using thedideoxy chain termination method.All three regions of P190 gene of FCC1/HN strain also werehighly conservative as compared with P190 gene of MAD20 (Papua New Guinea isolate),K1(Thailand isolate),Wellcome (West Africa isolate) and CAMP (Malaysia) strains ofPlasmodium falciparum.The C at position 81 in the second conserved block of P190 gene ofFCC1/HN isolate was substituted by T,which did not change the amino acid determined by thecodon corresponding to the substitution.The genes sequenced were cloned into rpGEX-2T,aglutathione S-transferase gene fusion system for expression.
Abstract:<正> It has been found that vasoactive intestinal peptide (VIP) stimulates the growth ofseveral kinds of tumor cells.There is no report so far about the effect of VIP on the growth ofhepatoma cells.Using the tetrazolium colorimetric assay (MTT assay) and cell countingmethod,it was investigated that the effect of VIP on the growth of a cultured rat hepatomaFSK-7902 cells.The results showed that VIP stimulated the proliferation of the rat hepatomacells obviously.The addition of 1μmol/L VIP caused a significant increase in the number of thecultured rat hepatoma cells on 3rd day and maximal increase occured on 4th day and 5th day ( P<0.01).The growth promoting effect was greater as the concentration of VIP increased.Thelowest effective concentration of VIP was 0.5μmol/L.Exposure to VIP for 12 h followed by re-moval of the peptide resulted in sustained growth for several days.
Abstract:<正> To evaluate the diagnostic value of the abnormal systolic blood pressure (SBP) re-covery ratio of exercise test and its mechanisms,142 patients with suspectable angina pectorisunderwent coronary angiography and treadmill exercise test.The SBP recovery ratios at the1st,3rd,and 6th min of the recovery period in patients with coronary artery disease (CAD)were significant higher than those in the control subjects.Of the hymodynamic variables,car-diac index (CI),stroke volume index (SI) and ejection fraction (EF) were lower,left ventricu-lar end-diastolic pressure (LVEDP) and systemic vascular resistance (SVR) were higher in pa-tients with abnormal SBP response than in patients with normal SBP response.The sensitivityof abnormal SBP recovery ratio for identifying patients with CAD was 68% and the specificitywas 90%.The false positivity rate of abnormal SBP recovery ratios in 21 patients with hyper-tension without CAD was 38%.These results indicate that an abnormal SBP recovery ratio is auseful index for identifying patients with CAD.When the ST segment response to exercise isnormal or borderline,it can increase the sensitivity and the accuracy of the exercise test.Theabnormal SBP response appears to be due to exercise-induced impairment of left ventricularfunction and increase in systemic vascular resistance.
Abstract:<正> A generator to induce simulated underpressure of blast waves has been designed andapplied for the study of underpressure injuries in animals.The generator consists of a vacuumchamber,a vacuum pump,an experimental chamber,a membrane-rupturing device,and apressure-measuring system.When the pressure in the vacuum chamber is reduced with the suc-tion of the vacuum pump to a given level,the membrane separating the vacuum chamber andthe experimental chamber,in which the experimental animal is fixed,is ruptured instantly by asharp pointer.Underpressure occurs in the experimental chamber as soon as its pressure is re-duced rapidly.The physical parameters of the simulated underpressure of blast waves rangefrom-13 to-90 kPa for the peak level,1 to 90 ms for the decreasing time and 14 to 2000 msfor the duration.Animal experimentation with the generator verified that underpressure can in-flict injuries on living bodies as well.
Abstract:<正> A simple and rapid method was established to study vascular permeability by in vitroperfused endothelial cell monolayers cultured on micropore filter membrane.It can be used todetermine filtration coefficient (Kf) to small molecules and osmotic reflection coefficient (σ) toproteins of the endothelial monolayer.Hanks’ balanced salt solution (HBSS) or 5g/L albuminin HBSS was used to perfuse the confluent endothelial monolayer at the hydrostatic pressure of2.45kPa (25cm H2O).Control Kf values were 10.1±0.75 and 3.6±0.75μl·min-1·cm-2·kPa(-1)(±,n=3) respectively for the perfusion of HBSS and albumin HBSS,suggesting that al-bumin may decrease endothelial monolayer permeability to water and small molecules.After ex-posure of endothelial monolayer to 10-8mol/L platelet-activating factor (PAF) for 30min,Kfvalues increased to 193.1% and 133.3% respectively.Protein clearance rate (μl.min-1·cm-2)and osmotic reflection coefficient of the control endothelial monolayer were 8.0±3.22 and 0.37±0.09 respectively and those of the PAF treated endothelial monolayer 12.2±2.95μl·min-1·cm-2 and 0.18±0.06,revealing increased permeability to albumin.Computer-assisted imageprocessing demonstrated that PAF treatment decreased cell area while increased cell form factorand intercellular space,suggesting that endothelial cells retracted and rounded,which may bean important mechanism of PAF-induced increase of vascular permeability.
Abstract:<正> The correlation of secondary brain injury with thromboxane A2(TXA2) and prostacy-clin (PGI2) levels following head injury was studied in rats and patients.Thromboxane B2(TXB2) and 6-keto-PGF1α in rat brain homogenate and TXB2 in cerebral spinal fluid (CSF) ofpatients with severe head injury were determined by RIA.The effects of ligustrazini hydrochlo-rioi were also tested.The results showed that the concentration of TXB2 and 6-keto-PGF1α inrat brain was progressively increased within 3h after injury (P<0.01),and the ratio betweenTXB2 and 6-keto-PGF1α(T/K value) increased during the early stage after injury,along withthe tissue damage aggravation.After the use of ligustrazini hydrochlorioi,TXB2 content inbrain decreased sharply accompanied with a drop in T/K value.Moreover,TXB2 content inCSF increased within 7 d after injury (P<0.01),and it reached its peak value on the 3rd day.This could indicate that the metabolic imbalance between TXA2 and PGI2 might be one of theimportant factors in the development of secondary brain injury,and ligustrazini hydrochlorioiproved to have a protective effect on the brain tissue by normalizing the metabolism of TXA2and PGI2.In addition,the metabolic disorders of TXA2 in the brain tissue of head injured pa-tients have much to do with the outcome of the patients.
Abstract:<正> Forty children with thalassemia major were treated with gelfoam particles for partialsplenic embolization (PSE).The embolization of the spleen ranged from 50 to 85 per cent.Af-ter PSE,the spleen shrank greatly,and the symptoms of anemia improved.The immunologicalfunction of the body was the same as before PSE.All patients showed a marked reduction intransfusion requirements.Therefore,it is believed that PSE is an effective therapy for tha-lassemia major.
Abstract:<正> In order to clarify whether gentamicin (GM) could damage the renal glomeruli,theultrastructural changes of the renal cortex were observed in rabbits after they received a singleinjection of 100 mg/kg GM or 100 mg·kg-10·d-1 GM for 3 consecutive days.The animals ofthe control group received equal amount of normal saline.It was found that some myelin figuresappeared in the epithelial cells of the proximal tubules as early as 1 h after a single GM injec-tion;severe swelling,degeneration,and obvious changes of the organelles were seen in the en-dothelial cells of the glomerular capillaries,the podocytes and the epithelial cells of the proxi-mal tubules 24 h after an injection of GM;and fusion of the processes of podocytes,thickeningof the glomerular membrane and necrosis of the epithelial cells of a part of the’proximal tubuleswere encountered 24h after the 3rd GM injection.These findings demonstrate that GM can in-jure not only the proximal tubules but also the glomeruli in rabbits.
Abstract:<正> Twenty samples of endothelia removed from normal and post penetrating keratoplas-ty (0.5,1,2,3 months after penetrating keratoplasty) were observed by scanning electron mi-croscopy.The photographs of the endothelia in graft-host junction were analyzed by computer-assisted image analysis system,and the morphometric indexes examined were area of the cells,perimeters,density,figure coefficient,long axis,coefficient of variation of the area,and oth-ers.Results showed that the morphology and the density of the endothelial cells changed obvi-ously after operation and improved slowly but progressively with time although at 3 monthspostoperatively some differences still existed.By using the new techniques,the experiment con-firmed and enriched the theories on the corneal endothelial wound-healing,revealing some ofthe new characters of the endothelial wound-healing following penetrating keratoplasty.
Abstract:<正> In the present investigation,measurement of leukocyte chemiluminescence (CL) wasundertaken using micro-whole blood assay in 76 patients with coronary heart diseases (CHDs).The findings showed that there was a significant increase in leukocyte CL in patients with un-stable angina pectoris (UAP) and acute myocardial infarction (AM1) (P<0.01) and the in-crease was greater in AMI than that in UAP (P<0,01).It was found that leukocyte CL inAMI was correlated with the extent of the infarct and increased with the size of the infarct.Leukocyte CL in AMI patients who had received fibrinolytic treatment was markedly greaterthan that before treatment (P<0.05),suggesting that leukocytes may play an important rolein the pathogenesis of CHDs and may be involved in myocardial ischemia-reperfusion induced in-jury.The findings of the present study demonstrate that measurement of leukocyte CL may beof benefits for prognosis and treatment of CHDs.
Abstract:<正> A model of incomplete cerebral ischemia caused by bilateral ligation of the commoncarotid arteries in rats was used to examine the effect of naomaikang on the relief of cellulardamage following cerbral ischemia.Cellular damage and the formation of peroxidation productswere reduced in rats pretreated with naomaikang.It has been suggested that the free radical re-action involved with tissue damage,particularly with the injury of neurons in cerebral ischemia.The results showed that the degree of lipid peroxidation can be lowered by the pretreatmentwith naomaikang in the model of incomplete cerebral ischemia,and suggested that this drug isbeneficial to the recovery of cerebral ischemia in humans.
Abstract:<正> Plasma motilin concentrations were measured in 135 women during the second andthird trimesters of pregnancy and 3~5 d after delivery and the results were compared withthose of 20 healthy nonpregnant women.The mean plasma motilin concentration (323.96±125.10 ng/L) in women during the second trimester of pregnancy was lower than in healthynonpregnant women (366.12±96.23 ng/L) (P<0.05),but that was significantly higher thanin women during the third trimester of pregnancy (121.04±27.00 ng/L) (P<0.01);theplasma motilin concentration in women during 3~5d after delivery (443.05±140.79 ng/L)was significantly higher than that in pregnant women (P<0.01).The results showed thatpregnancy appears to have a profound inhibitory effect on plasma motilin,and this may partlybe responsible for the gastrointestinal hypomotility associated with pregnancy.
Abstract:<正> Pulmonary injury after acute H2S inhalation was studied with bronchoalveolar lavage(BAL),ultracentrifuge,and optical and electron microscopy.The changes of the activities oflactate dehydrogenase,alkaline and acid phosphatase,and angiotension converting enzymes inBAL fluid were used as the indicator of cellular damages,those of leucocytic count and the indi-cator of inflammatory response,and those of the concentration of protein and Evans blue as theindicator of vascular permeability.In addition the effects of H2S on lipid peroxidation,naturalantioxidative system and energy substances and the changes of phospholipid concentration inBAL fluid were also observed.The results were as follows:1.Inhalation of H2S exerted a severe cytotoxic effect on the lung tissues resulting in dam-ages on various types of cells and a severe edematogenic effect on lung parenchyma.2.The development of pulmonary edema in H2S intoxication resulted from a combination ofdifferent pathogenic factors.3.The biochemical changes and their recovery occurred earlier than the pathologicalchanges and their recovery.The efficacy of 6 categories of drugs including 25 medicaments against H2S intoxication wasevaluated in mice,and 10 drugs were found to be prophylactically effective.The effects of vari-ous methemoglobin-forming agents and some other drugs were also studied in their treatment ofH2S intoxication in rabbits and dogs.It was concluded that methemoglobin-forming agentscould be used as specific antidotes but could not provent or alleviate the lung damages due toH2S inhalation unless they were administered in association with dexamethasone,vitamin E oranisodamine.A scheme of the medical treatment for H2S intoxication was presented.
