The influence of actinomycin D and puromycin on the mediation of cellularimmunity by mouse S
180
ascitic tumor cell-specific iRNA was investigated with
3
H-Leu-LAItest as an index. The results showed that mouse splenic lymphocytes sensitized with iRNAmaintained their LAI activity after the translation or transcription was blocked bytreatment with puromycin or actinomycin D respectively;and that when first treated withactinomycin D or puromycin
then incubated with iRNA and relevant antigen
thelymphocytes did not show any LAI activity. These results suggest that transcription andtranslation processes in lymphocytes are involved in the mediation of LAI reaction byiRNA
and during the treatment of cancer
the effectivity of iRNA would be reduced by theprevious administration of tumoricidal agents which are suppressive to the RNA and/orprotein synthesis in patient’s lymphocytes.
