Abstract:<正>Objective: To observe the expression of proto-oncogenes in the process of airway remodeling in asthma. Methods: Guinea pig was used as an asthma model challenged by ovoglobulin. Dot-blot, Northern-blot molecular hybridization and immunohistochemistry techniques were used to detect the expression of c-fos, c-myc, c-jun and c-sis. Results: Expression of c-fos and c-myc mRNA could not be detected or detected at very low level in the control group. There were greatly increased expression of c-fos and c-myc mRNA after guinea pigs were challenged by ovoglobulin. Thirty minutes after the challenge, the expression of c-fos and c-myc mRNA reached to the peak and returned to normal level 4 h after the challenge. Immunohistochemistry studies showed that Fos, Myc, Jun and Sis expressed at low level in control group and increased after ovoglobulin stimulation. Abstract:<正>Objective: To investigate the influence of anti-keratin autoantibodies (AK auto Abs) on telom-erase activity of squamous cell carcinoma cultured in vitro and the mechanisms of the inhibitory effects of AK auto Abs on squamous cell carcinoma. Methods: Influence of AK auto Abs on the proliferation of Tca cells was observed by MTT colorimetry. Telomerase activity of cultured Tca cells and human keratinocytes was determined by telomeric repeat amplication protocol-ELISA (TRAP-ELISA) and polyacrylamide gel elec-trophoresis (PAGE). After being treated with AK auto Abs for 36 h at a concentration of 4, 8, 16 mg/L respectively, the changes of telomerase activity of Tea cells were also detected by TRAP-ELISA and PAGE. Abstract:<正>Objective: To explore the method to repair injured peripheral nerve using conduits made of human hair keratin (HHK). Methods: The tibial nerves of rabbits were transected leaving a gap 10 mm in length between the 2 severed ends, which were either routinely sutured or bridged using HHK nerve conduits. Electro-physiological , anatomical and histological examinations were performed at different time postoperatively. Results: Electrophysiological study showed more obvious improvement in the neural function recovery in rabbits with HHK conduits bridging as compared with that in rabbits with routine suture. In the former group, HHK conduits were gradually degraded and absorbed with large amount of myelinated nerve fibers and Schwann cells regenerated around HHK conduits. In the latter group, however, the nerve tissues around the suture were degenerated and replaced by connective tissues. Conclusion: HHK may induce the regeneration of the nerve fibers and provides an ideal approach to repair nerve damages.
Abstract:<正>Objective: To investigate the association between the diversity of urease gene and urease activity of clinical isolates of Helicobacter pylori (H. pylori). Methods: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) of urease gene and rapid urease activity test were used to study the urease activity of different clinical isolates of H. pylori. Results: H. pylori clinical isolates were divided into 4 types according to their PCR-RFLP results of urease gene and urease activity. Abstract:<正>Objective: To explore the characteristics of crisis state at erythrocytic stage of Plasmodium berghei chloroquine-resistant (RC) strain. Methods: Agarose electrophoresis, optical and transmission electron microscopes were used. Patterns of genomic DNA structures and ultra structures of the erythrocytic parasites were observed in ICA mice (infected with the RC strain) during rising and declining of parasitemia. Results : During the declining parasitemia, the erythrocytic stage parasites of the RC strain showed round or o-val appearance with intact plasma membrane and shrank nuclei with no metabolic window, mitochondria or other membranaceous structures. Their DNA electrophoretogram revealed a ladder pattern which evidently differed from the parasites of the RC strain in the rising parasitemia and the chloroquine-sensitive (N) strain. Conclusion; The crisis state of the erythrocytic stage parasites of the P. berghei chloroquine-resistant (RC) strain is characterized by apoptosis.
Abstract:<正>Objective: To compare the cleavage activity of ribozymes directed against 2 sites of PDGF receptorP subunit cDNA gene in vitro. Methods: The 608 bp fragment of PDGF receptor β subunit cDNA was clonedinto T-vector under the control of T7 promoter, named pPDGFR-β. Two ribozymes were designed to cleavethe CUU sequence at codon 45 and codon 252 of PDGF receptor β subunit mRNA respectively. These 2 ham-merhead ribozyme genes were cloned into vector PI. 5 between 5’ -cis ribozyme and 3’ -cis ribozyme to gener-ate the plasmids of pRZ1 and pRZ2. The pPDGFR-β, pRZl and pRZ2 were linearized and then transcribedwith T7 promoter in vitro. Results: The RZ1 showed high cleavage activity in vitro, but the RZ2 showed nocleavage activity under the same condition. Conclusion: The cleavage site selection is an important factor in-fluencing the cleavage activities of ribozymes.

Abstract:<正>Objective: To investigate the association of changes in G and D cells in the antral mucosa with the production of gastrin and somatostatin during gastric ulcer and the healing process. Methods: Experimental gastric ulcer was induced with acetic acid in 42 Wistar rats and another 7 normal rats served as control. Changes in the production of gastrin and somatostatin in the plasma, gastric fluid and the antral tissues of the rats were measured by radio immunoassay, and the number and distribution of G and D cells were respectively determined by immunochemistry and Quantimet500 image analysis system. Results: In rats with gastric ulcer, the gastrin levels in the plasma, gastric fluid and the antral tissues increased while somatostatin levels were reduced, which were corrected in the healing process. Methods: Immunocytochemistry, Western blot and electrophoretic mobility shift assay (EMSA) were used. Results: The expression of NF-kB in cultured mouse bone marrow stromal cells (BM-SCs) on the level of protein was elevated after exposure to 60Co in the dosage of 8. 0 Gy with the use of im-munocytochemistry and Western blot. The activity of nuclear factor-kappa B in cultured BMSCs was significantly increased after exposure to gamma irradiation by using EMSA. The activity peak was at the 4th h after irradiation. Conclusion: Our results suggest that the activation of nuclear factor-kappa B in the BMSCs after irradiation may be involved in the protection of BMSCs against apoptosis and in the recovery of hematopoiesis after radiation.
Abstract:<正>Objective: To evaluate the therapeutic effects of radiosurgery on brain tumor using 99Tcm-MIBI brain single-photon emission computed tomography (SPECT). Methods: Fifteen normal volunteers and 49 patients with brain tumor underwent 99Tcm-MIBI brain SPECT, and the tumor to non-tumor ratio (T/N) was calculated and compared before and after radiosurgery. The patients were regrouped according to different schedules for postoperative reexamination, and diagnostic sensitivity and specificity of 99Tcm-MIBI SPECT evaluated against that of conventional CT and magnetic resonance imaging. Results: After radiosurgery, the lesions were reduced or even disappeared in 22 cases, and tumor remnants or recurrence were found in 27 cases. The sensitivity, specificity and accuracy of "Tc"’-MIBI brain SPECT were 85. 2%, 68. 2% and 77. 6% , respectively. Abstract:<正>Objective: To analyze the effect of lymphocytes on proliferation of keratinocytes in patients with psoriasis. Methods: Lymphocytes in lesion and peripheral blood were isolated and amplified, then cultured together with normal keratinocytes. By MTT method, the living cells were quantified in the mixed culture. Results: Compared with normal controls, lymphocytes from lesion and peripheral blood of psoriasis both promote the proliferation of keratinocytes (P<0. 01 and P<0. 05 respectively). The concentrations of IL-2 and IFN-7 in the mixture of lesion lymphocytes and keratinocytes were significantly higher than that of controls. Tripterygium glycosides inhibited this promotion. Conclusion: Lymphocytes in patients with psoriasis (mainly Th1 cell) play an important role in proliferation of keratinocytes. This psoriasis cell model is useful for studies on signal transduction in psoriasis.
Abstract:<正>Objective: To assess the clinical efficiency of the transfusion of leucocyte filtrated RBC concentrates to prevent febrile nonhemolytic transfusion reactions (FNHTRs). Methods: One hundred patients with liver cirrhosis, gastric ulcer or cancer were subjected to receive RBC concentrates after leucocyte filtration. Another 50 patients with similar diseases were selected to receive non-filtrated RBC concentrates. The incidence of FNHTRs in all patients was investigated. Results: There was no FNHTR in 100 transfusions with leucocyte filtrated RBC concentrates, while FNHTRs occurred in 8 of 50 patients with non-filtrated RBC concentrates, with the incidence of 16%. Conclusion: FNHTRs to RBC transfusion can be prevented with leucocyte filtration.

Abstract:<正>Objective: To explore the affecting factors of liver cancer recurrence after hepatectomy. Methods: The BP artificial neural network - Cox regression was introduced to analyze the factors of recurrence in 1 457 patients. Results: The affecting factors statistically significant to liver cancer prognosis was selected. There were 18 factors to be selected by uni-factor analysis, and 9 factors to be selected by multi-factor analysis. Conclusion: The 9 factors selected can be used as important indexes to evaluate the recurrence of liver cancer after hepatectomy. The artificial neural network is a better method to analyze the clinical data, which provides scientific and objective data for evaluating prognosis of liver cancer.
Abstract:<正>Objective: To analyze the relationship of Stat3 expression with clinical stages, tissue types, p53 and proliferation cell nuclear antigen (PCNA) in human lung carcinoma, and to evaluate the role of Stat3 in the pathogenesis of lung carcinoma. Methods: Immunohistochemical method were used to detected Stat3, p53 and PCNA in different tissues of patients (n = 42) with lung carcinoma who accepted neither radiotherapy nor chemotherapy. The expression level of Stat3 in non-small cell lung carcinoma (NSCLC) was higher than that in small cell lung carcinoma (SCLC). Abstract:<正>Objective: To explore the role of vascular endothelial growth factor (VEGF) in the angiogenesis and development of human gastric carcinoma. Methods: The expressions of VEGF and its receptor KDR (ki-nase-domain insert containing receptor) in human gastric cancer tissue and SGC-7901 cells were detected with immunohistochemical staining. Microvessel density (MVD) was obtained after immunostaining for Factor-VIII. VEGF in SGC-7901 cell line was detected with Western blot.
